Septic arthritis: immunopathogenesis, experimental models and therapy J. Venom. Anim. Toxins incl. Trop. Dis.

Septic arthritis is an inflammatory disease of the joints that is started by an infection whose most common agent is Staphylococcus aureus. In this review we discuss some of the most arthritogenic bacterial factors and the contribution of innate and specific immune mechanisms to joint destruction. Special emphasis is given to the induction of experimental arthritis by S. aureus in mice. The improvement of therapy by association of antibiotics with down-modulation of immunity is also included.

Detection of Toxoplasma gondii (T. gondii) DNA in blood can help to diagnose the disease in its acute phase; however, it must be considered that hemoglobin, present in blood, can inhibit polymerase activity, making impracticable the detection of DNA in samples. Mice were experimentally infected via oral route with ME49 and BTU2 strains cysts and RH strain tachyzoites; polymerase chain reaction was used to detect T. gondii DNA in mice sera 18, 24, 48, 96, and 192 hours post infection (PI). AbstractBackground Jorge Lobo’s disease, also known as lacaziosis, is a cutaneous-subcutaneous mycosis with chronic evolution. It is caused by the fungus Lacazia loboi. Herein we report a study that relates the genotoxicity caused by L. loboi in isogenic mice with nutritional status, through a normal or restricted diet.Methods DNA damage was assessed in the peripheral blood by the comet assay (tail intensity).Results The results for leukocytes showed increases in the mean tail intensity in mice under dietary restriction, in infected mice under dietary restriction and in infected mice ingesting a normal diet.Conclusion These results indicate that dietary restriction and L. loboi infection may increase DNA damage levels in mice, as detected by the comet assay.

Although cryopreservation of mouse spermatozoa is simpler, less time-consuming, and less costly than of embryos for preservation of the genetic materials, maintenance of cryopreserved spermatozoa still has a high running cost because of the need for a constant supply of liquid nitrogen. Stimulatory factors for interleukin-12 production from murine splenic macrophages co-cultured with Babesia microti and Babesia rodhaini infected erythrocytes OAK

Stimulatory factors for interleukin 12 (IL-12) production associated with Babesia microti and Babesia rodhaini infected erythrocytes were examined using an in vitro assay system established, since a remarkable increase of serum IL-12 concentration and differentiation of helper T cell (Th cell) into helper T cell type 1 (Th1 cell) was observed in early phase of infection with Babesia spp. in mice. Beneficial Effect of Hyperbaric Oxygen Therapy on the Follicular Survival in the Mouse Ovary after Transplantation OAK

A large proportion of follicles are lost during the initial ischemia that occurs after transplantation of ovarian tissues. Thus, the effect of hyperbaric oxygen therapy (HBO) on the follicular loss of ovarian tissues after transplantation was examined in mice. Ovarian slices from ICR mice were transplanted under the kidney capsule in ovariectomized ICR. Hyperbaric oxygen with 100% oxygen was initiated for 30 min at 2.5 atmospheres absolute immediately after transplantation, and this treatment was repeated at 48-h intervals for 2 weeks. Aggravation of pathogenesis mediated by aflatoxin B1 in mice infected with Trypanosoma brucei rhodesiense OAK

Aflatoxins are known to alter the pathogenesis of many infectious diseases, but such effects have not been evaluated in trypanosome infections. The aim of the present work was to assess the effects of aflatoxin B1 on the pathogenesis of Trypanosoma brucei rhodesiense infection using a murine model. Mice fed on 0.50 mg/kg aflatoxin b. wt. were infected with T. b. rhodesiense and compared to trypanosome infected and uninfected aflatoxin-fed controls. The clinical and pathological changes were determined and the quantitative data statistically analysed using standard methods. The mRNA expression patterns of EGF, HB-EGF, Amphiregulin, EGF receptor, IGF-1, CSF-1, IL-1 alpha, IL-1 beta, IL-1 receptor type 1, IL-1 receptor antagonist, LIF, COX-1, COX-2, Mucin-1, calcitonin, and rat USAG-1 mouse homologue, all of which are involved in the process of conceptus implantation to the endometrium, were examined during the estrous cycle by means of real-time quantitative PCR. Protective immunity of blood plasma fraction containing Babesia rodhaini exoantigens in mice OAK

Babesia spp. are major hemoprotozoan parasites in animals. Exoantigens, which are soluble antigens released by the parasites during their asexual development, have been successfully applied for the development of a vaccine against babesiosis. In this study, we examined the ability of exoantigens to induce protective immunity in mice against a rodent Babesia parasite, Babesia rodhaini. The BALB/c mice immunized with the blood plasma fraction of B. rodhaini-infected mice (B. rodhaini exoantigens) showed a significant decrease in the lethal rate when challenged with B. rodhaini. Exposure of C57BL/6J Male Mice to an Electric Field Improves Copulation Rates with Superovulated Females OAK

It is well-known that there are considerable strain differences in the relative copulation rates between male and superovulated female mice. In particular, the C57BL/6J strain of mice has a lower rate of successful copulation. We examined the effect of exposure to an electric field on sexual behavior in C57BL/6J male mice. When C57BL/6J males were exposed to a 50 Hz, 45 kV/m electric field for 30 min per day for 11 days and placed in a cage with a superovulated female of the same strain, the successful copulation rates of males was significantly improved compared with unexposed males (P<0.05). These results suggest that the exposure of C57BL/6J male mice to an electric field improves their sub-fertility activity in mating with superovulated females.

Preliminary studies on the effects of orally-administered Transforming Growth Factor-beta on protozoan diseases in mice OAK

Transforming growth factor beta-1 (TGF-β1) is a pleiotropic cytokine with both pro- and antiinflammatory properties, depending on its environment and concentration. The present study evaluated the effects of orally-delivered TGF-β1 on mice parenterally-infected with various protozoan parasites. We report that while orally-administered TGF-β1 seems to confer partial protection against murine chronic babesiosis and acute trypanosomosis, no beneficial clinical effects were observed against acute babesiosis, malaria or toxoplasmosis. Through immuno-suppression, aflatoxins could affect drug and vaccine efficacy. Such effects have not been evaluated in treatment of many diseases including trypanosomiasis. We assessed the effect of aflatoxin B1 on the efficacy of suramin, the drug used for treatment of early stage sleeping sickness, in a murine model. Mice were fed daily on a diet containing 0.50 mg aflatoxin/kg body weight or a placebo.
