Innovative rapid solid-phase immunobead assay for the detection of okadaic acid and related DSP toxins in shellfish ArchiMer

The solid-phase immunobead assay (Ciguatect super(TM)) has been optimized to detect toxins associated with diarrheic shellfish poisoning (DSP). The Ciguatect super(TM) test kit utilizes a membrane to bind the toxins and a monoclonal antibody-latex bead solution as the determination step. The presence or absence of okadaic acid (OA) and related toxins is determined by binding the toxins to a membrane attached to a plastic strip and exposing the toxins ladened membrane to a monoclonal antibody-colored latex bead complex which has a high specificity for OA and related polyether compounds. At 15 degree C, different concentrations of Phaeodactylum tricornutum are given to Crassostrea gigas . Ingestion, filtration, respiration, ammonia excretion have been measured. In 1983, the parasitism of Mytilus edulis L. by Proctoeces maculatus (Loose, 1901) Odhner, 1911 (Trematoda, Digenea, Fellodistomatidae), was recorded for the first time by the authors from mussels farming on stakes (Western Cotentin). After analysing the distribution of the prevalence on the shore and bibliographical data, it was suggested that this parasite must follow a biological cycle that had never been observed on the French coasts and that the most probable host of eggs producer - adult or progenetic metacercaria - would not be a fish but the Prosobranch Gasteropod Nucella lapillus . This paper is a contribution to the study of the Bonamia ostreae epizooty in french european oyster Ostrea edulis : parasite process (hemocytic parasitosis); experimental contaminations on parks and in tanks. First results of an epidemiological watch conducted in oyster culture sites show in molluks mytilidae, pectinidae, cardiidae, veneridae: the absence of Bonamia ostreae forms; the existence of different types of parasitism or commensalisus of inequal pathological importance and especially a parasite of the Marteilia genus in Tapes sp . digestive epithelium. The population of Pinctada margaritifera and Pinctada maculata lay eggs throughout the year. Temperature variations induce huge but asynchronous laying. Preferential periods of gonad maturity correspond to the season changings in both species. However. some differences between the two specics are noticed. Both species are protandric hermaphrodites. Experiments were canied out to assess the copper-chrome-arsenic (CCA) trealment effeets on the Blue mussel Mytilus edulis physiology. Experimental design included two complementary approaches: 1) scope for growth esrimates based upon the energy balance between respiration energy loss and energy gained from feeding processes. 2) a biological early waming system for valve behavior monitoring under CCA exposures. Since 1991, high mortality rates of larvae and juveniles of different bivalve mollusc species associated with herpesvirus infections have been reported in different countries. The aim of this study was to determine if more than one helpesvirus species was responsible for these infections or just one species exhibiting polymorphism. The investigations concentrated on the analysis of the virus genome, studies of virus transmission in different mollusc species and on the potential role of bivalve adults in the occurrence of infections. Caracterisation des hemocytes d''un mollusque bivalve marin, la nacre, Pinna nobilis L. 1758 ArchiMer

The study of tinctorial affinities, in vitro behaviour and ultrastructure of the hemocytes of Pinna nobilis allows us to group them into two main families: basophilic, neutrophilic and acidophilic granulocytes; hyalinocytes, three categories can be distinguished among these cells: small hyalinocytes, hyalinocytes containing smooth endoplasmic reticulum and the macrophage population. The hemocytes participate in the regulation of metabolism by the storage and degradation of glycogen. They contribute to the detoxification process by the sequestration of mineral elements in their lysosomes. Their role in antibacterial defense is suggested by their bacterian phagocytic activity.

Reproduction et recrutement de la coque Cerastoderma edule L. a Saint-Pol-de-Leon (Bretagne Nord) ArchiMer

Investigation on the recruitment of Cerastoderma edule was carried out at St-Pol-de-Leon over three years. This approach is intended to disclose in the fluctuations of recruitment the part of variability linked with reproduction and spawning. Stages of sexual maturity are used together with a condition index. The reproductive activity spread out over the most part of the year, ripe individuals being observed even in winter. Main spawning took place in summer and autumn. Seasonal and interannual fluctuations of recruitment are associated with the variations of the condition index. Bilan energetique d'une population naturelle de Crepidula fornicata dans le bassin de Marennes-Oleron ArchiMer

An energy budget has been evaluated for a natural population of Crepidula fornicata living at low water of spring tides in the oyster farming basin of Marennes-Oleron. This budget of production was estimated according to the position along the chain. The average biomass for an individual representative of the population was equivalent to 1.87 Kjoules, and the annual consumption C was 20.08 Kjoules/year. The energy budget expressed in percent of energy consummates was: C = Pg + Pr + Ps + R + F; 100 = 4.6 + 1.6 + 0.8 + 48 + 45. This budget is compared with literature data on other herbivorous gastropods and with those calculated for Crassostrea gigas in the same locality.

Cockle mortality events have been reported in northern France since 2012. In the present study, we describe and investigate the implication of a potential bacterial causative agent in cockle mortality. Bacteria isolated from five different cockle mortality events were characterized and studied. Using phenotypic analysis combined with DNA–DNA hybridization (DDH) and whole genome sequencing, the isolates were shown to belong to Vibrio aestuarianus , a species regularly detected in France during oyster mortality events. The quality of the scallop embryos is determined by the spawners characteristics as well as by the external factors they support during the maturation and the spawning phases. All these parameters are not still well controlled during the artificial conditioning in the hatcheries. As a consequence, the quality of the embryos is drastically variable and with them, the larvae performances in terms of survival and growth rates. It would be interesting to get a simple and quick test which would allow the estimation of the spawns quality. The germs decreasing of artificially contaminated oysters Crassostrea gigas has been studied in experimental tanks similar to a shellfish depuration plant, without any chemical or physical water treatment. The oyster were treated by three different water management pattern, compared to a control tank and the germs decreasing studied by MPN method of fecal Coliforms and Streptococus. As a result: 1) it appaears somewhat difficult to conclude in this first attempt about the most efficient treatment. 2) the most important factor in shellfish depuration working is obviously the physiological activity (and stage) of the animals.

Viral contamination and depuration of oysters under field conditions ArchiMer

Shellfish consumption is clearly associated with the transmission of enteric viruses. The channel of transmission of enteric viruses to human beings could be decreased particularly if the systems of depuration of shellfish were optimized, standardized and if their viral efficiency was controlled. The purpose of this work was to study the contamination of oysters and to assess the efficiency of several depuration processes. Our study have demonstrated that in some cases, 50% of oysters samples contain enteric viruses or H.A.V. antigen and the quantities of viruses range from 17 to 246 FF for the rotaviruses and from 124 to 200 for the enteroviruses per 100 g of tissue. The behaviour of F specific bacteriophage in depurating shellfish with reference to their use as pollution indicator organisms ArchiMer

The behaviour of sex-pilli specific (F+) bacteriophage during the depuration process was investigated using Pacific oysters (Crassostrea gigas) and edible mussels (Mytilus edulis) naturally contaminated with untreated sewage. F+ bacteriophage was eliminated considerably slower than E. coli. This effect was most pronounced in oysters which showed average T90 bacteriophage values of 62.5 hours compared with 11 hours for E. coli. F+ bacteriophage in mussels was largely confined to the digestive tract. The majority of E. coli were similarly located but, in contrast, counts were also distributed throughout the other internal tissues.
