Monoxenic cultures of arbuscular mycorrhizal fungi (AMF) in transformed roots have been used to better understand the symbiosis with these fungi, but few species have been successfully established in vitro. The object was to establish monoxenic cultures of Gigaspora decipiens and Glomus clarum and to verify the effects of temperature and pH on spore formation. The tree fern Cyathea corcovadensis (Raddi) Domin is an endangered species in the state of Rio Grande do Sul, Brazil. It currently occurs only in the northern segment of the coastal region. Spore storage would help in conservation programs since it maintains genetic variability and provides material for in vitro cultures. Current study evaluates the effect of low temperatures combined to different spore storage times on the germination and initial gametophyte development of C. corcovadensis. The aim of this study was to establish an efficient protocol for the in vitro propagation of the endangered, endemic and decorative species Dianthus pinifolius Sibth. et Sm. The effects of different concentrations of 6-benzylaminopurine (BAP) and naphtalenacetic acid (NAA), and different explant types (single-node cuttings, terminal buds and shoot cuttings) on shoot multiplication were examined on Murashige and Skoog (MS) and half-strength MS media. The best results were obtained for shoot cuttings on the MS medium supplemented with 0.5 mg/L BAP and 0.1 mg/L NAA, achieving a maximum rate of regeneration (100%) and a total of 15.4 newly-developed shoots per explant. The aim of the present research was to verify the in vitro growth of orchids in different systems of micropropagation, being cultivated in a bioreactor, with natural ventilation and conventional systems. Cattleya walkeriana plants were obtained from the germination of seeds in culture medium.
