Sea bass Dicentrarchus labrax nervous necrosis virus isolates with distinct pathogenicity to sea bass larvae ArchiMer
Breuil, Gilles; Mouchel, Olivier; Fauvel, Christian; Pepin, Jean-francois.
Reproduction of nodavirus disease was performed by experimental infection of sea bass eggs during fertilization or at larval stage 4 with 2 genetically distinguishable nodavirus strains (Sb1 and Sb2) isolated from sea bass collected along the Atlantic and Mediterranean French coast. The pathogenicity of the virus strains was assigned after detection of the virus by ELISA and immunohistochemistry (IHC). The Atlantic (Sb1) strain was more pathogenic than the Mediterranean (Sb2) strain during the fertilization step whilst both strains were pathogenic following experimental exposure of 4 d old larvae. Virus lesions developed in the brain 4 to 6 d following experimental exposure. Experimental ELISA proved very sensitive for detecting the nodavirus in Sb1 or Sb2...

Rapport de mission aux Etats-Unis du 4 au 22 janvier 1988 ArchiMer
Fauvel, Christian.
During this visit, research work on the physiology and zootechny of reproduction of different US teams were compared to ours. It is clear that we have the same problems to solve and that research programs are rather similar, but they must take into account the local conditions. This fact limits the possibilities of cooperation. Nevertheless a very good knowledge of each work has been noted.

The nme gene family in zebrafish oogenesis and early development ArchiMer
Desvignes, T.; Fauvel, Christian; Bobe, J..
After the recent report of the expression of several nme genes in the zebrafish gonads, the present study aimed at further analyzing the expression of nme genes in the ovary with special attention for the nme transcripts that are maternally inherited and could thus participate in the determination of oocyte developmental competence. The expression levels of all groups I and II nme genes were characterized by QPCR in a panel of zebrafish tissues. The nme genes exhibiting an ovarian expression were subsequently monitored throughout oogenesis and early development, and their expression sites characterized using in situ hybridization. Here, we show that nme2b1, nme3, nme4, and nme6 are highly expressed in the ovary and present in the zebrafish oocyte...

Fertilization strategies for Sea Bass Dicentrarchus labrax (Linnaeus, 1758): effects of pre-incubation and duration of egg receptivity in seawater ArchiMer
Siddique, Mohammad A. Momin; Butts, Ian A. E.; Linhart, Otomar; Macias, Adrian D.; Fauvel, Christian.
Studying gamete biology can provide important information about a species fertilization strategy as well as their reproductive ecology. Currently, there is a lack of knowledge about how long sea bass Dicentrarchus labrax eggs can remain viable after being activated in seawater. The objectives of this study were to understand the effects of pre-incubation of fresh and overripe sea bass eggs in seawater and to determine the duration of egg receptivity. Pooled eggs (fresh and overripe) from four females were pre-incubated in seawater for 0 min (control), 0.5 min, 1 min, 3 min, 10 min and 30 min and then fertilized by pooled sperm from four males. The fresh eggs had a higher fertilization success than overripe eggs. Our results revealed a significant effect of...

Fitness of early life stages in F1 interspecific hybrids between Dicentrarchus labrax and D. punctatus ArchiMer
Ky, Chin-long; Vergnet, Alain; Molinari, Nicolas; Fauvel, Christian; Bonhomme, Francois.
Inter- and intraspecific crossbreeding experiments were conducted to evaluate the aquaculture potential of hybrids in the genus Dicentrarchus, focusing on fertilisation and hatching success. The experimental design consisted of 24 controlled crosses in which individual and pooled fertilisations were made between wild D. labrax individuals (8 dams and 5 sires originating from West Mediterranean and Atlantic populations) and wild D. punctatus (6 sires). Three experiments were successively performed: (1) dams from the Mediterranean population, individually considered to have good egg quality, (2) dams from Mediterranean population, individually considered to have poor egg quality and (3) crossbreeding using pooled eggs from the Atlantic population. In each...

Comparative analysis of male germ cell proliferation and apoptosis in wild and captive Atlantic bluefin tuna (Thunnus thynnus L.) ArchiMer
Zupa, R.; Fauvel, Christian; Mylonas, C. C.; Santamaria, N.; Valentini, L.; Pousis, C.; Papadaki, M.; Suquet, Marc; De La Gandara, F.; Bello, G.; Metrio, G.; Corriero, A..
The most commonly observed reproductive dysfunction in male fishes reared in captivity is reduction in sperm volume and quality. The Atlantic bluefin tuna Thunnus thynnus (Osteichthyes: Scombridae) is one of the few large pelagic and migratory marine fishes maintained in captivity with the purpose of establishing breeding populations to support an aquaculture industry. The objectives of the present study were to compare male germ cell proliferation and apoptosis between wild and captive individuals at two different phases of the spermatogenetic cycle, and to evaluate sperm motility characteristics of captive individuals. Histological observations were performed to analyze testicular activity, and germ cell proliferation and apoptosis were evaluated through...

Reproductive dynamics of swordfish (Xiphias gladius) in the southwestern Indian Ocean (Reunion Island). Part 2: fecundity and spawning pattern ArchiMer
Poisson, Francois; Fauvel, Christian.
Batch fecundity and relative fecundity of swordfish (Xiphias gladius) in the southwestern Indian Ocean were estimated from seven gravid swordfish females (size range 127-225 cm lower jaw-to-fork length, LJFL) with unovulated, hydrated oocytes collected onboard Reunion-based (France) longline swordfish fishing vessels between December 1999 to January 2001. To investigate the spawning pattern of swordfish, we used data collected through a combination of two at sea sampling regimes. A total of 17 007 geo-located size data of swordfish were recorded during 8 years (1993-2001) and a total of 1727 (size range 75-289 cm, LJFL) swordfish gonads (1107 females and 620 males) were sampled from May 1998 to January 2001. The estimated batch fecundity ranged from 995...

La domestication des Gadidés : le cas de la morue et du lieu jaune ArchiMer
Suquet, Marc; Fauvel, Christian; Gaignon, Jean-louis.
The term "domestication" has a legislative meaning when it applies to species submitted to selection pressures, but also a literary one, according to the time since which the species live closed to men and the possibility of controlling their reproduction. In terms of legislation, none of both species of gadids object of this article, cod and pollack, can be considered as domestic. From a literary point of view, 140 years of research and the control of the reproduction, give to the cod a domestic status. It is not the case with pollack, only studied since ten years and the reproduction of which is still badly controlled. The weak differences observed between the performances of breeding and wild cods focuse the still limited impact of the domestication on...

Acoustical monitoring of fish density, behavior, and growth rate in a tank ArchiMer
Conti, Stéphane; Roux, Philippe; Fauvel, Christian; Maurer, Benjamin; Demer, David.
A challenge for the aquaculture community has long been the development of harmless techniques for monitoring fish in a tank. Acoustic telemetry has been used to monitor fish swimming behavior, and passive acoustics have been used to monitor fish feeding, but new techniques are needed to monitor non-invasively their numbers and growth rates. Recently, it has been demonstrated that the acoustical total scattering cross section of fish swimming in a tank can be measured from multiple reverberation time series. These measurements have been used successfully to estimate the number of fish in a tank in laboratory conditions, and to characterize their acoustical signatures. Here, we introduce a novel method for acoustically monitoring fish numerical density and...

Reproductive dynamics of swordfish (Xiphias gladius) in the southwestern Indian Ocean (Reunion Island). Part 1: oocyte development, sexual maturity and spawning ArchiMer
Poisson, Francois; Fauvel, Christian.
The reproductive dynamics of swordfish (Xiphias gladius) was investigated from catches of the Reunion Island-based longline swordfish fishery between 19-25 degrees S and 48-54 degrees E. Thus, 1727 swordfish (size range 75-289 cm, lower jaw-to-fork length, LJFL) were sampled on board commercial fishing vessels during the period of May 1998 to January 2001. Reproductive activity was assessed using macroscopic gonad characteristics, trends of gonadal indexes for both sexes, oocyte size-frequency distributions and microscopic investigation of oocyte development stages. Size at first maturity (L-50) for female and male swordfish was estimated in Indian Ocean for the first time. L50 was estimated at 170 cm and 120 cm (LJFL) for females and males, respectively....

Body reserves mediate trade-offs between life-history traits: new insights from small pelagic fish reproduction ArchiMer
Brosset, Pablo; Lloret, Josep; Munoz, Marta; Fauvel, Christian; Van Beveren, Elisabeth; Marques, Virginie; Fromentin, Jean-marc; Menard, Frederic; Saraux, Claire.
Limited resources in the environment prevent individuals from simultaneously maximizing all life-history traits, resulting in trade-offs. In particular, the cost of reproduction is well known to negatively affect energy investment in growth and maintenance. Here, we investigated these trade-offs during contrasting periods of high versus low fish size and body condition (before/after 2008) in the Gulf of Lions. Female reproductive allocation and performance in anchovy (Engraulis encrasicolus) and sardine (Sardina pilchardus) were examined based on morphometric historical data from the 1970s and from 2003 to 2015. Additionally, potential maternal effects on egg quantity and quality were examined in 2014/2015. After 2008, the gonadosomatic index increased for...

Some characteristics of sperm motility in European hake (Merluccius merluccius, L., 1758) ArchiMer
Groison, A. -l.; Fauvel, Christian; Suquet, Marc; Kjesbu, O. S.; Le Coz, Jean-rene; Mayer, I.; Cosson, J..
P>The objective of this paper is to characterize some of the sperm motility parameters in European hake (Merluccius merluccius), which is considered to be a species with aquaculture potential. The total ATP, ADP and AMP concentrations were determined using high-performance liquid chromatography on hake sperm samples collected during the winter-early spring in the Bay of Biscay (France) (n = 22) and on hake sperm samples collected during the summer-early autumn in waters off Western Norway (n = 5). The Adenylate Energy Charge (AEC) was deduced from these data. Computer Assisted Sperm Analysis (CASA) was used to measure a series of parameters characterizing the motility and the sperm swimming performances. Changes in salinity of the swimming medium...

Genetic Inactivation of European Sea Bass (Dicentrarchus labrax L.) Eggs Using UV-Irradiation: Observations and Perspectives ArchiMer
Colleter, Julie; Penman, David J.; Lallement, Stephane; Fauvel, Christian; Hanebrekke, Tanja; Osvik, Renate D.; Eilertsen, Hans C.; D'Cotta, Helena; Chatain, Beatrice; Peruzzi, Stefano.
Androgenesis is a form of uniparental reproduction leading to progenies inheriting only the paternal set of chromosomes. It has been achieved with variable success in a number of freshwater species and can be attained by artificial fertilization of genetically inactivated eggs following exposure to gamma (gamma), X-ray or UV irradiation (haploid androgenesis) and by restoration of diploidy by suppression of mitosis using a pressure or thermal shock. The conditions for the genetic inactivation of the maternal genome in the European sea bass (Dicentrarchus labrax L.) were explored using different combinations of UV irradiation levels and durations. UV treatments significantly affected embryo survival and generated a wide range of developmental abnormalities....

Egg quality in domesticated and wild seabass (Dicentrarchus labrax): A proteomic analysis ArchiMer
Crespel, Amlie; Rime, Helene; Fraboulet, Erwann; Bobe, Julien; Fauvel, Christian.
In order to improve Our understanding of factors determining egg quality in fish, seabass egg batches obtained from 34 females held under standard conditions, were assessed for biological performances and a fraction from each batch was subjected to proteomic analysis. Comparison of proteomic patterns from eggs of different quality revealed differential protein spots among which one was identified as Nucleoside Diphosphate Kinase, a highly conserved protein involved in cell proliferation. This study shows that molecular markers may help understand factors determining egg quality in fish.

Artificial insemination mastery in red drum, Sciaenops ocellatus, requires a first experimental step wOOng at getting basic biological data. Regarding males, a relation between optical density of the solution (OD) measured by spectrophotometry and sperm concentration (SC) has been established : OD O,161*SC + 0,119. Mean sperm concentration measured is 4,36.10 10 spz.mL-1. This study shows an influence of the solution of dilution on spermatozoa motility and its evolution during short term preservation. As far as females are concemed, the follow-up of final maturation through the observation of samples taken during reguIar strippings, seems to he a good technique to limit overripening and spontaneous liheration of ovules. Analysis of turbot (Scophthalmus maximus) broodstock pilot scale production. ArchiMer
Omnes, Marie-helene; Normant, Yvon; Suquet, Marc; Fauvel, Christian.
Rearing turbot (Scophthalmus maximus) reaches the commercial scale. With the hatchery output increasing, more attention needs to be focussed on broodstocks performance both in terms of quantity and viability of eggs. The techniques of shifting the environmental conditions allow a production of eggs all year round. However, egg quality tends to be highly variable. This paper examines over a two-year period, the improvement in egg quality and its consequences on the production of larvae by a strict management of spawners.
