The soft-shell clam Mya arenaria is one of the most ancient invaders of European coasts and is present in many coastal ecosystems, yet little is known about its genetic structure in Europe. We collected 266 samples spanning a latitudinal cline from the Mediterranean to the North Sea and genotyped them at 12 microsatellite loci. In parallel, geometric morphometric analysis of shell outlines was used to test for associations between shell shape, latitude and genotype, and for a selection of shells we measured the thickness and organic content of the granular prismatic (PR), the crossed-lamellar (CL) and the complex crossed-lamellar (CCL) layers. Strong population structure was detected, with Bayesian cluster analysis identifying four groups located in the... |

Lamellibrachia vestimentiferan tubeworms form aggregations at hydrocarbon cold seeps in the deep Gulf of Mexico (GoM), creating structures that provide living space for other fauna. In the GoM, three Lamellibrachia taxa vary in morphology and depth ranges: Lamellibrachia luymesi (300–950 m), Lamellibrachia sp. 1 (950–2,604 m), and Lamellibrachia sp. 2 (1,175–3,304 m). While Lamellibrachia sp. 2 is consistently identified as a separate species, L. luymesi and sp. 1 cannot be discriminated using barcoding markers cytochrome oxidase subunit 1 (COI) and large ribosomal subunit rDNA (16S). To determine if limited gene flow was a factor in the formation of these taxa, we employed more quickly evolving markers, including mitochondrial cytochrome B (CYTB),... |

The history of colonization and dispersal in fauna distributed among deep-sea chemosynthetic ecosystems remains enigmatic and poorly understood because of an inability to mark and track individuals. A combination of molecular, morphological and environmental data improves understanding of spatial and temporal scales at which panmixia, disruption of gene flow or even speciation may occur. Vestimentiferan tubeworms of the genus Escarpia are important components of deep -sea cold seep ecosystems, as they provide long-term habitat for many other taxa. Three species of Escarpia, Escarpia spicata [Gulf of California (GoC)], Escarpia laminata [Gulf of Mexico (GoM)] and Escarpia southwardae (West African Cold Seeps), have been described based on morphology, but... |

Understanding the factors explaining the observed patterns of genetic diversity is an important question in evolutionary biology. We provide the first data on the genetic structure of a Mediterranean octocoral, the yellow gorgonian Eunicella cavolini, along with insights into the demographic history of this species. We sampled populations in four areas of the Mediterranean Sea: continental France, Algeria, Turkey, and the Balearic and Corsica islands. Along French coasts, three sites were sampled at two depths (20 and 40 m). We demonstrated a high genetic structure in this species (overall F-ST = 0.13), and most pairwise differentiation tests were significant. We did not detect any difference between depths at the same site. Clustering analyses revealed... |

Genetic linkage maps consist of ordering molecular markers across the genome and require a high number of markers for a good coverage of the genome. Such maps represent a framework which enables the identification and localisation of Quantitative Trait Loci (QTLs) for traits of interest, such as growth or disease resistance, with the final aim of achieving genetic improvement through marker-assisted selection (MAS). Data on bivalves are scarce. No genetic map has yet been constructed in any mussel or flat oyster species. We report construction of a preliminary genetic linkage map in the blue mussel, Mytilus edulis (n=14). Amplified Fragment Length Polymorphism (AFLP) markers were used in a mapping family containing 86 full-sibs issued from a controlled... |

Because captivity constitutes a drastic environmental change, domestication is expected to induce a rapid genetic selection for behavioural traits. In this study, we searched for genetic differences in behaviour among brown trout juveniles from two strains differing for their domestication history, i.e. an almost pure native wild Mediterranean population (W) and an Atlantic domesticated strain (D). In order to assess pure genetic differences among strains, males from the two origins were mated with Mediterranean females to produce two experimental crosses (WW and WD). The swimming activity characteristics of individual WW and WD juveniles were compared before and after the application of a stress (light switched off suddenly, followed by a 5-min period of... |

Background: Human intestinal schistosomiasis caused by Schistosoma mansoni and urinary schistosomiasis caused by Schistosoma haematobium are endemic in Ethiopia. Although schistosomes look morphologically uniform, there is variation in infectivity, egg productivity and virulence due to variation in their genetic make. Knowing the genetic diversity and population structure of S. mansoni isolates will enable to understand and consider the possible variability in terms of infectivity, egg productivity and virulence. Methods: Between 2010 and 2011, genetic diversity and population structure of Schistosoma mansoni isolates from four endemic areas of Ethiopia was assessed using previously published 11 polymorphic microsatellite loci. Miracidia were hatched from... |

Cyprinid herpes virus 3 (CyHV-3) diseases have been reported around the world and are associated with high mortalities of koi (Cyprinus carpio). Although little work has been conducted on the molecular analysis of this virus, glycoprotein genes identified in the present study seem to be valuable targets for genetic comparison of this virus. Three envelope glycoprotein genes (ORF25, 65 and 116) of the CyHV-3 isolates from the USA, Israel, Japan and Korea were compared, and interestingly, sequence insertions or deletions were observed in these target regions. In addition, polymorphisms were presented in microsatellite zones from two glycoprotein genes (ORF65 and 116). In phylogenetic tree analysis, the Korean isolate was remarkably distinguished from USA,... |

The reef fish Myripristis berndti (Jordan & Everman 1903) is a pantropical species. A genetic analysis was conducted on 353 individuals from 10 localities distributed across the SW Indian Ocean (SWIO) in order to determine patterns of connectivity in the SWIO. Both the mtDNA sequences (711-bp cytochrome b sequences) and the microsatellites (8 newly developed loci) reveal spatial patterns of differentiation within the SWIO. There is, however, a discrepancy between the structure observed with each kind of marker. MtDNA revealed that 3 peripheral populations (NW Kenya, SE Reunion, and SW Europa) were isolated from the 7 more central populations, which form a more densely connected population network, while microsatellite data indicated a more restricted... |

The European flat oyster Ostrea edulis and the blue mussel Mytilus edulis are both very valuable commercial species across Europe. Despite their economical importance, only a few microsatellite markers have so far been developed in those species. The aim of this study was to develop new microsatellites for O. edulis and M. edulis that could be used for population genetics studies, genetic variability assessment of stocks, parentage analysis or genetic and QTL mapping studies. For Ostrea edulis, an enriched library was made by ecogenics GmbH (Zurich, Switzerland) from size selected genomic DNA ligated into SAULA/SAULB-linker and enriched by magnetic bead selection with biotin-labelled (GT)13 and (CT)13 oligonucleotide repeats (Gautschi et al. 2000a,b). Hybridization between divergent lineages often results in reduced hybrid viability. Here we report findings from a series of independent molecular analyses over several seasons on four life stages of F1 hybrids between the newts Triturus cristatus and T. marmoratus. These two species form a bimodal hybrid zone of broad overlap in France, with F1 hybrids making up about 4% of the adult population. We demonstrate strong asymmetry in the direction of the cross, with one class (cristatus-mothered) making up about 90% of F1 hybrids. By analyzing embryos and hatchlings, we show that this asymmetry is not due to prezygotic effects, as both classes of hybrid embryos are present at similar frequencies, implicating differential selection on the two hybrid classes... |

Monogamy is rare in fishes and is usually associated with elaborate parental care. When parental care is present in fishes, it is usually the male that is responsible, and it is believed that there is a relationship between the high energetic investment and the certainty of paternity (except in the case of sneaker males). Osteoglossum bicirrhosum is considered a monogamous fish, and has particular behavioral traits that permit the study of mating systems and parental care, such as male mouthbrooding. We investigated the genetic relationships of males with the broods found in their oral cavities in Osteoglossum samples collected in a natural environment in the lower Purus river basin, Amazonas, Brazil. Fourteen broods were analyzed for parentage (268 young... |

The rio São Francisco basin contains many endemic species, such as Prochilodus argenteus and P. costatus, which have great commercial importance. However, information about the main recruitment sites and genetic studies containing extensive sampling of these species are scarce. To investigate the roles of the marginal lagoons in the maintenance of genetic variability and in the population structure, we analyzed six microsatellite loci in nine sampling groups of P. argenteusand five sampling groups of P. costatus. Our results showed high levels of genetic variability and low values of genetic differentiation for P. argenteus (FST =0.008, P< 0.05) and for P. costatus(FST =0.031, P < 0.05). In addition, high values of gene flow combined with a small... |

The objective of this work was to estimate the mating system parameters of a andiroba (Carapa guianensis) population using microsatellite markers and the mixed and correlated mating models. Twelve open‑pollinated progeny arrays of 15 individuals were sampled in an area with C. guianensis estimated density of 25.7 trees per hectare. Overall, the species has a mixed reproductive system, with a predominance of outcrossing. The multilocus outcrossing rate (t m = 0.862) was significantly lower than the unity, indicating that self‑pollination occurred. The rate of biparental inbreeding was substantial (t m ‑ t s = 0.134) and significantly different from zero. The correlation of selfing within progenies was high (r s = 0.635), indicating variation in the... |

Abstract: The objective of this work was to identify polymorphic simple sequence repeat (SSR) markers for varietal identification of cotton and evaluation of the genetic distance among the varieties. Initially, 92 SSR markers were genotyped in 20 Brazilian cotton cultivars. Of this total, 38 loci were polymorphic, two of which were amplified by one primer pair; the mean number of alleles per locus was 2.2. The values of polymorphic information content (PIC) and discrimination power (DP) were, on average, 0.374 and 0.433, respectively. The mean genetic distance was 0.397 (minimum of 0.092 and maximum of 0.641). A panel of 96 varieties originating from different regions of the world was assessed by 21 polymorphic loci derived from 17 selected primer pairs.... |
