Estradiol and progesterone fecal metabolites analysis in crab-eating-fox (Cerdocyoun thous) Arq. Bras. Med. Vet. Zootec.
Paz,R.C.R.; Souza,N.P.; Furtado,P.V.; Brown,J.L..
ABSTRACT In this study, four crab-eating fox females (Cerdocyon thous) maintained at the Federal University of Mato Grosso Zoo, Cuiabá, Brazil, were investigated for 12 months, using feces measurement of estradiol and progesterone concentrations. Fecal collections were performed three times a week for hormone extraction. Two methods of analysis, Elisa (EIA) and Radioimmunoassay (RIA), were used in the measurement of progesterone (P4) and estradiol (E2) metabolites. The aim of this study was to compare and validate two different methods of hormone measurement for C. thous. There were no differences regarding the method used. The Radioimmunoassay technique proved to be more sensitive, however, both showed similar results.

Proacrosin-acrosin activity in capacitated and acrosome reacted sperm from cryopreserved bovine semen Biocell
Rosatti,M. I.; Beconi,M. T.; Córdoba,M..
Acrosin activity is associated with normal fertility in human and bovine spermatozoa. The aim of the study was to determine the variation of the enzyme activity in the proacrosin-acrosin system in capacitated and acrosome reacted cryopreserved bovine sperm. Enzyme activity was assessed spectrophotometrically using N-a-benzoyl-DL-arginine p-nitroanilide (BAPNA) as specific substrate for acrosin at pH 8. Capacitation with heparin and quercitin failed to induce conversion of proacrosin to acrosin. An increase in acrosin activity produced by the presence of progesterone, in a dose-dependent manner, was related with the induction of true acrosome reaction. The total level of acrosin activity registered showed that 96% of acrosin of capacitated sperm samples and...

Glucose transporters: expression, regulation and cancer Biol. Res.
MEDINA,RODOLFO A.; OWEN,GARETH I..
Mammalian cells depend on glucose as a major substrate for energy production. Glucose is transported into the cell via facilitative glucose transporters (GLUT) present in all cell types. Many GLUT isoforms have been described and their expression is cell-specific and subject to hormonal and environmental control. The kinetic properties and substrate specificities of the different isoforms are specifically suited to the energy requirements of the particular cell types. Due to the ubiquitousness of these transporters, their differential expression is involved in various disease states such as diabetes, ischemia and cancer. The majority of cancers and isolated cancer cell lines over-express the GLUT family members which are present in the respective tissue of...

Post partum reproductive assessment in lowland Tapir (Tapirus terrestris): a case report Braz. J. Vet. Res. Anim. Sci.
OLIVEIRA,Cláudio Alvarenga de; NOGUEIRA,Guilherme de Paula; CASTRO,João César Bedran de.
The lowland tapir is the biggest Brazilian terrestrial mammal, which belongs to the order Perissodactyla, suborder Hippomorpha, superfamily Tapiroidea and a member of the family Tapiridae. At tropical forest the tapir is involved with seed dispersal. The knowledge of this wild animal reproductive cycle is one way to help its preservation. The stress due to restrain of captive or free-ranging wild animal in order to sample collection limits endocrine study once it can be hazard for the estrous cycle. One possibility is to quantify gonadal hormones at the excreta. Progesterone milk levels were studied in a tapir housed at the Araçatuba Zoo, in São Paulo, Brazil. Milk samples, vaginal cytology and rectal temperature were collected during lactation. The...

Evaluation of extraction methods for progesterone determination in rabbit (Oryctolagus cuniculus) feces by radioimmunoassay Braz. J. Vet. Res. Anim. Sci.
KORNDÖRFER,Clotilde Maria; MEIRELLES,Cyro Ferreira; BUENO,Ives Cláudio da Silva; ABDALLA,Adibe Luiz.
The purpose of this study was to find a practical procedure for the extraction of progesterone (P4) from feces and to determine if the P4 plasma profiles during pregnancy were reflected in the total fecal P4 of pregnant rabbits. The rabbit was used as model for the techniques. Plasma and feces were collected from 11 rabbits during a period of 42 days. Three different methods of P4 extraction were used. The total P4 was measured by solid-phase radioimmunoassay (RIA) with 125I-P4 as the tracer. Results suggested that it was possible to extract total P4 from rabbit feces with methanol and petroleum ether. Plasma and fecal P4 profiles were compared for both pregnant and ovariectomized rabbits. It was possible to differentiate total P4 extracted from day two...

Influence of progesterone on GAD65 and GAD67 mRNA expression in the dorsolateral striatum and prefrontal cortex of female rats repeatedly treated with cocaine BJMBR
Souza,M.F.; Toniazo,V.M.; Frazzon,A.P.G.; Barros,H.M.T..
Female rats are intensely affected by cocaine, with estrogen probably playing an important role in this effect. Progesterone modulates the GABA system and attenuates the effects of cocaine; however, there is no information about its relevance in changing GABA synthesis pathways after cocaine administration to female rats. Our objective was to investigate the influence of progesterone on the effects of repeated cocaine administration on the isoenzymes of glutamic acid decarboxylase (GAD65 and GAD67) mRNA in brain areas involved in the addiction circuitry. Ovariectomized, intact and progesterone replacement-treated female rats received saline or cocaine (30 mg/kg, ip) acutely or repeatedly. GAD isoenzyme mRNA levels were determined in the dorsolateral...

Estrogen and progesterone promote breast cancer cell proliferation by inducing cyclin G1 expression BJMBR
Tian,J-M.; Ran,B.; Zhang,C-L.; Yan,D-M.; LI,X-H..
Breast cancer is the most common cause of cancer among women in most countries (WHO). Ovarian hormone disorder is thought to be associated with breast tumorigenesis. The present study investigated the effects of estrogen and progesterone administration on cell proliferation and underlying mechanisms in breast cancer MCF-7 cells. It was found that a single administration of estradiol (E2) or progesterone increased MCF-7 cell viability in a dose-dependent manner and promoted cell cycle progression by increasing the percentage of cells in the G2/M phase. A combination of E2 and progesterone led to a stronger effect than single treatment. Moreover, cyclin G1 was up-regulated by E2 and/or progesterone in MCF-7 cells. After knockdown of cyclin G1 in MCF-7 cells...

Different stress modalities result in distinct steroid hormone responses by male rats BJMBR
Andersen,M.L.; Bignotto,M.; Machado,R.B.; Tufik,S..
Since both paradoxical sleep deprivation (PSD) and stress alter male reproductive function, the purpose of the present study was to examine the influence of PSD and other stressors (restraint, electrical footshock, cold and forced swimming, N = 10 per group) on steroid hormones in adult Wistar male rats. Rats were submitted to chronic stress for four days. The stressors (footshock, cold and forced swimming) were applied twice a day, for periods of 1 h at 9:00 and 16:00 h. Restrained animals were maintained in plastic cylinders for 22 h/day whereas PSD was continuous. Hormone determination was measured by chemiluminescent enzyme immunoassay (testosterone), competitive immunoassay (progesterone) and by radioimmunoassay (corticosterone, estradiol, estrone)....

Effect of sildenafil (Viagra®) on the genital reflexes of paradoxical sleep-deprived male rats BJMBR
Andersen,M.L.; Bignotto,M.; Tufik,S..
Since there is evidence that paradoxical sleep deprivation (PSD) elicits penile erection (PE) and ejaculation (EJ), and that the erectile response of rats is mediated by nitric oxide, the present study sought to extend the latter finding by assessing the effects of sildenafil on the genital reflexes of male Wistar rats subjected to PSD. We also determined the influence of sildenafil on hormone concentrations. In the first experiment, sildenafil at doses ranging from 0.08 to 0.32 mg/kg was administered intraperitoneally to rats that had been deprived of sleep for 4 days and to home cage controls (N = 8-10/group). The frequency of PE and EJ was measured for 60 min. PSD alone induced PE in 50% of the animals; however, a single injection of sildenafil did not...

Maia,O.B.; Jácomo,A.T.A.; Bringel,B.A.; Kashivakura,C.K.; Oliveira,C.A.; Teodoro,L.O.F.; Silveira,L.; Teixeira da Costa,M.E.L.; Malta,M.C.C.; Furtado,M.M.; Torres,N.M.; Mattos,P.S.R.; Viau,.P; Lima,T.F.G.; Morato,R.G..
Serum hormone levels were compared between captive and free-living maned wolves and seasonal variations of sex hormones were studied. Blood samples were collected from 16 male and 26 female adult animals from Brazilian zoos, and from 30 male and 24 female free-living adults to determine serum progesterone and testosterone by radioimmunoassay. Serum testosterone concentrations varied (P < 0.05) across seasons for 16 captive males, being higher in autumn (2184.7 ± 355.1 pg/mL) than in summer (1080.7 ± 205.4 pg/mL), winter (1270.1 ± 276.6 pg/mL) and spring (963.9 ± 248.1 pg/mL), although they did not differ between summer, winter and spring. Testosterone concentration of 30 free-living males differed (P < 0.05) between autumn (824.1 ± 512.2 pg/mL),...

Regulation of antioxidant enzyme activities in male and female rat macrophages by sex steroids BJMBR
Azevedo,R.B.; Lacava,Z.G.M.; Miyasaka,C.K.; Chaves,S.B.; Curi,R..
Human and animal immune functions present sex dimorphism that seems to be mainly regulated by sex hormones. In the present study, the activities of the antioxidant enzymes total superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GSH-Px) were measured in intraperitoneal resident macrophages from adult male and female rats. In addition to comparing males and females, we also examined the regulation of these enzyme activities in macrophages by sex steroids. GSH-Px activity did not differ between male and female macrophages. However, both total SOD and CAT activities were markedly higher in females than in males (83 and 180%). Removal of the gonads in both males and females (comparison between castrated groups) increased the difference in...

Roles of estrogen and progesterone in modulating renal nerve function in the rat kidney BJMBR
Graceli,J.B.; Cicilini,M.A.; Bissoli,N.S.; Abreu,G.R.; Moysés,M.R..
The maintenance of extracellular Na+ and Cl- concentrations in mammals depends, at least in part, on renal function. It has been shown that neural and endocrine mechanisms regulate extracellular fluid volume and transport of electrolytes along nephrons. Studies of sex hormones and renal nerves suggested that sex hormones modulate renal function, although this relationship is not well understood in the kidney. To better understand the role of these hormones on the effects that renal nerves have on Na+ and Cl- reabsorption, we studied the effects of renal denervation and oophorectomy in female rats. Oophorectomized (OVX) rats received 17β-estradiol benzoate (OVE, 2.0 mg·kg-1·day-1, sc) and progesterone (OVP, 1.7 mg·kg-1·day-1,sc). We assessed Na+ and...

Effects of metformin treatment on luteal phase progesterone concentration in polycystic ovary syndrome BJMBR
Meenakumari,K.J.; Agarwal,S.; Krishna,A.; Pandey,L.K..
The causes of luteal phase progesterone deficiency in polycystic ovary syndrome (PCOS) are not known. To determine the possible involvement of hyperinsulinemia in luteal phase progesterone deficiency in women with PCOS, we examined the relationship between progesterone, luteinizing hormone (LH) and insulin during the luteal phase and studied the effect of metformin on luteal progesterone levels in PCOS. Patients with PCOS (19 women aged 18-35 years) were treated with metformin (500 mg three times daily) for 4 weeks prior to the test cycle and throughout the study period, and submitted to ovulation induction with clomiphene citrate. Blood samples were collected from control (N = 5, same age range as PCOS women) and PCOS women during the late follicular (one...

Estrous cycle and stress: influence of progesterone on the female brain BJMBR
Lovick,T.A..
The female brain operates in a constantly changing chemical milieu caused by cyclical changes in gonadal hormones during the estrous cycle (menstrual cycle in women). Such hormones are highly lipophilic and pass readily from the plasma to the brain where they can influence neuronal function. It is becoming clear that the rapid reduction in peripheral circulating progesterone, which occurs during the late diestrous phase of the cycle, can trigger a withdrawal-like response, in which changes in GABA A receptor expression render hyper-responsive certain brain areas involved in processing responses to stressful stimuli. The periaqueductal gray matter (PAG) is recognised as an important region for integrating anxiety/defence responses. Withdrawal from...

Effect of medroxyprogesterone acetate on thyrotropin secretion in adult and old female rats BJMBR
Moreira,R.M.; Borges,P.P.; Lisboa,P.C.; Curty,F.H.; Moura,E.G.; Pazos-Moura,C.C..
Steroid hormones have been implicated in the modulation of TSH secretion; however, there are few and controversial data regarding the effect of progesterone (Pg) on TSH secretion. Medroxyprogesterone acetate (MPA) is a synthetic alpha-hydroxyprogesterone analog that has been extensively employed in therapeutics for its Pg-like actions, but that also has some glucocorticoid and androgen activity. Both hormones have been shown to interfere with TSH secretion. The objective of the present study was to investigate the effects of MPA or Pg administration to ovariectomized (OVX) rats on in vivo and in vitro TSH release and pituitary TSH content. The treatment of adult OVX rats with MPA (0.25 mg/100 g body weight, sc, daily for 9 days) induced a significant...

Behavioral effects of endogenous or exogenous estradiol and progesterone on cocaine sensitization in female rats BJMBR
Souza,M.F.; Couto-Pereira,N.S.; Freese,L.; Costa,P.A.; Caletti,G.; Bisognin,K.M.; Nin,M.S.; Gomez,R.; Barros,H.M.T..
Cocaine sensitization is a marker for some facets of addiction, is greater in female rats, and may be influenced by their sex hormones. We compared the modulatory effects of endogenous or exogenous estradiol and progesterone on cocaine-induced behavioral sensitization in 106 female rats. Ovariectomized female rats received progesterone (0.5 mg/mL), estradiol (0.05 mg/mL), progesterone plus estradiol, or the oil vehicle. Sham-operated control females received oil. Control and acute subgroups received injections of saline, while the repeated group received cocaine (15 mg/kg, ip) for 8 days. After 10 days, the acute and repeated groups received a challenge dose of cocaine, after which locomotion and stereotypy were monitored. The estrous cycle phase was...

Silva,Lorena Ferreira; Silva,Luiz Antonio Franco da; Rabelo,Rogério Elias; Silva,Wanessa Patrícia Rodrigues da; Cassali,Geovanni Dantas; Gonçalves,Aline De Biasi Bassani; Sant’Ana,Fabiano José Ferreira de.
ABSTRACT: This manuscript described the anatomopathological and immunohistochemical findings in a rare case of mammary fibroadenomatoid hyperplasia in a 12-month-old Holstein heifer. A yellow, multilobulated, firm 20cm x 9cm x 6.5cm mass affecting the right quarters of the udder was observed. Total mastectomy was performed. Microscopic evaluation revealed severe hyperplasia of the mammary epithelium and numerous well-differentiated and mildly pleomorphic acini. Additionally, moderate proliferation of the fibrous connective tissue and the myoepithelial cells near the proliferating acini was evident. About 50% of the proliferating epithelial cells showed positive nuclear labeling for estrogen and progesterone receptors, and approximately one-third were...
