Beneficial Effect of Hyperbaric Oxygen Therapy on the Follicular Survival in the Mouse Ovary after Transplantation OAK
HAYASHI, Satomi; ASANO, Tomoyoshi; KAKIZAKI, Ryujiro; SUZUKI, Hiroshi; 鈴木, 宏志.
A large proportion of follicles are lost during the initial ischemia that occurs after transplantation of ovarian tissues. Thus, the effect of hyperbaric oxygen therapy (HBO) on the follicular loss of ovarian tissues after transplantation was examined in mice. Ovarian slices from ICR mice were transplanted under the kidney capsule in ovariectomized ICR. Hyperbaric oxygen with 100% oxygen was initiated for 30 min at 2.5 atmospheres absolute immediately after transplantation, and this treatment was repeated at 48-h intervals for 2 weeks. The number of follicles was dramatically reduced at 2 weeks post transplantation. However, HBO was significantly effective in enhancing the survival of transplanted ovarian follicles. The survival rates of primordial and...

In Vivo Evidence that Local Cortisol Production Increases in the Preovulatory Follicle of the Cow OAK
ACOSTA, Tomas J.; TETSUKA, Masa; MATSUI, Motozumi; SHIMIZU, Takashi; BERISHA, Bajram; SCHAMS, Dieter; MIYAMOTO, Akio; 松井, 基純; 清水, 隆; 宮本, 明夫.
The aim of the present in vivo study was to monitor real-time fluctuations of cortisol (Cr) in the wall of preovulatory follicles using a microdialysis system (MDS) implanted in the theca layer as well as changes in ovarian venous plasma (OVP) and jugular venous plasma (JVP). Seven cows were superovulated using FSH and prostaglandin F2α injections. Dialysis capillary membranes were surgically implanted into the theca layer of mature follicles and connected to a microdialysis system. Fractions of the perfusates were collected from Day -1 (Day 0=LH surge) to Day 3. No difference in the concentrations of Cr between JVP and OVP was detected throughout the experiment. Circulating concentrations of Cr ranged from 20 to 35 ng/ml 8 h after surgery in ovulatory and...

Promotion of Ovarian Follicular Development by Injecting Vascular Endothelial Growth Factor (VEGF) and Growth Differentiation Factor 9 (GDF-9) Genes OAK
SHIMIZU, Takashi; 清水, 隆.
Ovarian follicular development in mammals is the complex process including endocrine, paracrine and autocrine. There is the development of four basic stages of ovarian follicles, i.e. the primordial, primary, secondary and tertiary or Graafian follicles. There are few blood vessels in the cortical area where primordial and primary follicles are assembled. The development of these follicles is stimulated by oocytes derived factor including growth differentiation factor 9 (GDF-9) or bone morphogenetic protein 15 (BMP-15). Porcine GDF-9 complementary DNA (cDNA) cloned, and then injected its gene into the ovary in gilts. The injection of porcine GDF-9 gene resulted in an increase in the number of primary, secondary and tertiary follicles, concomitant with a...

Expression of Pin1, a peptidyl-prolyl isomerase, in the ovary of eCG/hCG-treated immature female mice. OAK
SHIMIZU, Takashi; AKIYAMA, Hirotada; ABE, Yasuyuki; SASADA, Hiroshi; SATO, Eimei; MIYAMOTO, Akio; UCHIDA, Takafumi; 清水, 隆; 宮本, 明夫.
Protein phosphorylation on certain serine or threonine residues preceding proline (Ser/Thr-Pro) is a pivotal signaling mechanism in diverse cellular processes. Pin1 is a highly conserved enzyme that isomerizes only the phosphorylated Ser/Thr-Pro bonds in certain proteins, thereby inducing conformational changes. Although much protein is phosphorylated in the ovary, the role of Pin1 in the ovary is still unknown. The purpose of this study is to investigate the effects of gonadotropins on protein and mRNA expression of Pin1 in mice ovaries. Quantitative PCR analysis showed that the expression of Pin1 mRNA significantly increased in the ovaries of equine chorionic gonadotropin (eCG)-treated mice compared with those of untreated mice (P<0.05). However,...

Ultrasonography (US) has been applied to the ovary and the uterus of domestic animals from the late 1980s, and established in 1990s as a practical tool for animal production. US made it possible to detect pregnancy at a very early stage and, most importantly, to observe the real-time dynamics of follicular development and hence the discovery of follicular waves. This has greatly contributed to our understanding of ovarian physiology and helped us to develop several "pin-point" protocols for hormonal treatment. While US may not seem to fit preconceived ideas of a "green" technology, it does not contravene environmental priorities, and it is non-invasive ("ethical") and non-hormonal ("clean"). Using the US technology that is now commercially available at a...

Follicular Loss of the Cryopreserved Canine Ovary after Xenotransplantation OAK
Ishijima, Takako; Abe, Yasuyuki; Suzuki, Hiroshi; 鈴木, 宏志.
The effect of cryopreservation and subsequent xenotransplantation on the follicular reserve of the canine ovary by using non obese diabetic-severe combined immunodeficient (NOD-SCID) mice was examined. Vitrified-warmed canine ovarian tissues were placed into the ovarian bursa of mice, and then were removed and subjected to histological examination at 4 weeks after the transplantation. Over 30% of primordial follicles and 65% of early primary follicles survived after cryopreservation. However, regardless of breed or age, percentages of survived primordial follicles and early primary follicles after the transplantation ranged from 0-7% and from 0-15%, respectively. These results indicate that the majority of primordial follicles and early primary follicles...

A comparative study of the ovaries in some Brazilian bees (Hymenoptera; Apoidea) Pap. Avulsos de Zool. (São Paulo)
Martins,Gustavo Ferreira; Serrão,José Eduardo.
The present paper concerns the morphological features of ovaries in 33 species of bees with different social behavior patterns. The ovaries of bees were examined under light microscope. They are polytrophic-meroistic ovaries formed for an anterior germarium and a basal vitellarium. The germarium houses the germ cells and in the vitellarium there are follicles arranged linearly. In general the follicle is constituted by a nutritive chamber (a cluster of nurse cells) and an oocyte chamber, both covered by a single epithelial layer of follicular cells. The number of ovarioles per ovary and the number of mature oocyte per ovary were analyzed. Measurements of ovariole length, oocyte size, oocyte width, follicular epithelial height and the intertegular distance...

Teixeira,RSC; Cardoso,WM; Nogueira,GC; Câmara,SR; Romao,JM; Siqueira,AA; Sampaio,FAC; Moraes,TGV; Campello,CC; Buxade,CC.
This study aimed at evaluating the alternative method of zinc oxide and fasting to induce molt in Japanese quails. A total number of 190 48-week-old quails was used. They were at end of laying cycle, and presented low egg production. Quails molted by zinc oxide (Z) were fed a diet containing 25,000 ppm of zinc oxide, and received water ad libitum. Quails treated by fasting (F) received no feed and a day of water restriction. The treatment period was determined by the experimental level of body weight loss (BWL). Birds were submitted to different levels of BWL in order to analyze reproductive system regression (ovary + oviduct), and livability. The following groups were established according to their BWL: Control (untreated quails); F25 (25% BWL by F); F35...

Effect of Feed Restriction and Photoperiod on Reproduction and LEPR, MELR mRNA Expression of Layers Rev. Bras. Ciênc. Avic.
HAO,E; Chen,H; Ge,S; Huang,R.
ABSTRACT Photoperiod and nutrition are major factors that affect the reproductive efficiency particularly in female animals. In this study we examined the interaction of photoperiod and food restriction on growth, sexual maturation and receptor mRNA expressions of leptin, melatonin, and estrogen in abdominal fat and the ovary of pullets. There were no interaction effects between photoperiod and feeding level on body weight, abdominal fat weight, ovary weight at both 14 wk and 18 wk. Abdominal fat weight of feed restriction group was significantly lower compared with the control group at the age of 14 wk, 18 wk, and age of the first egg (AFE) (p<0.05). Ovary LEPR (Leptin receptor) gene expression showed an interaction effect of the first egg. Restricted...

Yin,L; Ran,J; Lian,T; Yang,C; Li,S; Liu,Y.
ABSTRACT The objective of this study was to analyse the impact of dietary vitamin E supplementation on laying performance, serum reproductive hormones concentration and gene expression in ovary and follicles of anti-season breeding goose. A total of 210 anti-season breeding geese were divided into seven treatments with six replications. Each group was supplied with diets containing different vitamin E (DL-α-tocopherol acetate) contents (0, 10, 20, 40, 80, 160, 320mg/kg). We observed that the egg production and laying rate improved significantly at doses of 10 and 80 mg/kg, while the highest egg weight appeared in the 320 mg/kg group. Meantime, 80 mg/kg of vitamin E supplementation significantly improved the concentration of follicle-stimulating hormone...

Gross morphology and ultrastructure of the female reproductive system of Diaphorina citri (Hemiptera: Liviidae) Rev. Bras. Zool.
Dossi,Fabio Cleisto Alda; Cônsoli,Fernando Luis.
The morphological traits of the female reproductive system of Diaphorina citri were examined in detail. Diaphorina citri has telotrophic ovaries with ovarioles organized as a "bouquet", displaying a rudimentary terminal filament and a syncytial tropharium. The vitellarium carries a single growing oocyte at each maturation cycle, which is connected with the tropharium by a nutritive cord. Morpho-functional changes occur during oocyte development, mainly during mid to late vitellogenesis. Morphological events such as the patency of the follicular cells and the intense traffic of vesicles through para- or intracellular processes, suggest a possible route for endosymbiont invasion of D. citri reproductive tissues. Similar events have been demonstrated to be...

Oocyte development in cattle: physiological and genetic aspects R. Bras. Zootec.
Britt,Jack H..
Oocytes in cattle are formed during embryogenesis and develop within individual follicles in the cortex of the ovary. Dormant primordial follicles become active and undergo progressive development at regular intervals commencing during the late fetal stage and continuing throughout adulthood. Once activated, follicles and oocytes in a cohort either grow to maturation and ovulation or undergo atresia, ultimately depleting the ovaries of germ cells. It takes an estimated 100 days for a follicle and its oocyte to reach the mature ovulatory stage. Within an individual, number of follicles in one ovary is similar to number in the other ovary; however, there are large differences among individuals in total number of follicles present in both ovaries. In...

Morais,Hévea de; Spuri,Renata; Gonçalves,Tarcísio de Moraes; Carvalho,Rafaela Rodrigues de; Andrade,Renato Campos; Pinto,Tássia Louregiani Carvalho; Souza,José Camisão de.
The objective of this study was to investigate whether the use of rbST and eCG prior to ultrasound-guided follicular aspiration (OPU) improves oocyte yield and quality in Tabapuã and Holstein heifers. The study was conducted in two phases, 20 days apart, in a change-over design. The dominant follicle was ablated two days (D-2) before two treatments: stimulation (6 Holstein and 8 Tabapuã), 500 mg of rbST (Boostin®) on D0 and 500 IU of eCG (Folligon) on D2; and control (6 Holstein and 8 Tabapuã), in which heifers received injections of the excipient. Heifers were aspirated on D4. Oocytes were subjected to a well established commercial in vitro embryo production protocol (Vitrogen®) and embryos were evaluated seven days after fertilization. There was an...

Histological characterization of cellular types during Scinax fuscovarius oogenesis (Lutz) (Anura, Hylidae) Zoologia
Oliveira,Classius de; Santos,Lia Raquel de Souza.
This paper describes morphological characteristics of the ovarian germinative cells of the Scinax fuscovarius (Lutz, 1925). The ovary is organized in primordial germinative cells (oogonia) and follicular structures (ovarian follicle) - oocytes surrounded by follicular cells. Oogonia: their nests are peripherically localized, containing cells with large and oval nucleus. Oocytes I: basophilic cytoplasm; the spherical nucleus presents few nucleoli, one or two; the follicular cells (one tenuous layer) surround these previtellogenic cells. Oocytes II: the cell is larger and the cytoplasm becomes more basophilic; the nucleus presents few nucleoli and contains many chromosomes in the periphery (beginning the perinucleolar stage). Oocytes III: the cytoplasm...

Morphological aspects of the ovary of Columba livia (Gmelin) (Columbidae, Columbiformes) Zoologia
Ribeiro,Maria das Graças; Teles,Maria Eloiza de Oliveira; Maruch,Sandra Maria das Graças.
The study of the female genital system of Columba livia (Gmelin, 1789) is part of the research on the reproductive biology of Brazilian birds. This work describes the morphological aspects of the ovary of Columba livia, as well as the histological differences observed in the ovarian follicles during the ovocyte maturation process. Ten Columba livia females were studied, whose ovaries had been dissected, fixed in Bouin and Helly solutions, and histologically processed for staining (Hematoxylin-Eosin; Gomori's trichromic and Weigert's technique and Gomori's technique for connective tissue fibers). The ovary has an irregular surface, is located in the abdominal cavity, and relates to the cranial portion of the left kidney and caudal extremity of the left...

Desenvolvimento morfológico e dos ovários de Kalliapseudes schubarti Mañe-Garzon (Crustacea, Tanaidacea) do Canal de São Sebastião, São Paulo, Brasil Zoologia
Leite,Fosca Pedini Pereira; Leite,Paulo Eduardo Pereira.
The morphological stages of the Kalliapseudes schubarti Mañe-Garzon, 1949 females were easily determinated and they were similar, but the male stages show distint forms probably due to the ambient stimuli. The description of the male stages demonstrate that K. gianucai (Bacescu, 1979) is another male stage of K. schubarti, probably a secondary copulatory stage. The morphology and development of the ovary is related both with the maturity and with the morphological modifications of females stages.

HISTOLOGICAL DESCRIPTION OF THE REPRODUCTIVE SYSTEM OF MALE AND FEMALE HATCHLINGS OF THE MAGDALENA RIVER TURTLE (Podocnemis lewyana) Acta biol.Colomb.
SÁNCHEZ-OSPINA,ANDRÉS C; RODRIGUEZ,BERARDO; CEBALLOS,CLAUDIA P..
In this study we described the macroscopic and microscopic histology of the reproductive system of male and female Podocnemis lewyana neonates (3.5 months old). We found macroscopic differences in the morphology of the gonads between the sexes, with ovaries being twice as long as testes, and testes being twice as wide as ovaries. Microscopically, we identified several immature elements, such as the lack of a muscle layer in the oviduct of females, and simple epithelia instead of pseudostratified epithelia in the oviduct and epididymis described in reptile adults. We also found a black pigment observed macroscopically in the mesovarium, and macroscopically and histologically in the epididymis. This pigment is consistent with the center of melano-macrophages...
