Individual monitoring of gonad development in the European flat oyster Ostrea edulis by in vivo magnetic resonance imaging 5
Davenel, Armel; Gonzalez, Ricardo; Suquet, Marc; Quellec, Stephane; Robert, Rene.
Previous studies have shown that magnetic resonance imaging is a very appropriate non-invasive technique for quantifying the growth of somatic and gonadic tissues and sex determination in the Pacific oyster, Crassostrea gigas. Despite a thinner gonad, harder to distinguish than that of the Pacific oyster, we showed in this study that it was possible to distinguish and quantify the development of the gonad of the European flat oyster, Ostrea edulis, using a 3D MRI sequence with a suitable finer resolution. Compared to T2-weighted images, theoretically the most appropriate for good anatomical description, T1-weighted images were more suited to gonad observation. The gonad development was quantified by the number of voxels with higher grey level. Larvae were...

Experimental shift of diet and DIC stable carbon isotopes: Influence on shell delta C-13 values in the Manila clam Ruditapes philippinarum 5
Poulain, C.; Lorrain, Anne; Mas, R.; Gillikin, D. P.; Dehairs, F.; Robert, Rene; Paulet, Yves-marie.
The influences of diet and seawater dissolved inorganic carbon (DIC) on the carbon isotope composition of shell aragonite (Cinched in the Manila clam Ruditapes philippinarum reared under laboratory conditions were investigated. Clams were exposed to two successive negative carbon isotope shifts: a first shift in diet (delta C-13(phytoplankton)) and a second shift, 35 days later, in DIC (delta C-13(DIC)). Both successive shifts induced a decrease in delta C-13(shell). These results are the first to experimentally confirm an incorporation of respired carbon derived from food and carbon from DIC into shell carbonate of adult bivalves. Skeletal delta C-13 responded to changes in the delta C-13 of both diet and DIC in less than 7 days. Consequently, proxies...

Scallop larval survival from erythromycin treated broodstock after conditioning without sediment 5
Holbach, Marine; Robert, Rene; Boudry, Pierre; Petton, Bruno; Archambault, Philippe; Tremblay, Rejean.
Pathogenic bacteria are known to be one of the main factors affecting Pecten maximus larval survival in hatcheries. As a result, juvenile production often relies on the use of antibiotics during larval culture. However, limitations of the usage of chemicals such as chloramphenicol in aquaculture have been reinforced due to their negative environmental impact and alternatives are accordingly needed. Thus, the importance of bacterial transfer from oocytes to larvae has been studied here as well as procedures to limit larval mortality in P. maximus. In order to reduce bacterial contamination during larval development, we focused on two periods, broodstock conditioning and post-fertilization. The animals were conditioned for 2 months with two erythromycin...

The effects of eight single microalgal diets on sex-ratio and gonad development throughout European flat oyster (Ostrea edulis L.) conditioning 5
Gonzales, Ricardo; Quillien, Virgile; Robert, Rene.
To determine the effects of food quality on Ostrea edulis reproduction, European flat oysters were conditioned during two sets of experiments, carried out in spring and autumn, during 40 days at 19 degrees C, in 50 l transparent flow-through tanks, in triplicate, and fed constantly at 900 mu m(3) mu l(-1), with eight different types of microalgae. Four species were fed per group of trials: Isochrysis affinis galbana, Chaetoceros gracilis, Skeletonema marinoi, and Tetraselmis suecica were fed to flat oysters in the first set; whereas Rhodomonas salina, Thalassiosira weissflogii, Thalassiosira pseudonana, and Pavlova lutheri were provided during the second set of experiments. At the beginning and end of both conditioning periods, oysters were sampled and...

Influence of phytoplankton diet mixtures on microalgae consumption, larval development and settlement of the Pacific oyster Crassostrea gigas (Thunberg) 5
Rico Villa, Benjamin; Le Coz, Jean-rene; Mingant, Christian; Robert, Rene.
Microalgae commonly used as feed for bivalves, Pavlova lutheri (P), Isochrisis affinis galbana (T) and Chaetoceros calcitrans forma pumilum (Cp), were fed to Pacific oyster Crassostrea gigas to assess their nutritional value for larval development and metamorphosis during two experiments. Monospecific, bispecific and trispecific diets were firstly evaluated during 3 weeks from D larvae to young postlarvae. Then bispecific diets, based on different T and Cp proportions, were assessed during a similar period. Concurrently, ingestion was studied through the whole larval and postlarval development for each diet and/or diet mixture. Because lipids are assumed to be a key nutrient for bivalves, biochemical analysis was undertaken on the second set of trials...

An evaluation of Mg/Ca, Sr/Ca, and Ba/Ca ratios as environmental proxies in aragonite bivalve shells 5
Poulain, C.; Gillikin, D. P.; Thebault, Julien; Munaron, Jean-marie; Bohn, M; Robert, Rene; Paulet, Yves-marie; Lorrain, Anne.
The influence of salinity and water chemistry on Mg/Ca, Sr/Ca and Ba/Ca ratios in the aragonitic shells of the Manila clam were investigated. Clams were reared at constant temperature (20 °C) under different controlled conditions of salinity, commonly encountered in their natural habitat. Clams were held in three tanks with a constant salinity of 35 for the first 35 days and then two tanks were changed to lower salinities (20 and 28) for the next 29 days. Individual shell Mg/Ca, Sr/Ca and Ba/Ca ratios were studied through time. Despite stable conditions (temperature, salinity, and Mg/Cawater) for clams reared at salinity 35 during the experiment, Mg/Ca shell ratios increased through the time. Moreover the salinity decrease at t35 slowed the increase of...

Nutrional inadequacy of Nannochloris atomus and Stichoccocucs bacillaris for the oyster Crassostre gigas (Thunberg) larvae 5
Robert, Rene.
The evaluation of Nannochloris atomliS and StichoccocllS bacillaris as nutrient sources for oyster larvae has been attempted in the present work. After a study of their grawth and size distributions, these two species were used as food for Crassostrea gigas larvae. Despite praper sizes (2.5 to 20 µm3) the larval development of Crassostrea gigas was poor. Evaluation of ingestion and digestion of both algae, by means of epifluorescence microscopy, on 2 day and 6 day old larvae showed that they were normally ingested but weakly digested. Consequently, their nutritional inadequacy for larval molluscs may be also related to their indigestibility.

Pavlova lutheri: Production, preservation and use as food for Crassostrea gigas larvae 5
Ponis, Emanuele; Parisi, G; Chini Zittelli, G; Lavista, F; Robert, Rene; Tredici, M. R..
The haptophyte Pavlova lutheri was cultivated in a 4-l flat alveolar photobioreactor (FAP) in semi-continuous mode during 35 days, achieving a mean volumetric productivity of 0.4 g l-1 day-1. The microalgal culture was harvested from the photobioreactor at an average concentration of 3.5 g l-1 and subjected to different storage conditions in which the effects of temperature (1 °C, 4 °C and 25 °C), air supply and preservative (ascorbic or propionic acid) addition on the viability of the stored suspensions were investigated. Low temperatures (1 and 4 °C) and air-bubbling extended the shelf-life of the stored suspension, while no beneficial effects of ascorbic or propionic acid were found. The influence of temperature and air-bubbling on fatty acid content...

Evaluation of toxicity of Deepwater Horizon slick oil on spat of the oyster Crassostrea virginica 5
Vignier, Julien; Rolton, Anne; Soudant, Philippe; Chu, Fu-lin E.; Robert, Rene; Volety, Aswani K..
The 2010 explosion of the Deepwater Horizon (DWH) oil rig generated the largest marine oil spill in US history with millions of barrels of crude oil released in the Gulf of Mexico (GoM). The eastern oyster, Crassostrea virginica, is an ecologically and economically important species in the northern GoM. Due to its biological characteristics (sessile, filter feeding), juvenile oysters may have been affected. This study investigated the effects of surface-collected DWH oil prepared as high-energy water-accommodated fraction (HEWAF) on the survival of 2-month-old oyster spat, and evaluated the potential impacts of HEWAF on particle clearance rate and spat tissue. Exposure of oysters to a range of oil/HEWAF (0-7-66-147-908-3450 mu g tPAH50 (sum of 50...

Growth, fatness and gross biochemical composition of the japanese oyster Crassostrea gigas in stanway cylinders in the bay of Arcachon, France 5
Robert, Rene; Trut, Gilles; Borel, Michel; Maurer, Daniele.
The Stanway cylinder was tested in the bay of Arcachon from September 1989 to August 1991. Trials were mainly performed on the Japanese oyster Crassostrea gigas in a wind-exposed area. The seed, spat, 18 month old oysters development and fattening of 2 year old oysters were studied. Compared to plastic bags the Stanway cylinder has high positive effects on survival and growth of Crassostrea gigas seed. It also has positive effects on commercial sized Crassostrea gigas fattening. Nevertheless, the cylinder has negative effects on the growth of Crassostrea gigas spat and 18 month old oysters, except for width. However, oyster quality is greatly improve : higher condition index, higher carbohydrate contents and higher shell densities were recorded. Because...

The food value of starch rich flagellates for Pecten maximus (Linné) larvae. Preliminary results 5
Robert, Rene; Moal, Jeanne; Campillo, Maria-josé; Daniel, Jean-yves.
Pecten maximus larvae exhibit amylase as a main digestive enzyme indicating the possibility of starch hydrolysis. The food value of Rhodomonas salina and Porphyridium cruentum, both rich in starch, on the development of Pecten maximus larvae was investigated. The larval development was studied by means of an image analysis technique and epifluorescence microscopy was used for detecting ingestion and digestion of each phytoplankton species. The biochemical contents of algae and larvae were also assessed. The best growth of Pecten maximus larvae was observed when fed with the mixture PTS (plurispecific control). No growth was observed for larvae fed with Porphyridium cruentum of which most of the population died on day 16 (90%). This algal species was...

Effects of hydrodynamic factors on Pecten maximus larval development 5
Holbach, Marine; Robert, Rene; Miner, Philippe; Mingant, Christian; Boudry, Pierre; Tremblay, Rejean.
Hatchery production of great scallop, Pecten maximus, remains unpredictable, notably due to poor larval survival. Large-scale flow-through systems up to 3500 L have been developed to avoid the use of antibiotics in static systems. Alternatively, small-scale flow-through systems have been successfully applied for oysters but they proved to be unsuitable to rear scallop larvae. By focusing on physical factors presumed to limit P. maximus larval development, this study aimed to optimize great scallop larvae rearing parameters under controlled conditions. First, the influence of aeration on larval performances, energetic metabolism and antioxidant defences were studied both in static and flow-through systems. Aeration depressed larval food intake, regardless...

Croissance des larves de Crassostrea gigas et de Mytilus galloprovincialis en présence d'algues monocellulaires isolées du tractus digestif des véligères du milieu naturel 5
His, Edouard; Robert, Rene.
The collection of Crassostrea gigas veligeres in their natural environment and the homogenate technique (HIS et al., 1985) enable the monospecific culture of nine algae which larvae feed on. Four of them could be mass-produced: Nannochloris atomus, Stichococcus bacillaris, Chlamidomonas bullosa and Tetraselmis Tetrathele. Their feeding quality has been tested using the cross-growth method, and using Crassostrea gigas and Mytilus galloprovincialis veligeres. Although regularly found in the homogenates, and thus fed on by larvae, the first three algae gave poor results. Only Tetraselmis tetrathele has good nutritional qualities for Crassostrea gigas veligeres.

Interet du compteur de particules ZB-ZBI et de l'analyseur C1000 pour la numeration des algues unicellulaires de culture. 5
Robert, Rene.
Marine unicellular algae growth rates were studied with: a Coulter Counter in conjunction with a channelizer, and a hemacytometer. Compared to the hemacytometer a good precision of the culture density was observed when the Coulter Counter was used in conjunction with a channelizer. Nevertheless care had to be taken when determining the growth of the diatoms.

Ontogenic changes in the contents of dopamine, norepinephrine and serotonin in larvae and postlarvae of the bivalve Pecten maximus 5
Cann Moisan, Christine; Nicolas, Laurence; Robert, Rene.
In the present study, norepinephrine (NE), epinephrine (E), dopamine (DA) and 5-hydroxy-tryptamine (5HT) have been quantified by high performance liquid chromatography (HPLC) in Pecten maximus during larval and post-larval development. With average values ranging from 2 to 5 pg mug(-1) of protein, NE remained low through the whole larval life, while epinephrine (E) was undetected. DA and 5HT were the most abundant monoamines with significant variations between larval, pre-metamorphic and post-larval stages. During the first 20 days, corresponding to D larval and umboned larval stages, levels of DA and 5HT increased from 15 to 30 and 10 to 15 pg mug(-1) of protein, respectively. Then during the last week of larval life, at the approach of metamorphosis, DA...

Microalgues et nutrition larvaire en écloserie de mollusques 5
Robert, Rene; Trintignac, Pascal.
The production of live microalgae remains the only reliable technique for feeding bivalve mollusc larvae in a controlled environment. However, these algae must answer a number of size, nutritive value and mass-production criteria. The current knowledge in this field is assessed in this work which will lead us to the conclusion that only a few species meet these requirements.

Determination of stocking density limits for Crassostrea gigas larvae reared in flow-through and recirculating aquaculture systems and interaction between larval density and biofilm formation 5
Asmani, Katia; Petton, Bruno; Le Grand, Jacqueline; Mounier, Jerome; Robert, Rene; Nicolas, Jean-louis.
The first aim of this study was to determine the stocking density limits for Pacific oyster Crassostrea gigas larvae reared in flow-through system (FTS) and recirculating aquaculture systems (RAS). The second aim was to examine biofilm formation on the larval tank wall and its interaction with larvae growth. Three larvae concentrations were tested: 50, 150, and 300 mL−1. Chemical parameters and larvae performance were measured. The biofilm was observed by scanning electron microscopy, and its bacterial composition was investigated by pyrosequencing analysis of part of the 16S rRNA gene. The highest growth (13 µm day–1), survival (87%) and metamorphosis (50%) rates were observed in FTS at 50 larvae mL–1, while lower and similar performances occurred at...

Impacts of Deepwater Horizon oil and associated dispersant on early development of the Eastern oyster Crassostrea virginica 5
Vignier, J.; Donaghy, L.; Soudant, P.; Chu, F. L. E.; Morris, J. M.; Carney, M. W.; Lay, C.; Krasnec, M.; Robert, Rene; Volety, A. K..
The explosion of the Deepwater Horizon (DWH) oil platform resulted in large amounts of crude oil and dispersant Corexit 9500A® released into the Gulf of Mexico and coincided with the spawning season of the oyster, Crassostrea virginica. The effects of exposing gametes and embryos of C. virginica to dispersant alone (Corexit), mechanically (HEWAF) and chemically dispersed (CEWAF) DWH oil were evaluated. Fertilization success and the morphological development, growth, and survival of larvae were assessed. Gamete exposure reduced fertilization (HEWAF: EC201 h = 1650 μg tPAH50 L− 1; CEWAF: EC201 h = 19.4 μg tPAH50 L− 1; Corexit: EC201 h = 6.9 mg L− 1).
