SCORPION VENOM: NEW PROMISE IN THE TREATMENT OF CANCER Acta biol.Colomb.
GÓMEZ RAVE,Lyz Jenny; MUÑOZ BRAVO,Adriana Ximena; SIERRA CASTRILLO,Jhoalmis; ROMÁN MARÍN,Laura Melisa; CORREDOR PEREIRA,Carlos.
ABSTRACT Cancer is a public health problem due to its high worldwide morbimortality. Current treatment protocols do not guarantee complete remission, which has prompted to search for new and more effective antitumoral compounds. Several substances exhibiting cytostatic and cytotoxic effects over cancer cells might contribute to the treatment of this pathology. Some studies indicate the presence of such substances in scorpion venom. In this review, we report characteristics of the principal scorpion venom components found in recent literature and their potential activity against tumor cells. There are different toxin groups present in the venom, and it seems that their mode of actions involves ionic channel blocking, disruption of the cell membrane...

Proteins and Peptides at the Interfaces of Nanostructures Anais da ABC (AABC)
BRITO,ADRIANNE M.M.; BELLETI,ELISANGELA; MENEZES,LUCIVALDO R.; LANFREDI,ALEXANDRE J.C.; NANTES-CARDOS,ISELI L..
Abstract: The present review focuses on the proteins and peptides at the interfaces of nanostructured metals and semiconductors as a result of their use in synthesis in-situ and functionalization of nanostructures. We start the review with an introduction about the peculiar properties of nanostructured materials and their applications. In the following, the chemical and structural properties of peptides and proteins that allow their use as reducing, stabilizing, and functionalization agents are discussed. Proteins and peptides have not only the chemical groups for the metal ion reducing but also provide templates for directing the crystalline growing of nanostructures to the desired shapes and sizes. Proteins and peptides are also used mainly for the...

Histidines 13 and 14 in the Aβ sequence are targets for inhibition of Alzheimer's disease Aβ ion channel and cytotoxicity Biol. Res.
DÍAZ,JUAN CARLOS; LINNEHAN,JOHN; POLLARD,HARVEY; ARISPE,NELSON.
The fact that Alzheimer's beta amyloid (Aβ) peptides forms cation channels in lipid bilayers was discovered during the course of our experiments in the laboratory of "Guayo" Rojas at NIH in Bethesda, Maryland (USA). Recently, we found that the Aβ ion channel could be blocked selectively with small peptides that copy the amino acid sequence of the predicted mouth region of the Aβ channel pore. We now have searched for the essential amino acid residues required for this blocking effect by mutations. We found that the ability of peptides to block Aβ channel activity could be lost by replacement of histidines 13 and 14 by alanine or lysine. The amino acid substitution also resulted in the loss of the capacity of the peptides...

The complete sequence of the mitochondrial genome of the Chinook salmon, Oncorhynchus tshawytscha Biol. Res.
WILHELM,VIVIAN; VILLEGAS,JAIME; MIQUEL,ÁLVARO; ENGEL,ESTEBAN; BERNALES,SEBASTIÁN; VALENZUELA,PABLO D.T.; BURZIO,LUIS O..
The complete sequence of the mitochondrial genome of Chinook salmon, Oncorhynchus tshawytscha, has been determined. The circular genome consisting of 16,644 base pairs encodes thirteen proteins, the 12S and 16S ribosomal RNAs, and 22 transfer RNAs. These genes are ordered in the same way as most other vertebrates. The nucleotide and amino acid sequences of the ribosomal RNAs and the thirteen protein-coding genes were compared with those of other salmonids such as Oncorhynchus mykiss, Salmo salar, Salvelinus fontinalis, Salvelinus alpinus and Coregonus lavaretus. The sequence features of the control region (D-loop), the origin of L-strand replication and a putative peptide codified by the 16S mitochondrial RNA are described and discussed.

Production of Hydrolysate of Okara Protein Concentrate with High Antioxidant Capacity and Aglycone Isoflavone Content BABT
Figueiredo,Vitória Ribeiro Garcia de; Justus,Ariana; Pereira,Dafne Garcia; Georgetti,Sandra Regina; Ida,Elza Iouko; Kurozawa,Louise Emy.
Abstract This work aimed to evaluate the enzymatic hydrolysis of okara protein concentrate with respect to degree of hydrolysis (DH) in order to obtain a protein hydrolysate with high antioxidant capacity and aglycones isoflavone content. A central composite rotatable design was carried out to evaluate the influence of temperature (40 to 70°C), enzyme:substrate ratio (0.5 to 5.0%, g/100g protein) and pH (7.0 to 9.0) on DH. The optimal condition was 55°C, pH 9 and enzyme:substrate ratio of 5.0%, resulting a DH value of 35.5%. After protein hydrolysis at optimal condition, the antioxidant capacities of hydrolysate increased from 58.29 to 383.49 μM Trolox equivalent/g solids (ABTS method) and 2.41 to 15.32 μM Trolox equivalent/g solids (FRAP method) when...

Action of a pancreatin and an Aspergillus oryzae protease on whey proteins: correlation among the methods of analysis of the enzymatic hydrolysates BABT
Morais,Harriman Aley; Silvestre,Marialice Pinto Coelho; Silveira,Josianne Nicácio; Silva,Ana Cristina Simões e; Silva,Viviane Dias Medeiros; Silva,Mauro Ramalho.
The objectives of this study were to hydrolyze whey proteins using a pancreatin and an Aspergillus oryzae protease; to evaluate the degree of hydrolysis (DH) and the peptide profile; and to establish the correlations among the analytical methods. Ten hydrolysates were prepared at different reaction times and the highest DH was obtained by the protein content method. Good correlations (r > 0.87) between the methods of formaldehyde and orthophthalaldehyde (OPA), formaldehyde and osmometry as well as osmometry and OPA were observed using pancreatin. Similar results were obtained between OPA and soluble protein content for the A. oryzae protease. The action of pancreatin produced the highest contents of di- and tripeptides (9.07, 7.12 and 6.46%) and the...

Epitope of dengue virus E protein detect human antibodies associated with mild disease: a potential peptide for vaccine development BJID
Sánchez-Burgos,Gilma; Herrera-Nájera,Carla; Cedillo-Rivera,Roberto; Dumonteil,Eric.
ABSTRACT The antigenic potential of seven immunogenic peptides of the dengue virus was evaluated in the sera of patients with dengue confirmed by IgM/IgG serology. Antibodies IgM and IgG against dengue virus peptides were analyzed by ELISA in 31 dengue sero-positive and 20 sero-negative patients. The P5 peptide showed significant IgG immunoreactivity mostly in the sera of patients with dengue without warning signs in comparison with patients with dengue with warning signs, correlating with mild disease. This finding suggests that the low antibody response against P5 epitope could be a risk factor for higher susceptibility to dengue virus infection with warning signs, and that P5 could be a potential antigen for vaccine development.

Endogenous vasopressin and the central control of heart rate during dynamic exercise BJMBR
Michelini,L.C..
The present article contains a brief review on the role of vasopressinergic projections to the nucleus tractus solitarii in the genesis of reflex bradycardia and in the modulation of heart rate control during exercise. The effects of vasopressin on exercise tachycardia are discussed on the basis of both the endogenous peptide content changes and the heart rate response changes observed during running in sedentary and trained rats. Dynamic exercise caused a specific vasopressin content increase in dorsal and ventral brainstem areas. In accordance, rats pretreated with the peptide or the V1 blocker into the nucleus tractus solitarii showed a significant potentiation or a marked blunting of the exercise tachycardia, respectively, without any change in the...
