Post-biotinylation of the alkyl azide derivative of trifluoromethyl phenyldiazirine (TPD) was elucidated to apply a photoaffinity biotinylation technique. A photo-modified polyvinilidene difluoride (PVDF) membrane was used as a photolabeled component and we introduced biotin by Staudinger-Bertozzi ligation. The 15 pmol amount of biotinylated reagent was still effective for the visualization of cross-linked product on the matrix. The results show the potential utility of alkyl azide carrying TPD derivatives in the application of photoaffinity biotinylation, which could be useful for the ligands with tight structural requirements. |

During recent decades, milk production per cow has increased drastically due to improved management, nutrition, and genetic selection; however, the reproductive performance of high-producing dairy cows has been declining. One of the factors responsible for this low reproductive performance is negative energy balance (NEB). NEB affects the onset of first ovulation in early postpartum cows. It is generally accepted that early first ovulation positively relates to the resumption of normal ovarian function, first service, and conception rate in dairy cows. タイヤ性能を改善し土壌踏圧を解明するための基礎研究として, タイヤ変形を考慮しての接地面積計測方法を確立した。走行時のタイヤ断面形状を楕円で近似し, タイヤのラジアル方向および横方向変形量, 沈下量, 踏面長さ等の測定値から, タイヤ走行時の3次元接地面積を求めた。また, レーザセンサ距離計により土壌面上部定位置から踏圧面までの距離をメッシュ状に測定できる土壌変形計測装置を開発し, タイヤ静荷重時の3次元接地面積を求めた。これより, 静荷重時接地面積は走行時の約1.3倍となった。さらに, 同装置により静荷重時の土壌深さの違いによる土壌変形量を測定した。その結果, 変形前の土壌の10cmより浅い土壌層にラグによる土壌変形がみられた。 Methods for the determination of 3-D contact area between a tire and soil were presented to improve tire performance and to clarify soil compaction. Effects of the antiparasitic drug moxidectin were studied in laboratory and field experiments in Hokkaido, Japan by pour-on administrations (500 μg/kg) on a target pest Haematobia irritans Linnaeus, nontarget coprophagous flies represented by Neomyia cornicina (Fabricius), and the dung beetle Caccobius jessoensis Harold. The concentration of moxidectin excreted into cattle dung was maximum at 3 days post-treatment both in the first and second trials, and then it diminished. No moxidectin was detected on or after day 21 post-treatment in the first trial, and on or after day 28 post-treatment in the second trial. Acidic electrolyzed water (EW) (pH 2.6-5.8) and alkaline EW (pH 11.2-12.1) were examined as potential disinfectants against foot-and-mouth disease virus (FMDV). Using acidic EW with pH 2.6 and alkaline EW with pH > 11.7, the viral titer decreased in vitro by > 4.0 log values, 2 min after the virus was mixed with EW at a 1: 10 dilution. The strong virucidal effect of acidic EW (pH 2.6), but not that of alkaline EW (> 11.7), seemed to depend on the chlorine level in the solution. Genetic analysis revealed that viral RNA was substantially reduced, especially by alkaline EW. |
