Nitrogen source and pH interact and modulate lipase secretion in a non-clinical strain of Candida parapsilosis Biological Sciences

Lipases (E.C. 3.1.1.3) are serine-hydrolases, and act on long chain fatty acid ester bonds. They exhibit specific and enantioselective activities, which are desirable for many industrial applications. This study aimed at screening and optimizing the production of lipases by wild yeast strains from a variety of substrates, as well as characterizing the enzyme. An initial selection was made in oxygenated oil-supplemented minimum medium, and the enzymatic activity of the supernatant was tested over p- nitrophenyl palmitate. ABSTRACT A series of arylamidines 3a-j was designed, synthesized and investigated for antimicrobial activity. Structures of the compounds were confirmed by IR, 1H-NMR and 13C-NMR and a 2D spectroscopic study was performed. A preliminary screening of the antimicrobial tests clearly showed that three out of ten arylamidines, viz, 3f, 3g and 3i, were effective against all the gram-negative bacteria: Klebsiella pneumoniae, Pseudomonas aeruginosa and Salmonella enteric; and against the yeast, candida albicans. Further, the Minimum Inhibitory Concentrations (MIC) against the bacteria and yeast were determined. Abstract Yeasts are unicellular fungi widespread in the environment, and studies suggest that there is a positive correlation between yeast and polluted aquatic environments. The aim of this study was to analyze the diversity and resistance of yeasts isolated from water samples collected along the Arroio Dilúvio in Porto Alegre to antifungals. It is also used to produce a wide range of pharmacologically interesting molecules. Several pathogens affect garlic plants, especially Athelia (Sclerotium) rolfsii, a fungus that is widespread and causes large economic losses. It causes direct damage to crops and leads to plant stress, which induces secondary metabolite production in plants. Abstract: This study aimed to evaluate the effect of the commercial probiotic (Saccharomyces cerevisiae), antibiotic (florfenicol), and its combination for Nile tilapia culture. Larvae and adults of Spermologus rufus Boheman can cause severe damage to seeds of Brazilian pine, Araucaria angustifolia. In this study, yeasts were isolated from seeds of A. angustifolia and from the digestive tract of S. rufus to verify their similarity and determine the existence of feeding sites in order to elaborate an artificial diet for this insect. Pichia spp. were found in seeds and Pichia spp. and P. stipitis Pignal in the digestive tract of S. rufus. The low variability among observed microorganisms suggests a food preference, which can facilitate the elaboration of artificial diets.

A mastite representa um dos problemas mais sérios que afetam as fazendas de gado leiteiro. As mastites clínica e subclínica em bovinos, devido a leveduras, são causadas principalmente por espécies do gênero Candida. O objetivo deste trabalho foi identificar quais espécies dessa levedura estavam presentes em amostras de leite obtidas da glândula mamária de vacas apresentando mastite. Um total de 428 amostras de leite foi coletado nos estados de São Paulo, Paraná, Santa Catarina e Rio Grande do Sul. A identificação foi realizada por meio de critérios fenotípicos e fisiológicos. Yeasts discarded in industrial processes can be used as a nutritional supplement and to extract cellular components with biotechnological aims. In this study, the humoral immune response of Swiss mice treated with mannoproteins (MP) from the yeast Saccharomyces uvarum was evaluated. The mice were treated with MPs at different doses and times and inoculated with 2% sheep red blood cells. This work aimed to evaluate nine strains of yeast, previously identified as good producers of lipids in honey medium, for selecting the most suitable strain for the production of lipids in cheese whey medium and compared two well known extraction methods of lipids from the culture medium. The highest yield of total lipids was 1.27 g.L-1 produced by Cryptococcus laurentii 11. A comparison was made between the two culture media: cheese whey and liquid YEPG, and two lipid extraction methods: Bligh and Dyer and Folch et al. for C. laurentii. Production of alcohol by simultaneous saccharification and fermentation of low-grade wheat flour BABT

Two samples of low-grade wheat flour, namely low-grade 1 (LG1) and low-grade 2 (LG2), with different carbohydrate and fibrous content, were used as substrates. The samples were liquefied using various concentrations of alpha- or beta-amylase, in order to optimize the production of fermentable sugars; the enzyme alpha-amylase revealed higher performance. After liquefaction, the simultaneous saccharification and fermentation was conducted in a jar fermentor. Amyloglucosidase was used for saccharification, and dry baker's yeast, S. cerevisiae, for fermentation simultaneously. The objective of this work was to study the production of biomass with copper bioaccumulation in submerged fermentation using sugarcane molasses. Candida pelliculosa BARU 05 isolated from Baru (Dipteryx alata) was selected for its good capacity to accumulate the copper. Fermentation was carried out using the medium composed by sugarcane molasses at 5 °Brix enriched with (g/L) CuSO4.5H2O 0.1; yeast extract, 10.0; (NH4)2SO4, 5.0 ; KH2PO4, 5.0 MgSO4, 0.5, inoculum 10 % of total volume (100 ml), pH 6.0, and incubation at 30 °C, 120 rpm for 120 h. Identification of yeasts Isolated from processed and frozen cocoa (Theobroma cacao) pulp for wine production BABT

The alternative use of cocoa (Theobroma cacao) for wine production was tested. The pulp samples, obtained from Formosa farm, Itacaré, Brazil, were diluted, homogenized and inoculated on Sabouraud dextrose agar medium (SDA) and incubated at 28º C for 5-8 days. Selected colonies were tested for the ability to ferment cocoa pulp and divided into fermentative, non-fermentative and weak/late fermentative species. Isolates characterized as fermentative were further tested in a small-scale wine production plant and identified. Apple wine processing with different nitrogen contents BABT

The aim of this work was to evaluate the nitrogen content in different varieties of apple musts and to study the effect of different nitrogen concentrations in apple wine fermentation. The average total nitrogen content in 51 different apples juices was 155.81 mg/L, with 86.28 % of the values above 100 mg/L. The apple must with 59.0, 122.0 and 163.0 mg/L of total nitrogen content showed the maximum population of 2.05x 10(7); 4.42 x 10(7) and 8.66 x 10(7) cell/mL, respectively. Therefore, the maximum fermentation rates were dependent on the initial nitrogen level, corresponding to 1.4, 5.1 and 9.2 g/L.day, respectively. The nitrogen content in the apple musts was an important factor of growth and fermentation velocity.

ABSTRACT In Brazil, ethanol is obtained by fermentat of sugar cane juice using Saccharomyces cerevisiae. The cane juice extraction generates the bagasse that has been used for obtaining generation biofuel. However, the sugarcane bagasse has 30% pentose that cannot be fermented to ethanol by S. cerevisiae. Thus the aim of this study was to isolate a yeast able to ferment xylose to ethanol. Growth of Nile tilapia Oreochromis niloticus fed diets with different levels of proteins of yeast BABT

This experiment was based on observations of 72 juveniles of Nile tilapia (Oreochromis niloticus), sexually reverted with an initial mean weight of 37.27 ± 4.92g, distributed in 12 cages of 100 l to evaluate the effects of the yeast inclusion as proteins source in the diet. The fishes were distributed in a completely randomized design with four treatments (0; 20; 40; and 60%) of yeast protein in substitution to the protein of traditional sources with three repetitions. Effects of the treatments were not observed (p > 0.05) on the survival and to food conversion. It was observed a quadratic effect on weight gain (Y = 73.39 + 0.173X - 0.0034X²; R²= 0.9986).
