Genetical improvement of marine fish and shellfish : a french perspective ArchiMer

In France, marine fish and shellfish farming is based mainly on the breeding of wild species, whose natural populations are traditionally exploited. Research programs are conducted by IFREMER, in co-operation with INRA and Universities. French research programs for the genetical improvement of marine fish and shellfish concern primarily seabass (Dicentrarchus labrax) and oysters (Ostrea edulis and Crassostrea gigas). The following programs are developed: •The knowledge of wild and farmed populations. The most recent prodjects involve the highly variable microsatellite markers. The shellfish safety has long been considered a public health problem. The implementation of a regulation based on a bacterial indicator of faecal contamination has significantly reduced outbreaks of bacterial origin. However, epidemics of viral gastroenteritis due to the presence of noroviruses from human sewage persist. We demonstrated few years ago that oysters are not just a passive filter but are able to select some viral strains, via the presence of specific ligands, very similar to ligands observed in humans for these same virus. Ongoing developments, in conjunction with the application of metagenomic tools, will allow us to better understand the fate of some human pathogens after their release into the coastal environment.

Certain species of the marine diatom genus Pseudo‐nitzschia are responsible for the production of the domoic acid (DA), a neurotoxin that can bioaccumulate in the food chain and cause amnesic shellfish poisoning (ASP) in animals and humans. This study extends our knowledge by reporting on the first observation of the potentially toxic species Pseudo‐nitzschia simulans from this region. Official routine monitoring of phycotoxins and toxinproducing algae is regularly implemented in the marine environment through phytoplankton identification in water samples, as well as by official analysis in shellfish in order to verify compliance with the maximum limits set in regulation (EC) no.853/2004. These activities are conducted by the official REPHY network, coordinated by IFREMER (French Research Institute for Exploitation of the Sea). Contamination of Clams with Human Norovirus and a Novel Hepatitis A Virus in Cameroon ArchiMer

Shellfish constitute an important protein source but may be contaminated by viruses from various origins. A study performed on clams collected in Cameroon showed a high prevalence of norovirus and hepatitis A virus. After sequencing, the hepatitis A virus showed similarities with the genotype V simian strains. The need for a one-health approach to protect humans from emerging diseases is evident by the high number of microbial pathogens that become zoonotic following insect bites or consumption of contaminated meat. Potential of F-specific RNA bacteriophages to discriminate sources of faecal pollution in French shellfish ArchiMer

In Europe, shellfish harvesting areas are classified according European Directive EC N°854/2004, using classic faecal indicator levels (Escherichia coli). A better indicator is needed, however, because shellfish may contain other human pathogens. Other indicator organisms have therefore been evaluated and, among these, coliphages meet most of the requirements to serve as indicators and models for enteroviruses. Infectivity of GI and GII noroviruses established from oyster related outbreaks ArchiMer

Noroviruses (NoVs) are the major cause of acute epidemic gastroenteritis in industrialized countries. Outbreak strains are predominantly genogroup II (GII) NoV, but genogroup I (GI) strains are regularly found in oyster related outbreaks. The prototype Norwalk virus (GI), has been shown to have high infectivity in a human challenge study. Whether other NoVs are equally infectious via natural exposure remains to be established. Mathematical models for biological processes are presented as a set of tools for research, which ilave been created for specifie tasks impact studies, energy budget, test of hypothesis. Before using it, the mecanisms involved must be analysed, depending upon wether a whole ecosystem or one animal is studied (biological scale), physical conditions are taken into account (physical scale), with a predetermined scale for time. The no general method for analysis can be, as conditions of studies are so different, but sorne typical cases can show the approach of the modelizer and the recent developments of its tools.

Mr. Jensen noted that "we are committed to the principal that shellfish must be as safe to eat as other ordinary food" and that "the problems of assuring the sanitary quality of coastal areas will increase rather than diminish. Three-year study to assess human enteric viruses in shellfish ArchiMer

The main pathogenic enteric viruses able to persist in the environment, such as hepatitis A virus (HAV), Norwalk-like virus (NLV), enterovirus (EV), rotavirus (RV), and astrovirus (AV), were detected by reverse transcription-PCR and hybridization in shellfish during a 3-year study. Oyster samples (n = 108), occasionally containing bacteria, were less frequently contaminated, showing positivity for AV (17%), NLV (23%), EV (19%), and RV (27%), whereas mussel samples, collected in areas routinely impacted by human sewage, were more highly contaminated: AV (50%), HAV (13%), NLV (35%), EV (45%), and RV (52%). Fishing and trade of living crustaceans are dynamic commercial sectors in France. 25 000 tons of local or imported crabs, lobsters , crawsfishes are yearly treated. Based upon a literature review and fishmongers interviews, this study describes the current pratices and problems associated to live crabs and lobsters storage. Supply conditions of live crustaceans, marketing networks, and biological, ecological and physiological characteristics of crustaceans were discussed. Morever, storage facilities, their operating rules and factors affecting the survival rate under storage and transportation conditions were reviewed.

Exposure to the Neurotoxic Dinoflagellate, Alexandrium catenella, Induces Apoptosis of the Hemocytes of the Oyster, Crassostrea gigas ArchiMer

This study assessed the apoptotic process occurring in the hemocytes of the Pacific oyster, Crassostrea gigas, exposed to Alexandrium catenella, a paralytic shellfish toxins (PSTs) producer. Oysters were experimentally exposed during 48 h to the toxic algae. PSTs accumulation, the expression of 12 key apoptotic-related genes, as well as the variation of the number of hemocytes in apoptosis was measured at time intervals during the experiment. Results show a significant increase of the number of hemocytes in apoptosis after 29 h of exposure. Calcein and manganese experiments for marking the shell of the common cockle (Cerastoderma edule): tidal rhythm validation of increments formation ArchiMer

This work focuses on investigating the potential of calcein and manganese as growth markers of the common cockle (Cerastoderma edule) in the bay of Somme (France). Recapture of shells, previously marked using a chemical marking and then bred in natural conditions, was performed in order to determine the shell growth patterns. Calcein marking has shown a fluorescent increment in shells after only 30 min immersion time at 150 mg L-1, but also for shells immersed 3 h at 50 mg L-1. The Ciguatect super(TM) test kit utilizes a membrane to bind the toxins and a monoclonal antibody-latex bead solution as the determination step. The role of models in assessing the impact of sewage overflows on feacal water contamination ArchiMer

Climate change contributes to local weather modifications such as storm events and heavy rainfall, and in particular, changes in the frequency and intensity of extreme storm and rainfall events. Observations have indicated that rainfall-runoff processes, like sewage overflow may induce fecal contamination in coastal water. Pathogens present in runoff water could therefore contaminate shellfish and be responsible for food-borne disease outbreaks.
