Antigenic characterization of Brazilian bovine viral diarrhea virus isolates by monoclonal antibodies and cross-neutralization BJMBR
Botton,S.A.; da-Silva,A.M.; Brum,M.C.S.; Weiblen,R.; Flores,E.F..
Nineteen Brazilian isolates of bovine viral diarrhea virus (BVDV) were characterized antigenically with a panel of 19 monoclonal antibodies (mAbs) (Corapi WV, Donis RO and Dubovi EJ (1990) American Journal of Veterinary Research, 55: 1388-1394). Eight isolates were further characterized by cross-neutralization using sheep monospecific antisera. Analysis of mAb binding to viral antigens by indirect immunofluorescence revealed distinct patterns of reactivity among the native viruses. Local isolates differed from the prototype Singer strain in recognition by up to 14 mAbs. Only two mAbs - one to the non-structural protein NS23/p125 and another to the envelope glycoprotein E0/gp48 - recognized 100% of the isolates. No isolate was recognized by more than 14...

Production and characterization of monoclonal antibodies to a Brazilian bovine herpesvirus type 5 BJMBR
Oldoni,I.; Weiblen,R.; Inkelmann,M.A.; Flores,E.F..
Antigens of a bovine herpesvirus type 5 (BHV-5), isolated from a cow with a neurological infection in Rio Grande do Sul State, Brazil, were used to immunize BALB/c mice to produce monoclonal antibodies (mAbs). Eleven hybridomas secreting mAbs directed at BHV-5 antigens were obtained after two fusions and screening of 356 hypoxanthine-aminopterin-thymidine-resistant clones. The mAbs reacted at dilutions up to 1:500 (hybridoma culture supernatant) and up to >1:10,000 (ascitic fluid) in an indirect fluorescent antibody assay (IFA) and in immunoperoxidase staining of BHV-5-infected cells. Four mAbs (1D12, 2E2, 2G10 and 4E4) showed virus-neutralizing activity against the parental BHV-5 isolate. Five mAbs (1F3, 2A6, 2F9, 2G10 and HB24L) reacted in Western...

Production and characterization of monoclonal antibodies to Brazilian isolates of bovine viral diarrhea virus BJMBR
Kreutz,L.C.; Donis,R.; Gil,L.H.V.; Lima,M.; Hoffman,A.N.; Garcez,D.C.; Flores,E.F.; Weiblen,R..
Three Brazilian isolates of bovine viral diarrhea virus (BVDV), antigenically distinct from the standard North American isolates, were selected to immunize BALB/c mice in order to obtain hybridoma cells secreting anti-BVDV monoclonal antibodies (mAbs). Two hybridoma clones secreting mAbs, reacting specifically with BVDV-infected cells (mAbs 3.1C4 and 6.F11), were selected after five fusions and screening of 1001 hypoxanthine-aminopterin-thymidine-resistant clones. These mAbs reacted in an indirect fluorescent antibody (IFA) assay with all 39 South and North American BVDV field isolates and reference strains available in our laboratory, yet failed to recognize other pestiviruses, namely the hog cholera virus. The mAbs reacted at dilutions up to 1:25,600...

Monoclonal antibodies against an outer membrane protein from pathogenic Leptospira BJM
Lüdtke,Charli Beatriz; Coutinho,Mariana Loner; Jouglard,Sandra Denise Dorneles; Moreira,Cecília Nunes; Fernandes,Cláudia Hartleben Pinho; Brod,Claudiomar Soares; Haake,David A.; Ko,Albert Icksang; Dellagostin,Odir Antonio; Aleixo,José Antonio Guimarães.
Two hybridomas secreting monoclonal antibodies (MAbs) that react with a lipoprotein (LipL32) of the outer membrane of pathogenic Leptospira were obtained. For hybridoma production, spleen cells from BALB/c mice imunized with recombinant LipL32 (rLipL32) were fused to SP2/O-Ag14 cells, selected in HAT medium and screened in an indirect ELISA. One MAb produced was of the IgG2b isotype and the other was an IgM. MAbs specificity was confirmed by indirect ELISA and immunoblotting using purified rLipL32 and whole-cell antigen preparations from Escherichia coli (E. coli) expressing LipL32 and from pathogenic and non-pathogenic serovars. Both Mabs reacted with most of the pathogenic serovars tested and none reacted with non-pathogenic Leptospira. The MAbs...

Production and caracterization of monoclonal antibodies for the detection of Salmonella enterica in chicken meat BJM
Schneid,Andréa dos Santos; Lüdtke,Charli Beatriz; Diel,Cristiane; Aleixo,José AntonioGuimarães.
A panel of 13 monoclonal antibodies (MAbs) that react against outer membrane proteins of Salmonella Enteritidis was obtained. Two MAbs were classified as IgM, six were IgG2a, three were IgG3 and one was of the IgG2b isotype. The reactivity of the MAbs against different serovars of Salmonella enterica and other bacteria was investigated using an indirect ELISA. Five MAbs reacted only against Salmonella Enteritidis. Two MAbs presented crossed reactions with thermo-extracted antigens of Klebsiella pneumoniae, Citrobacter freundii and Enterobacter aerogenes. MAb 424H presented wide spectrum of reactivity, detecting antigens of Salmonella belonging to serogroups B, C, D, E and G. The detection limit of different serovars of Salmonella in a indirect ELISA with...

MONOCLONAL ANTIBODIES TO IDENTIFY TOMATO MOSAIC TOBAMOVIRUS (TOMV) BJM
Duarte,Keila M.R.; Gomes,Luiz Humberto; Gesztesi,Jean-Luc; Lopes,José Daniel; Tavares,Flávio C.A..
Monoclonal antibodies were obtained against Tomato mosaic tobamovirus (ToMV) isolated in Brazil. One antibody (8G7G2) isotyped as IgG2b (kappa light chain) showed strong specificity and very low cross reaction with the Tobacco mosaic virus (TMV). It can be used in identification of tomato mosaic virus (ToMV).

Therapeutic monoclonal antibodies: scFv patents as a marker of a new class of potential biopharmaceuticals BJPS
Pucca,Manuela Berto; Bertolini,Thaís Barboza; Barbosa,José Elpidio; Galina,Simone Vasconcelos Ribeiro; Porto,Geciane Silveira.
Monoclonal antibodies represent the fastest growing class of biopharmaceutical products and have a host of applications in medical research, diagnosis, therapy, and basic science. The production of recombinant monoclonal antibodies has revolutionized the generation of immunoglobulins, and their use represents a strategic breakthrough, affecting the global pharmaceutical market for therapeutic proteins. In the present work, a review of scFv, and the number of related patents, has been carried out. The results show that several countries have scFv patents, most notably the United States, China and United Kingdom. The target of these scFv antibodies was also assessed and the results demonstrate that most are directed toward cancer therapy.
