Colletotrichum truncatum is the most common pathogen fungus associated with soybean anthracnose. Although the lignin-degrading enzyme laccase has been implicated in pathogenicity of a wide range of plant pathogenic fungi, its biological role in the Colletotrichum -soybean disease system is unknown. The extent of the infection in our country led us to examine laccase production in Argentinean Colletotrichum strains isolated from diseased soybean plants from different geographic locations. Ten strains (eight of them identified as C. truncatum) , were screened for in vitro laccase production. Role of Light and Dark Cycle and Different Temperatures in the Regulation of Growth and Protein Expression in Oscillatoria agardhii Strain BABT

The cyanobacterium Oscillatoria agardhii was isolated from the fresh water Mawatha lake, Jaipur and was grown in Zarrouk's medium at 25 ± 2°C, illuminated with white fluorescent light at the intensity of 2 500 lux with 12:12 h light and dark photoperiod. The effects of photoperiod and temperature on the growth and protein expression by Oscillatoria agardhii were studied under different controlled culture conditions (ALR, ALC, CLR, CLC, and NDL), measuring optical density, cell count and dry weight. Detection of Genetic Relationship in the Tree of Life of Some Quail Through Molecular Markers Analyses BABT

ABSTRACT Quail is an important and interesting group of galliform birds. The Common quail (Coturnix coturnix); the Japanese quail (Coturnix japonica); the Panda quail (Coturnix japonica); the Dotted white quail (Coturnix japonica) and the Bobwhite quail (Colinus virginianus) were used in this study. PCR-RFLP and SDS-proteins were performed to reveal the genetic characterization and genetic relationship of the studied quails. Analysis of fragments generated by digestion of PCR product with restriction enzyme NlaIII recorded highly polymorphic restriction profiles. Rhizosphere Bacterial Composition of the Sugar Beet Using SDS-PAGE Methodology BABT

ABSTRACT The rhizosphere zone has been defined as the volume of soil directly influenced by the presence of living plant roots or soil compartment influenced by the root. During the growing season of 2014, the rhizobacteria of 23 sugar beet plants sampled from 12 sites in the west and north west of Iran were inventoried. Using a cultivation-dependent approach, a total of 217 bacteria were isolated from the rhizosphere. The bacterial isolates were tentatively grouped and documented based on polyacrylamide gel electrophoresis of whole-cell proteins and were found to represent 43 different protein electrotypes. Detection of the basement membrane-degrading proteolytic activity of Paracoccidioides brasiliensis after SDS-PAGE using agarose overlays containing Abz-MKALTLQ-EDDnp BJMBR

We have characterized, in the Paracoccidioides brasiliensis yeast phase, an exocellular SH-dependent serine proteinase activity against Abz-MKRLTL-EDDnp and analogous fluorescent-quenched peptides, and showed that it is also active against constituents of the basement membrane in vitro. In the present study, we separated the components of P. brasiliensis culture filtrates by electrophoresis and demonstrated that the serine-thiol exocellular proteinase has a diffuse and heterogeneous migration by SDS-PAGE, localizing in a region between 69 and 43 kDa. The hydrolytic activity was demonstrable after SDS-PAGE using buffered agarose overlays of Abz-MKALTLQ-EDDnp, following incubation at 37oC, and detection of fluorescent bands with a UV transilluminator....

Characterization of cry2-type genes of Bacillus thuringiensis strains from soil-isolated of Sichuan basin, China BJM

Sichuan basin, situated in the west of China, is the fourth biggest basin in China. In order to describe a systematic study of the cry2-type genes resources from Bacillus thuringiensis strains of Sichuan basin, a total of 791 Bacillus thuringiensis strains have been screened from 2650 soil samples in different ecological regions. The method of PCR-restriction fragment length polymorphism (PCR-RFLP) was used to identify the type of cry2 genes. The results showed that 322 Bacillus thuringiensis strains harbored cry2-type genes and four different RFLP patterns were found. The aim of the present research was to evaluate the protein polymorphism degrees among forty-eight C. albicans isolates from fourteen anatomical sites of clinical patients by polyacrylamide gel electrophoresis (SDS-PAGE) and numerical analyzes, in order to identify subspecies and their similarities in some infectious niches. Cell cultures were grown in YEPD medium, collected by centrifugation, and washed in cold saline solution. The whole-cell proteins were extracted by cell disruption using glass beads and submitted to SDS-PAGE technique. Comparison of electrophoretic protein profiles of Campylobacter jejuni subsp. jejuni isolated from different animal species BJM

Electrophoretic protein profiles of Campylobacter jejuni subsp. jejuni strains isolated from feces of seven animal species, including man, were compared. Fourteen strains (two from each species) plus two human strains and the reference one, were ruptured by ultrasound and their total soluble proteins were analyzed by SDS-PAGE technique in a 12% polyacrylamide gel with computerized densitometric reading by the molecular analyst software. All the strains had bands in common that correspond to 45 and 66 Kda molecular weight. The disagreement corresponded to a 97 to 200 Kda molecular weight region. Improved adsorption-desorption extraction applied to the partial characterization of the antilisterial bacteriocin produced by Carnobacterium maltaromaticum C2 BJM

Bacteriocins are ribosomally produced peptides useful for food biopreservation. An improved adsorption-desorption process is proposed for the partial purification of the bacteriocin produced by the fish isolate Carnobacterium maltaromaticum C2. Analyzis of extract by SDS-PAGE indicated this method may offer an alternative to improve the yield of purification of bacteriocins.

The Mycoplasma gallisepticum strains [wild-type S6 (208) and a vaccine-type F-K810] grown in Frey´s and Hayflick´s media were analyzed by SDS-PAGE. No visual changes in the protein profiles of these strains were observed regardless of the media composition used, although the polyacrylamide gel electrophoretograms showed minor differences do exist when densitometer traces of the gel are compared. Both strains were easily differentiated on SDS-PAGE analysis by a peptide band p75, that is specific for MG F-K810 strain, used as vaccine.

Molecular analysis of endophytic bacteria from the genus Bacillus isolated from tropical maize (Zea mays L.) BJM

Endophytic bacteria play an important role in agriculture by improving plant performance and adaptation against biotic and abiotic stresses. In the present study molecular methods were used for identifying Bacillus endophytic bacteria isolated from Brazilian sweet corn. SDS-PAGE of wholecell protein extract of fortytwo isolates revealed a high number of scrutinable bands. Twenty-four isolates were identified in nine different groups of duplicated bacteria and eighteen were identified as unique. Some high-accumulated polipeptides with variable length were observed in almost isolates. Partial sequencing of 16S ribosomal gene revealed that all isolates are Bacillus sp. and among thirteen isolates with similar protein profiles, two were different strains....

Retenção de placenta no proteinograma de vacas Holandesas Ciência Rural

Com o objetivo de avaliar a influência da retenção de placenta (RP) no proteinograma de fêmeas bovinas da raça Holandesa, de propriedades comerciais, foram utilizadas 129 vacas com RP e 145 vacas com parto e pós-parto fisiológicos e sem nenhum tratamento no período avaliado. As amostras de sangue foram divididas nos momentos: 1odia pós-parto (DPP), 2o-3o, 4o-5o, 6o-7o, 8o-14o, 15o-29o, 30o-59o e 60o-90o DPP. O fracionamento das proteínas foi realizado por eletroforese em fita de acetato de celulose e em gel de poliacrilamida, contendo dodecil sulfato de sódio (SDS-PAGE), nas quais se avaliou o comportamento de 19 bandas proteicas identificadas pelos respectivos pesos moleculares, que variaram entre 23KDa e 187KDa. Não houve influência da RP na proteína...

Growth characteristics and dynamics of protein synthesis in callus cultures from Glycine wightii (Wight & Arn.) Verdc. Ciência e Agrotecnologia

Cotyledon explants were first cultured on MS medium supplemented with 4.52 M 2,4-D and 0.46 mM kinetin. The development of the calli was followed (0, 4, 8, 12, 16, 20, 24, 28 and 32 days after) and the growth curve was determined, based in fresh and dry weight. The growth curve presented sigmoidal form with four distinct phases. The highest growth percentage was observed at the exponential phase and the lowest at the stationary phase. These results indicated that cotyledon callus subculture should be performed 20 days after inoculation.
