Purification of the exopolysaccharide produced by Alteromonas infernus: identification of endotoxins and effective process to remove them ArchiMer
Grivaud-le Du, Alicia; Zykwinska, Agata; Sinquin, Corinne; Ratiskol, Jacqueline; Weiss, Pierre; Vinatier, Claire; Guicheux, Jerome; Delbarre-ladrat, Christine; Colliec-jouault, Sylvia.
Alteromonas infernus bacterium isolated from deep-sea hydrothermal vents can produce by fermentation a high molecular weight exopolysaccharide (EPS) called GY785. This EPS described as a new source of glycosaminoglycan-like molecule presents a great potential for pharmaceutical and biotechnological applications. However, this unusual EPS is secreted by a Gram-negative bacterium and can be therefore contaminated by endotoxins, in particular the lipopolysaccharides (LPS). Biochemical and chemical analyses of the LPS extracted from A. infernus membranes have shown the lack of the typical LPS architecture since 3-deoxy-d-manno-oct-2-ulopyranosonic acid (Kdo), glucosamine (GlcN), and phosphorylated monosaccharides were not present. Unlike for other...

Purification procedure for the isolation of a P-I metalloprotease and an acidic phospholipase A 2 fromBothrops atrox snake venom J. Venom. Anim. Toxins incl. Trop. Dis.
Menaldo,Danilo L.; Jacob-Ferreira,Anna L.; Bernardes,Carolina P.; Cintra,Adélia C. O.; Sampaio,Suely V..
Background Snake venoms are complex mixtures of inorganic and organic components, mainly proteins and peptides. Standardization of methods for isolating bioactive molecules from snake venoms is extremely difficult due to the complex and highly variable composition of venoms, which can be influenced by factors such as age and geographic location of the specimen. Therefore, this study aimed to standardize a simple purification methodology for obtaining a P-I class metalloprotease (MP) and an acidic phospholipase A2 (PLA 2 ) from Bothrops atroxvenom, and biochemically characterize these molecules to enable future functional studies. Methods To obtain the toxins of interest, a method has been standardized using consecutive isolation steps. The purity level of...

General characterization of venom from the Moroccan snakes Macrovipera mauritanica and Cerastes cerastes J. Venom. Anim. Toxins incl. Trop. Dis.
Oukkache,N; Lalaoui,M; Ghalim,N.
Ophidian envenomation accidents constitute a serious public health problem in many countries around the globe. Over 5 million such accident cases occur each year causing more than 100,000 deaths. In Africa, more than 20,000 deaths per year are registered while 400,000 envenomation victims retain severe and permanent functional sequelae. In Morocco, snakebites are frequent and of greater severity in children. They occur mostly in rural areas. The incidence of these bites remains poorly understood and vastly underestimated. The epidemiological data are not well known due to the absence of a national registry, whereas a significant proportion of envenomations receive only traditional treatment methods in non-medical intensive care. This prompted us to...

DESENVOLVIMENTO SUSTENTÁVEL E SOJICULTURA EM RORAIMA: TRAJETÓRIAS ANTAGÔNICAS OU CONCILIÁVEIS? AgEcon
Gianluppi, Luciana Dal; Waquil, Paulo Dabdab.

Workshop Report: Advancing the Art of Analyzing Risks and Benefits of Dredged Material Management AgEcon
Wilson, James D..
The U.S. Army Corps of Engineers has been delegated responsibility for assuring the navigability of waters designated for that purpose in the United States. Dredging is among the activities required to fulfill this responsibility. In some places, the Corps finds difficulty in carrying out dredging projects because it lacks spaces where the sediments that must be removed ("dredged materials") can be placed or otherwise managed. In these cases, the public has expressed concerns about the impacts of dredged materials on the environment, either underwater or on land ("upland"). Thus, the Corps often finds it necessary to be able to evaluate and describe the costs or risks and the benefits of the possible alternatives that may be available. Scientists and...

Variability in ribosomal DNA genic and spacer regions in Verticillium dahliae isolates from different hosts Trop. Plant Pathol.
Otero,Laura; Ducasse,Daniel; Miller,Robert N. G..
Using PCR-based assays with specific primers for amplification of the ribosomal DNA intergenic spacer region (IGS) and a portion of the mitochondrial DNA small subunit ribosomal RNA gene (mtDNA SSU rRNA), the genetic variability among Verticillium dahliae isolates from olive (Olea europaea) and other host species from Argentina and Brazil was estimated. The derived UPGMA-generated phenograms based upon the restriction fingerprinting data of rDNA IGS products revealed genetic differences, correlating with the host of origin. Isolates infecting olive genetically distinct from those from cocoa (Theobroma cacao) and sunflower (Helianthus annuus). Digestion of mitochondrial DNA SSU rRNA PCR products revealed less variability, distinguishing only one isolate...
