Background: Toxoplasma gondii is an obligate intracellular parasite of the phylum Apicomplexa and a major pathogen of animals and immunocompromised humans, in whom it causes encephalitis. The impact of the host CCR5, TLR2 and TLR11 on production of nitric oxide, IL-6, IL-12 and growth of Toxoplasma gondii OAK

The current study investigated the impact of the CCR5, TLR2 and TLR11 on production of nitric oxide (NO), IL-6 and IL-12 and growth of T. gondii, high virulent RH strain and avirulent PLK strain. All examined knockout macrophages infected with PLK strain produced significant lower levels of IL-12. On the other hand, TLR2 and TLR11 macrophages infected with RH strain produced significantly reduced levels of IL-12 as compared to wild-type macrophages. Evaluation of novel oocyst wall protein candidates of Toxoplasma gondii OAK

Oocyst stage of Toxoplasma gondii is characterized by a durable wall that confers a strong protection to this protozoan parasite in face of harsh environmental conditions. Thus, it is considered the key for transmission of T. gondii. Analysis of oocyst wall composition is mandatory therefore; the aim of this study was to identify novel T. gondii oocyst wall proteins and test their use in detection of these oocysts in environmental samples. Five candidates of novel T. gondii oocyst wall proteins (TgOWPs) were identified and named TgOWP8 through TgOWP12. Background: Toxoplasma gondii is a highly prevalent protozoan that can infect all warm-blooded animals, including humans. Its definitive hosts are Felidae and its intermediate hosts include various other mammals and birds, including pigs. It is found in the meat of livestock which is a major source of human infection. A combination of antigenic regions of microneme proteins have been previously reported as being protective against chronic toxoplasmosis. In this work, we evaluated immune responses induced by immunizing BALB/c and C57BL/6 mice intradermally with plasmid DNA encoding the protein sequences of Toxoplasma gondii AMA1, MIC2, M2AP and BAG1. Mice immunized with the AMA1 gene developed high levels of serum IgG2a and c antibodies as well as cellular immune responses associated with IFN-γ synthesis suggesting a modulated Th1 type of response. The Influence of the regulation of Toxoplasma gondii TgMIC2 transgene on host cell infection OAK

Toxoplasma gondii microneme protein 2 (TgMIC2), an apically stored adhesin, was shown to be a key participant involved in the initial attachment and invasion to a host cell. In this study, we had established the clonal line of T. gondii tachyzoites which over-expressed TgMIC2 using the tetracycline repressor (TetR)-based inducible gene expression system. The TgMIC2 over-expression significantly reduced tachyzoite propagation in vitro (p < 0.002) and significantly reduced the parasite virulence in mice (p = 0.002). The over-expression of exogenous TgMIC2 caused the increase of mRNA expression level of endogenous TgMIC2. Serodiagnosis of Toxoplasma gondii in ducks from Behera Governorate, Egypt OAK

Toxoplasma gondii is an important zoonotic parasite. The diagnosis of infection in animals is an important tool to prevent human infection. In the present study, Toxoplasma gondii was diagnosed by using the modified agglutination test (MAT) in ducks from Behera Governorate, Egypt. The prevalence of Toxoplasma gondii was 13.9%. The prevalence of Toxoplasma gondii was the highest in the Native breed (17.65%) and in 6-8-months age group (19.4%). End-point titer of 1:25, 1:50, and 1:100 were recorded in 61.9%, 19.05%, and 19.05% of the positive samples, respectively. Duck meat is one source of the Toxoplasma gondii infection for human in Egypt.
