Molecular characterization of parasites of the genus Perkinsus present in clams from French producing areas ArchiMer

Production of clams is the third most important bivalve production in France and was estimated at 3400 metric tons in 2001 among which 60% came from aquaculture and 40% from fisheries. Surveillance of clam diseases mainly relies on histology and thioglycolate culture, two techniques allowing detection of parasites of the genus Perkinsus but not specific enough to determine the parasite species. Based on a previous study aiming at estimating distribution and prevalence of perkinsosis in France, we selected 34 infected clams from 4 different producing areas in order to characterize the parasite species. A first linkage map for the European flat oyster, Ostrea edulis towards the identification of Bonamia resistance QTLs ArchiMer

The flat oyster Ostrea edulis is an endemic European oyster species, found on both Atlantic and Mediterranean coasts. Its aquacultural production has dramatically decreased due to two successive diseases caused by the intracellular parasites Marteilia refringens and Bonamia ostreae. Since 1985, Ifremer has undertaken a breeding program to produce oyster families which are tolerant to Bonamia. In this context, a genetic map was therefore established as a basis for Quantitative Trait Loci (QTLs) analysis. Diverse molecular signatures for ribosomally 'active' Perkinsea in marine sediments ArchiMer

Background: Perkinsea are a parasitic lineage within the eukaryotic superphylum Alveolata. Recent studies making use of environmental small sub-unit ribosomal RNA gene (SSU rDNA) sequencing methodologies have detected a significant diversity and abundance of Perkinsea-like phylotypes in freshwater environments. In contrast only a few Perkinsea environmental sequences have been retrieved from marine samples and only two groups of Perkinsea have been cultured and morphologically described and these are parasites of marine molluscs or marine protists. Recherches sur un protiste, parasite nouveau de l'huître plate des côtes françaises ArchiMer

Following a number of mortalities reported on the flat oyster farms of the Ile Tudy in June 1979, a Protiste, a parasite then unknown in France, was found in the sick oysters. Located mainly in the connective tissue and the branchial epithelium, this parasite is associated with tissue damage. It can be either of a free type or of an intracellular type, both corresponding to small size cells (2 to 3 µm) and displaying, next to their core, one or two mitochondrion and some structured electron-dense particles. Moreover, different mitosis stages, with nuclear membrane persistence, have been observed. These ultrastructural characteristics show some similarities and differences with the Haplosporidies and Marteilia groups.

Essais d'infestations expérimentales entre Crassostrea rhizophorae et Carcinus maenas ArchiMer

Following our aim of developing oyster culture in French Guyana and studying the different options available to increase a stock of new oyster species, we were lead to search for possible parasites in Crassostrea rhizophorae. These oysters from the French Guyanese coasts host some gregarine Nematopsis sp. spores, characterized by Comps, Grizel and Tige (1972). Through this work, we conducted some experimental infestation trials between Crassostrea rhizophorae spats parasited by these spores and the most common crab on metropolitan farms Carcinus maenas.

Bonamiosis due to the intrahaemocytic protistan parasite Bonamia ostreae is a European endemic disease affecting flat oysters Ostrea edulis. The parasite has been described in different ecosystems from estuaries to open sea and no clear correlations could be demonstrated between the development of the disease and environmental parameters such as temperature or salinity. First record of the microsporidian parasite Steinhausia mytilovum in Mytilus sp (Bivalvia : Mytilidae) from France ArchiMer

Steinhausia mytilovuln is a globally distributed microsporidian parasite which infects the oocytes of the blue mussels Mytilus edulis and M. galloprovincialis. Despite the intensive monitoring effort made on mussel populations, the parasite has not previously been reported in France. We report herein on the occurrence of S. mytilovum in Mytilus sp. from 1 cultured and 2 natural populations on the northern coast of France, thus extending the parasite's known distribution northwards. Etude d'un lot de moules transférées dans un centre touché par l'épizootie affectant l'huître plate ArchiMer

L'examen de Mytilus edulis sur coupes histologiques révélant la présence de différents stade de Marteilia refringens, (Grizel et coll. 1974), nous avons essayé de vérifier si ce mollusque était un hôte accidentel ou non de Marteilia refringens. Pour cela, un lot de moules provenant d'une rivière non atteinte par l'épizootie, fut placé en élevage, de Novembre 1974 à Mars 1976, en rivière de Penzé, centre touché par la maladie. Au cours de l'essai, seul un faible pourcentage de moules se parasitait, le lot de moules restait de bonne qualité et les mortalités étaient minimes.

Etude ultrastructurale d'une haplosporidie parasite de la palourde Tapes decussatus L. ArchiMer

The existence of a Haplosporidie parasite in the clam Tapes deucussatus has been demonstrated by Vilela (1950) with the description, based on classical histology observations, of a new species: Haplosporidium tapetis Vilela. Recently, the examination of the histological sections of the digestive glands of some clams from the Thau lagoon helped the scientists to find, in the stomachal epithelium, some forms comparable to the plasmodium of H. tapetis and to precise their ultrastructure.

Caractérisation moléculaire des parasites du genre Mycrocytos ArchiMer

Les différentes productions ostréicoles mondiales sont à l'heure actuelle sérieusement mise à mal par différentes épizooties. Les responsables majeurs de ces maladies, sont des parasites protozoaires. Deux espèces sont responsables de la Bonamiose, une en Europe, Bonamia ostrea et une en Nouvelle Zélande et en Australie, Bonamia sp.. Deux autres parasites provoquent des maladies différentes : Mikrocytos roughleyi qui sévit en Australie et Micrkorcytos mackini au Canada. Ces quatre espèces infectant des zones différentes ne doivent en aucun cas venir coloniser des régions ou pays indemnes. C'est pourquoi il est important de pouvoir arriver à identifier leur présence chez un hôte le plus rapidement possible. Ces protozoaires étant assez proches...

The parasite, Bonamia ostreae, was diagnosed in several South Brittany flat oyster's samples, from gills tissues's prints, stained in May-Grunwald-Giemsa and from corresponding digestive glands sections. The comparaison of results shows quantitative differences relating to the infested oyster's rate, and qualitative differences relating to the early stages of infestation. Gill's print technic seems more intersting for the only Bonamia ostreae diagnosis.

The flat oyster Ostrea edulis L. is the indigenous oyster of the Atlantic as well as the Mediterranean coasts of Europe. Its commercial exploitation dates back to Antiquity but its breeding is now threatened by two parasitic protozoa, among which Bonamia ostreae. Various aspects of the genetics of this species' natural and farmed populations have been studied with the aid of microsatellite markers. At first, the implementation and the screening of two partial genomic libraries made it possible to identify 28 new microsatellite loci. Over the last thirty years, the French production of flat oyster Ostrea edulis dropped from15,000 tons/year to 1,500 tons. Marteilia refringens, a parasite of the flat oyster's digestive gland, is partly responsible for this drop. The fight against this pathogen agent requires a better knowledge of its parasitic cycle. The main hypothesis is the existence of a heteroxenous cycle, that is to say a cycle involving more than one host. However, for thirty years this or these hosts have not been found, and the cycle of Marteilia refringens remains unknown.
