The mode of action of Pochonia as a biological control agent is the penetration by means of the hyphae into the target nematode eggs. The egg shell of nematodes has an outer vitelline membrane composed mainly by proteins. Com o objetivo de conhecer as alterações metabólicas promovidas pelo Soil-borne wheat mosaic virus (SBWMV), um dos vírus economicamente mais importantes da cultura do trigo (Triticum aestivum), foram analisados os níveis de proteínas solúveis e determinadas as atividades da peroxidase e da protease em quatro cultivares (BRS Guabiju, BRS 194, BRS 179, BR 23) e uma linhagem (PF 980524) de trigo com diferentes níveis de resistência ao vírus. Os dados obtidos foram submetidos à análise de variância, comparando-se as médias, pelo Teste de Duncan a 5%. As raízes de eucalipto (Eucalyptus urophylla) podem estar associadas a fungos como Pisolithus tinctorius, formando uma simbiose conhecida como ectomicorriza, mas também podem estar colonizadas por fungos patogênicos, como Rhizoctonia solani, agente causal do tombamento de plantas em viveiros. O objetivo deste trabalho foi verificar a presença de atividade inibitória de tripsina, uma serino-protease, em raízes de E. urophylla e a atividade de tripsina em filtrados desses fungos. Seleção de leveduras da Região Amazônica para produção de protease extracelular Acta Amazonica

O objetivo deste trabalho foi identificar 50 leveduras isoladas a partir de diferentes substratos da Região Amazônica e selecionar uma espécie de maior atividade proteolítica. Entre as 26 espécies identificadas predominaram Candida aquatica (12%) e Candida tropicalis (10%). A fermentação submersa foi realizada em Extrato de Malte suplementado com gelatina 1%, Candida intermedia foi a que expressou maior atividade proteolítica, halo = 25 mm e 273 U/mL, crescimento máximo a 30 ºC, viabilidade celular 6,2 x 10(6) UFC, em 48 horas, não demonstrou características de patogenicidade e a melhor densidade do inóculo foi 3%, utilizando-se culturas com 24 horas de crescimento em ágar Malte.

Proteolytic activities of bacteria, yeasts and filamentous fungi isolated from coffee fruit (Coffea arabica L.) - doi: 10.4025/actasciagron.v33i3.6734 Agronomy

One hundred forty-four microorganisms previously isolated from coffee fruit (Coffea Arabica) were grown on casein agar to evaluate their proteolytic activities. Fifty percent of filamentous fungi, 52.5% of bacteria and 2.6% of yeasts were able to secrete proteases. Positive isolates were further examined in liquid culture for their protease activities by hydrolysis of casein at different pH values (5.0, 7.0 and 9.0) at 30 oC. Bacillus megaterium, B. subtilis, Enterobacter agglomerans, Kurthia sp, Pseudomonas paucimobilis and Tatumella ptyseos demonstrated the highest proteolytic activities at pH 9.0. Schistosome serine protease inhibitors: parasite defense or homeostasis? Anais da ABC (AABC)

Serpins are a structurally conserved family of macromolecular inhibitors found in numerous biological systems. The completion and annotation of the genomes of Schistosoma mansoni and Schistosoma japonicum has enabled the identification by phylogenetic analysis of two major serpin clades. S. mansoni shows a greater multiplicity of serpin genes, perhaps reflecting adaptation to infection of a human host. Putative targets of schistosome serpins can be predicted from the sequence of the reactive center loop (RCL). Schistosome serpins may play important roles in both post-translational regulation of schistosome-derived proteases, as well as parasite defense mechanisms against the action of host proteases.

Detection of proteases from Sporosarcina aquimarina and Algoriphagus antarcticus isolated from Antarctic soil Anais da ABC (AABC)

Two psychrophilic bacterial samples were isolated from King George Island soil, in Antarctica. The phylogenetic analysis based on the 16S rRNA (rrs) gene led to the correlation with the closest related isolates as Sporosarcina aquimarina (99%) and Algoriphagus antarcticus(99%), with query coverage of 99% and 98%, respectively.The spent culture media from both isolates displayed proteolytic activities detected by sodium dodecyl sulfate polyacrylamide gel electrophoresis containing gelatin as protein substrate. Under the employed conditions, S. aquimarina showed a 55 kDa protease with the best activity detected at pH 7.0 and at 27°C. Avaliaram-se os efeitos da protease sobre o coeficiente de metabolizabilidade dos nutrientes em dietas contendo farinha de penas (2,0% na fase inicial e 3,0% na fase de crescimento) para frangos de corte machos, Cobb(r), de um a 32 dias de idade. Foram alojadas 336 aves em gaiolas metálicas equipadas com bandeja para coleta total de excretas. Foi utilizado delineamento inteiramente ao acaso, em arranjo fatorial 2x2 (com e sem adição da enzima protease 0,05% e duas valorizações da matriz nutricional da enzima), sendo sete repetições por tratamento, de 12 aves cada. Quinhentos e setenta e seis frangos machos foram distribuídos em 36 gaiolas, com três tratamentos: T1 - composição de milheto sem complexo enzimático; T2 - composição de milheto com complexo enzimático (CES) e T3 - composição de milheto com complexo enzimático (CEV). Kinetics Study of Extracellular Detergent Stable Alkaline Protease from Rhizopus oryzae BABT

In this study, extracellular alkaline protease was produced from Rhizopus oryzae in submerged fermentation using dairy waste (whey) as a substrate. Fermentation kinetics was studied and various parameters were optimized. The strain produced maximum protease at initial medium pH of 6.0 medium depth of 26 mm, inoculum size of 2% at incubation temperature of 35ºC for 168 h of fermentation. Alkaline protease was purified to homogeneity by ammonium sulphate fractionation followed by sephadex G-100 chromatography. The molecular mass of alkaline protease was 69 kDa determined by 10% SDS-PAGE. The optimum pH and temperature of alkaline protease was 9.0 and 40ºC, respectively. Characterization of Nomuraea rileyi strains using polymorphic DNA, virulence and enzyme activity BABT

The characterization of entomopathogenic microorganisms is important for the selection of more effective strains for use in integrated pest-control programs. Five Nomuraea rileyi strains (SA86101, GU87401, SR86151, CG128 and VA9101) were characterized using random amplified polymorphic DNA (RAPD) analysis, virulence studies and assessment of chitinolytic and proteolytic activity. RAPD analysis divided the strains into two groups with a similarity coefficient of 0,76%, group 1 consisting of strains SA86101, GU87401 and SR86151 and group 2 of strains CG128 and VA9101. Peptidase with Keratinolytic Activity Secreted by Aspergillus terreus During Solid-State Fermentation BABT

The aim of this study was to evaluate peptidase production by Aspergillus terreus in solid-state bioprocess and evaluate its parameters. The best conditions were 5.0 g of wheat bran as substrate, incubation temperature 30°C, inoculum 2.0x105spores/g and 75% saline volume, with production reaching 677 U/mL (5400 U/g culture medium) after 72 h of fermentation. Biochemical characterization of the crude enzymatic extract showed the optimum pH and temperature of 6.5 and 55°C, respectively. We investigated the occurrence of HIV-1 antiretroviral resistance in individuals failing to respond to highly active antiretroviral therapy (HAART) attended by RENAGENO from 2001-2004. One hundred and seventeen patients were selected for this study; their plasma viral RNA was extracted and the PR and RT genes sequenced to examine subtype, genetic polymorphisms and mutations associated with resistance to antiretroviral drugs. HIV-1 sequence analysis showed that 86/100 (86%) were infected with subtype B, 7/100 (7%) with subtype F and 7/100 (7%) with RT/PR hybrid forms (2 D/B, 2 F/B, 2 B/F and 1 D/F). This study aims to isolate and identify fungi present in shrimp Litopenaeus vannamei, and in their nursery waters, at two breeding farms in Brazil. Inhibitors of plant proteases can regulate the hydrolysis of proteins inside the cells and also participate in the mechanisms of plant defense against herbivore insects and pathogens. Here, we demonstrated that seeds of Eucalyptus urophylla exhibit activities of trypsin and papain inhibitors, two proteases commonly found in living cells. Low amounts of proteins of the crude protein extract of seeds and fractions partially purified by gel filtration, with inhibitory activity against trypsin, inhibited in vitro the mycelial growth of a compatible isolate of the ectomycorrhizal fungus Pisolithus tinctorius and allowed an unsatisfactory growth of another isolate from Pinus taeda, considered incompatible for this eucalyptus species.
