DNA damage in children and adolescents with cardiovascular disease risk factors Anais da ABC (AABC)
Kliemann,Mariele; Prá,Daniel; Müller,Luiza L.; Hermes,Liziane; Horta,Jorge A.; Reckziegel,Miriam B.; Burgos,Miria S.; Maluf,Sharbel W.; Franke,Silvia I.R.; Silva,Juliana da.
The risk of developing cardiovascular disease (CVD) is related to lifestyle (e.g. diet, physical activity and smoking) as well as to genetic factors. This study aimed at evaluating the association between CVD risk factors and DNA damage levels in children and adolescents. Anthropometry, diet and serum CVD risk factors were evaluated by standard procedures. DNA damage levels were accessed by the comet assay (Single cell gel electrophoresis; SCGE) and cytokinesis-blocked micronucleus (CBMN) assays in leukocytes. A total of 34 children and adolescents selected from a population sample were divided into three groups according to their level of CVD risk. Moderate and high CVD risk subjects showed significantly higher body fat and serum CVD risk markers than low...

Hibiscus acetosella extract protects against alkylating agent-induced DNA damage in mice Anais da ABC (AABC)
VILELA,THAIS C.; LEFFA,DANIELA D.; DAMIANI,ADRIANI P.; DAMAZIO,DAIANE DAL COL; MANENTI,ALINE V.; CARVALHO,TIAGO JOSÉ G.; RAMLOV,FERNANDA; AMARAL,PATRICIA A.; ANDRADE,VANESSA M. DE.
Abstract Hibiscus acetosella was shown to exert beneficial effects in humans and animal models however, the effects of this plant on DNA are unknown. The aim of this study was to determine the antigenotoxic and antimutagenic effects of H. acetosella extracts on alkylating agent methyl methanesulfonate (MMS) in vivo in mice. Initially, we performed analysis of phenolic compounds in extracts of H. acetosella by high-performance liquid chromatography (HPLC). Next, mice were divided into 8 groups and treated with distilled water or plant extract (0.1 ml/10 g) by gavage for 15 days, followed by intraperitoneal (ip) administration of saline solution or MMS (40 mg/Kg b.w) on day 16. Caffeic acid, following by gallic acid, gallocatechin, coumaric acid, and...

Chronic ethanol consumption in mice does not induce DNA damage in somatic or germ cells, evaluated by the bone marrow micronucleous assay and the dominant lethal mutation assay Biol. Res.
Ellahueñe,Manuel F; Pérez-Alzola,L Patricia; Olmedo,M Isabel.
Although alcohol is known to be a carcinogen for humans, ethanol-genotoxicity studies are incomplete. Ethanol seems not to be a bacterial mutagen, but the results are conflicting in rodent assays. We investigate the genotoxicity in the bone marrow micronucleus (MN) test and in the dominant lethal mutation (DLM) assay using two long-term ethanol exposure protocols. In the MN test, mice consumed three doses (5, 10 and 15% v/v) for 32 weeks. MN induction was compared to two control groups of 5- and 38-week-old mice (the ages of the treated mice when the treatment was initiated and when they were killed, respectively). For the three groups treated with ethanol there was no significant increase in MN induction as compared to the first control group, but...

Evaluation of inorganic and organic bone components after application of an apatite-coated Al2O3 implants as scaffolds for bone repair BABT
Maia Filho,Antonio Luiz Martins; Amaral,Fabrício Pires de Moura do; Martin,Airton Abrahão; Soares,Luís Eduardo Silva.
The aim of this work was to study the influence of uncoated and apatite-coated Al2O3 implants on bone regeneration after 30 days of surgery in New Zealand white rabbits. Twelve samples of Al2O3 implants were prepared and half of them (n = 6) were apatite-coated by the modified biomimetic method. Three experimental groups were tested as Group C - control, surgery procedure without cerami c implant, Group Ce - uncoated Al 2O3 implants (n = 6) and Group CeHA - apatite-coated Al2O3 implants (n = 6). The mineralization of the Al2O3 implants was analyzed in bone fragments using FT-Raman spectroscopy. Raman peaks at 959 cm-1 and 2940 cm-1 evaluated the inorganic and organic bone content, respectively. In vivo citotoxicity was analyzed using micronucleus test....

The mutagenic and antimutagenic effects of the traditional phytoestrogen-rich herbs, Pueraria mirifica and Pueraria lobata BJMBR
Cherdshewasart,W.; Sutjit,W.; Pulcharoen,K.; Chulasiri,M..
Pueraria mirifica is a Thai phytoestrogen-rich herb traditionally used for the treatment of menopausal symptoms. Pueraria lobata is also a phytoestrogen-rich herb traditionally used in Japan, Korea and China for the treatment of hypertension and alcoholism. We evaluated the mutagenic and antimutagenic activity of the two plant extracts using the Ames test preincubation method plus or minus the rat liver mixture S9 for metabolic activation using Salmonella typhimurium strains TA98 and TA100 as indicator strains. The cytotoxicity of the two extracts to the two S. typhimurium indicators was evaluated before the mutagenic and antimutagenic tests. Both extracts at a final concentration of 2.5, 5, 10, or 20 mg/plate exhibited only mild cytotoxic effects. The...
