Oocytes in cattle are formed during embryogenesis and develop within individual follicles in the cortex of the ovary. Dormant primordial follicles become active and undergo progressive development at regular intervals commencing during the late fetal stage and continuing throughout adulthood. Once activated, follicles and oocytes in a cohort either grow to maturation and ovulation or undergo atresia, ultimately depleting the ovaries of germ cells. It takes an estimated 100 days for a follicle and its oocyte to reach the mature ovulatory stage. The objective of this study was to investigate whether the use of rbST and eCG prior to ultrasound-guided follicular aspiration (OPU) improves oocyte yield and quality in Tabapuã and Holstein heifers. The study was conducted in two phases, 20 days apart, in a change-over design. The dominant follicle was ablated two days (D-2) before two treatments: stimulation (6 Holstein and 8 Tabapuã), 500 mg of rbST (Boostin®) on D0 and 500 IU of eCG (Folligon) on D2; and control (6 Holstein and 8 Tabapuã), in which heifers received injections of the excipient. Light and electron microscopy study of late embryonic development in the land snail Limicolaria flammea (Müller) (Pulmonata, Achatinidae) Zoologia

The late stages of embryogenesis in the achatinid land snail Limicolariaflammea (Müller, 1774) were described using light and electron microscopy. Embryos at various stages of development were present in the eggs during the first hour after they were laid, from 4-cell blastulae to morulae and fairly advanced stages. The advanced embryo which was fully developed on the second day bears a long cephalic sac, first to be developed, attached to the embryo and a podocyst which is attached to the foot of the embryo. Both of these structures are reduced in size as embryogenesis progresses until they finally disappear at about the 7th day after the egg was deposited. Embriogenesis and larval ontogeny of the "piau-gordura", Leporinus piau (Fowler) (Pisces, Anostomidae) after induced spawning Zoologia

Mature males and females of Leporinus piau (Fowler, 1941) were induced to reproduce through a hypophysation process. Extrusion occurred 12 h or 312 hours-degree after the hypophysation, at a water temperature of 26°C. Fertilized eggs were maintained in incubators at 24°C. Embryonic development, was evaluated using every 10 minutes, fresh egg samples which were analyzed under stereoscopic microscope. The larvae were collected at 24-hour intervals for seven days after hatching, fixed in Bouin's fluid and were submitted to routine histological techniques. Taxonomic relevance of seed and seedling morphology in two Amazonian species of Entada (Leguminosae) Acta Amazonica

We carried out a comparative morphological study to evaluate the taxonomic value of seed and seedling traits of Entada polystachya and E. simplicata (Leguminosae, Mimosoideae, Mimoseae). Seeds of both species were collected from wild populations in Roraima State, Brazil. Our results show consistent quantitative and qualitative differences between these species regarding their seeds and seedlings. Testa fracture lines, funiculus impression, lens characters, radicle shape and length, and cotyledon lobes length, seedling morphology group, number of pinnae at first node, and hypocotyl length permit a straightforward distinction of E. polystachya from E. simplicata, supporting a recent treatment of the latter taxon as a distinct species, endemic to Roraima...

Although this study aims to develop an improved method for the preservation of reserve lipids in plant tissues for different uses in plant anatomy, it mostly aims to develop an improved method for the identification of lipid reserves where synthesis or storage occurs. The proposed procedures entail only the utilization of (2-hydroxyethyl)-methacrylate (HEMA) as a dehydration agent. The common bean (Phaseolus vulgaris L.) is an important source of copper in human nutrition. The objective of this research was to investigate the possible maternal effects on copper concentrations in the seeds of the common bean, to estimate the heritability and the selection gains in early hybrid generations for the characteristic of copper concentration and to evaluate the potential for genetic improvement. The controlled crossings were performed among IAPAR 44 x IAPAR 31 and Diamante Negro x TPS Bonito cultivars. In order to produce transgenic goats with hG-CSF, a total of 24 adult Saanen and 48 adult undefined breed goats were used as donors and recipients, respectively. Donors were estrus-synchronized with vaginal sponges and superovulated by a treatment with 200 mg FSH given twice daily in decreasing doses over 3 days starting 48 h before sponge removal. Ovulation was induced by injecting 100µg GnRH 36 h after sponge removal. The recipients also received an estrus synchronization treatment. Donors were mated with fertile Saanen bucks and, approximately 72 h after sponge removal, zygotes were recovered surgically by flushing oviducts. Embryonic external morphogenesis of Rhammatocerus conspersus (Bruner) (Orthoptera: Acrididae: Gomphocerinae) and determination of the diapausing embryonic stage Anais da SEB

The grasshopper Rhammatocerus conspersus (Bruner) (Orthoptera: Acrididae: Gomphocerinae) is an occasional pest in pasturelands of Rio Grande do Sul State. It is a univoltine species with an embryonic diapause. Nymphal and adult stages occur during the warmer months (November-March). Eggs were dissected periodically for characterization of embryonic external morphogenesis in 1994 and 1995. Ten embryonic stages were illustrated. Two diapausing stages were verified in R. conspersus: one at 25% and another at 50% of total embryonic development.

ABSTRACT Piau porcine blastocysts were submitted to MALDI-TOF to identify the main phospholipids (PL). After that, in vivo blastocysts (D6) were vitrified (n=52), non-vitrified were used as control (n=42). After warming, blastocysts were in vitro cultured to assess re-expansion and hatching at 24 and 48 hours. Finally, at 48 hours, hatched blastocysts were submitted to RT-qPCR searching for BCL2A1, BAK, BAX and CASP3 genes. For MALDI-TOF, the ion intensity was expressed in arbitrary units. The effect of cryopreservation in IFN-tau, from bovine embryos produced in vitro was evaluated. Two treated groups (G1= fresh bovine embryos, n=59 and G2= freezed embryos, n=84) were used to study the effect of cryopreservation on IFN-tau secretion. After reaching the blastocyst phase, the embryos were kept on individual culture for additional period of 7 days. On days 3 and 7 after the beginning of embryos cultivation, samples of the media culture were taken for IFN-tau secretion titration. Detection of equine herpesvirus 1 DNA in a single embryo and in horse semen by polymerase chain reaction Arq. Bras. Med. Vet. Zootec.

The genome of one equine embryo and three equine semen specimens collected from a Brazilian farm were tested by polymerase chain reaction (PCR) for the presence of EHV-1 and EHV-4-specific timidine kinase (TK) sequences. The PCR detected specific EHV-1 TK gene sequences in all samples tested. The peripheral blood leukocytes (PBL) of the embryo donor mare also was amplified by EHV-1 TK primers. Infectious virus was not recovered from any specimens. The animals did not show any clinical signal of EHV-1 or EHV-4 infections. EHV-4 was not detected in the studied specimens. The results indicate that PCR was more sensitive than virus isolation in cell culture for detecting EHV-1 in semen of carrier horses.

In cattle, embryo development is characterized by the appearance of two distinct cell layers, the trophectoderm and the inner cell mass. The latter will undergo differentiation to form the embryonic disc consisting of the epiblast and hypoblast. The aim of this study was to ultrastructurally characterize the bovine embryo from different in vitro production techniques, with emphasis on trophectoderm and inner cell mass cells. Influence of climatic conditions on in vitro production of bovine embryos Arq. Bras. Med. Vet. Zootec.

Temperature and rainfall were analyzed daily during six years to evaluate their influence on in vitro production of bovine embryos. Weekly replications (n=480) were performed on 14,778 ovaries collected at slaughterhouses. Cumulus oocyte complexes (n=19,180) were fertilized with a pool of Bos taurus taurus semen in one incubator with 5% CO2. Presumable zygotes were cultured in gasified plastic bags with 5% CO2, 5% O2, and 90% N2. In the first year, cleavage and embryo yield were 60.3% and 15.6%, respectively, being lower (P<0.05) than in the following years.
