The shellfish industry is an important economic activity in France, occurring mostly in estuarine zones subject to pollution due to anthropogenic activities. The harmful effects of pollutants on species inhabiting these estuarine zones are not well known. Among marine species, bivalve mollusks---particularly Pacific oyster, Crassostrea gigas-may serve a model of interest. The species is sedentary and filter-feeding, which favors bioaccumulation of pollutants in their tissues. Parental exposure to the herbicide diuron results in oxidative DNA damage to germinal cells of the Pacific oyster Crassostrea gigas ArchiMer

Chemical pollution by pesticides has been identified as a possible contributing factor to the massive mortality outbreaks observed in Crassostrea gigas for several years. A previous study demonstrated the vertical transmission of DNA damage by subjecting oyster genitors to the herbicide diuron at environmental concentrations during gametogenesis. This trans-generational effect occurs through damage to genitor-exposed gametes, as measured by the comet-assay. The presence of DNA damage in gametes could be linked to the formation of DNA damage in other germ cells. The effect of the 2 pesticides most frequently used against mosquitoes was tested on a number of marine organisms. Growth inhibition was tested using Phaeodactylum tricornutum and Gyrosigma spencer (algae). To 0.5 ml of culture inoculum containing similar to 100000 cells in exponential growth phase, 9.5 ml of insecticide in varying concentrations were added and the growth was recorded every 48 hour; LG sub(50) was determined. The coastal areas are subject to multiple anthropogenic pressures including chemical pollution that can pose a real risk to the sustainability of aquatic species. The Arcachon Bay, macrotidal lagoon located on the French Atlantic coast, is the important ecosystem for oyster farming. But for several years, the oyster farms face lower recruitment and high mortality of oyster spat. Rapid method for determining the sensitivity of sulphate-reducing bacteria to biocides. ArchiMer

The Minimal Inhibitory Concentration (MIC) of a biocide is usually determined by adding it in various concentrations to a suitable growth medium which is then inoculated with the test bacterium. These cultures are then observed for up to 28 days for growth of the organism as shown by its production of black ferrous sulphide. A method has been developed whereby the MIC of a biocide can be found within 4 days. Instead of noting ferrous sulphide production, this new method measures changes in: 1) optical density and 2) sulphate reduction of a logarithmic culture of the test strain exposed to the biocide.

Shellfish industry is often realised in estuarine areas, subjected to pollutions due to agricultural activities. The harmful effects of pesticides on animals inhabiting these estuarine zones are poorly documented. Bivalve molluscs represent an interesting model because they are sedentary and they filter water intensively. Effects of pesticides on haemocytes functions were tested in Pacific oyster, Crassostrea gigas and analysed by flow cytometry. A combination of pesticide at environmental concentrations was tested in vivo. Animals used corresponded to two genetically selected populations produced in hatchery.

TOPHYPAC is a field study that was conducted in the Charente river estuary (Marennes-Oléron bay). The main goal was to assess the possible impact of chemical contamination with copper and pesticides on phytoplankton communities. Physical, chemical (nutrients, metals and pesticides) and biological (phytoplankton community) parameters were monitored in the field during three years, from 2011 to 2014. In vitro test-based comparison of pesticide-induced sensitivity in marine and freshwater phytoplankton ArchiMer

This study aims to assess the effects of two pesticides, namely the insecticide carbofuran and the herbicide isoproturon, on monospecifically cultivated marine and freshwater phytoplankton according to standard methods. In the presence of pesticide, growth rates were lower in marine species Chaetoceros gracilis and Phaeodactylum tricornutum than in freshwater species Chlorella vulgaris and Selenastrum capricornutum after 72 hours. The EC50 values were obtained with the REGTOX Macro software, and the NEC values by applying the DEBtox model.

Contamination of some aquatic species with the organochlorine pesticide chlordecone in Martinique ArchiMer

Martinique is a French overseas department whose economy relies heavily on agriculture. Organochlorine pesticides, mainly chlordecone, were used for banana cultivation to eradicate banana weevil over a period of 40 years. Chlordecone is chemically stable,and has a strong affinity for fatty tissues. It is therefore able to bioaccumulate in animals and thereby represent a threat to ecosystems and man. Soils from banana plantations in Martinique are heavily contaminated with chlordecone. Possible transfer of these molecules from agricultural watersheds to the aquatic environment and the organisms that live in it is feared. Des mortalités chez l'huître creuse, Crassostrea gigas, ont été associées au virus OsHV-1 (Oyster herpes virus 1) au stade larvaire et chez des juvéniles. Cependant, aucune mortalité liée à ce virus n'a été rapportée chez les animaux adultes. Cette observation laisse suspecter l'existence de mécanismes dirigés contre ce virus chez cette espèce. Pesticides used in viticulture create a potential risk for the aquatic environment due to drift during application, runoff and soil leaching. The toxicity of sixteen pesticides and one metabolite were evaluated on the growth of two marine microalgae, Tisochrysis lutea and Skeletonema marinoi, in 96-h exposure assays conducted in microplates. For each substance, concentrations of stock solutions were analytically measured and abiotic assays were performed to evaluate the chemical stability of pesticides in microplates. Comet assay in phytoplankton as biomarker of genotoxic effects of environmental pollution ArchiMer

The alkaline comet assay was tested on different microalgae: the clinofiagellates, Karenia mikimotoi and Alexandrium minuturn, and the diatom, Chaetoceros gracilis. The microalgae were exposed during their exponential growth to the model direct genotoxicant, hydrogen peroxide (1 h, 5 and 100 pM H2O2). Following H2O2 exposure, the comet assay was validated only for K. mikimotoi for which genotoxicity was observed from the lowest tested concentration of 5 pM with a concentration-dependent effect. C. gracilis was too small in size (4 mu m) to be correctly analysed. For A. minuturn, our lysis buffer was not strong enough to digest the cellulosic thecal plates. Pesticide contamination of the coastline of Martinique ArchiMer

In January and February 2002, the presence of certain agricultural pesticides throughout the coastline of the Caribbean island of Martinique was investigated. The tropical climate of the French West Indies is suitable for banana production, which requires intensive use of pesticides. An inventory of all pesticides used on the island (compounds and tonnage) was compiled. Surveys and analyses revealed the presence of pesticides in the plumes of seven rivers. The organochlorine chlordecone and metabolites of aldicarb were detected at nearly all of the monitored sites, even though the use of chlordecone has been prohibited since 1993. Combination of a pesticide exposure and a bacterial challenge: In vivo effects on immune response of Pacific oyster, Crassostrea gigas (Thunberg) ArchiMer

To assess the impact of pollution induced by pesticides on Pacific oyster, Crassostrea gigas, health in France, in vivo effects of combined pesticide exposure and bacterial challenge on cell activities and gene expression in hemocytes were tested using flow cytometry and real-time PCR. Characterisation and expression of four mRNA sequences encoding glutathione S-transferases pi, mu, omega and sigma classes in the Pacific oyster Crassostrea gigas exposed to hydrocarbons and pesticides ArchiMer

Hydrocarbon and pesticide pollution in coastal ecosystems can disturb marine bivalve metabolism. In this study, we characterised four full-length cDNA sequences encoding glutathione S-transferases (GSTs) in the Pacific oyster Crassostrea gigas. A BLAST X search showed that these four sequences encode GSTs from four different classes: GST pi, sigma, mu and omega. A phylogenetic analysis of GST was made to determine the position of oyster GST compared to invertebrate and vertebrate sequences.
