Potato yellow vein virus (PYVV) expresses yellowing symptoms on plants and it is transmitted by the whitefly vector Trialeurodes vaporariorum Westwood. No molecular viral detection from the vector extracts has been reported to date. The objectives of this study were I) amplification of the PYVV major coat protein (CP) gene from viruliferous vector extracts by RT-PCR using specific primers, II) cloning and III) sequencing and sequences analysis. Some Colombian Solanum phureja (Jus et Buk) field potato plants expressing yellowing symptoms and confirmed PYVV positive by RT-PCR were used as virus donor plants for feeding whiteflies. ABSTRACT: Severe mosaic symptoms, accompanied by yellow spots, abnormally small leaves, fruit malformation and cracking, reduced plant growth, and high levels of whitefly (Bemisia tabaci MEAM1) infestation were observed in passionflower (Passiflora edulis) orchards in southwestern Bahia, Brazil. The aim of this work was to identify the species of begomovirus infecting the passionflowers, its prevalence in southwestern Bahia, and the spatial and temporal dynamics of the disease. Leaf samples from symptomatic plants collected at 57 orchards located in ten counties were evaluated by PCR for begomovirus infection. Whiteflies are phytophagous insects, whose nymphs and adults suck the phloem sap, causing direct damage due to host plant weakness. In tomato (Lycopersicon spp.) crops, they are important vectors of limiting fitoviruses. The objective of this study was to determine the effects of Lycopersicon spp. genotypes on Bemisia tabaci (Gennadius, 1889) biotype B development under greenhouse conditions. The evaluated genotypes were LA462 (L. peruvianum), LA716 (L. pennellii), LA1584 (L. pimpinellifolium), LA1609 (L. peruvianum), LA1739 (L. hirsutum), P25 (L. esculentum), PI134417 (L. hirsutum f. glabratum) and Santa Clara (L. esculentum). Stunting and stem necrosis were noticed in soybeans (Glycine max) grown in 2000/2001 in West Central Brazil the same condition was also observed in the following year in plantations as far as 2,000 km from the initial area. Based on transmission (mechanical, graft, insect vector), purification and serology, electron microscopy and molecular studies the causal agent was determined to be a whitefly-borne carlavirus which is possibly related to Cowpea mild mottle virus (CpMMV). |

The oviposition preference of Bemisia tabaci biotype B (Homoptera: Aleyrodidae) for bean (Phaseolus vulgaris L.) genotypes containing arcelin in the seeds was evaluated. The tests were carried out under greenhouse conditions, in the dry and wet seasons, with the following genotypes: ARC 3s, ARC 5s (wild genotypes containing arcelin in the seeds); ARC 1, ARC 2, ARC 3, ARC 4 (near isogenic lines containing arcelin in the seeds - EMBRAPA) and Porrillo 70, Bolinha, IAPAR MD 808 (commercial genotypes without arcelin). In the free choice and no-choice oviposition tests, Bolinha and Porrillo 70 genotypes were preferred for oviposition by that insect.
