Mortalities of oyster seed of Crassostrea gigas associated with ostreid herpes virus OsHV-1 μVar have been observed in many oyster producing countries since 2008. The objective of this study was to further investigate whether low temperature treatments can offer a viable option to mitigate oyster mortalities. An experiment was set-up to further evaluate the effect of low temperature treatments (10 and 13°C vs. 21°C) and their duration (6 d to 83 d) on the survival of oysters previously infected with OsHV-1 μVar by means of exposure to field conditions in areas where mortalities were occurring. Average survival of oysters infected with OsHV-1μVar was 71% after 83 d at low temperatures compared to only 23% in controls maintained at 21°C. During... |

Climate change contributes to local weather modifications such as storm events and heavy rainfall, and in particular, changes in the frequency and intensity of extreme storm and rainfall events. Observations have indicated that rainfall-runoff processes, like sewage overflow may induce fecal contamination in coastal water. Pathogens present in runoff water could therefore contaminate shellfish and be responsible for food-borne disease outbreaks. Mathematical models for this were used to predict the effect of climate modifications on shellfish quality. Using a hydrodynamic model, simulations of water quality conditions under a variety of storm event scenarios were run and are presented here for a harvesting area in France. Major storm events occurring in... |

An inter-laboratory study of the accuracy of microbial source tracking (MST) methods was conducted using challenge fecal and sewage samples that were spiked into artificial freshwater and provided as unknowns (blind test samples) to the laboratories. The results of the Source Identification Protocol Project (SIPP) are presented in a series of papers that cover 41 MST methods. This contribution details the results of the virus and bacteriophage methods targeting human fecal or sewage contamination. Human viruses used as source identifiers included adenoviruses (HAdV), enteroviruses (EV), norovirus Groups I and II (NoVI and NoVII), and polyomaviruses (HPyVs). Bacteriophages were also employed, including somatic coliphages and F-specific RNA bacteriophages... |

Since 1991, sporadic, but high mortality in larvae of the Pacific oyster (Crassostrea gigas) has been observed during the summer in various French hatcheries. This mortality was concomitant with the detection of a virus by transmission electron microscopy. This virus, by virtue of its morphological characteristics and developmental cycle, seems to belong to the Herpesviridae family. The pathogenicity of this viral agent has been demonstrated in the laboratory by experimentally induced mortality on healthy larvae. Moreover, concomitant mortality has been reported in larvae of Crassostrea gigas and Ostrea edulis reared in the same tanks, with detection of a herpes-type virus. |

Over the last decade, RNA interference pathways have emerged in eukaryotes as critical regulators of many diverse biological functions including, among others, transcriptional gene regulation, post-transcriptional gene silencing, heterochromatin remodelling, suppression of transposon activity, and antiviral defences. Although this gene silencing process has been reported to be relatively well conserved in species of different phyla, there are important discrepancies between plants, invertebrates and mammals. In penaeid shrimp, the existence of an intact and functional RNAi machinery is supported by a rapidly growing body of evidence. However, the extent to which this process participates to the host immune responses remains poorly defined in this non-model... |

The single-stranded genomic RNA of Taura syndrome virus (TSV) is 10205 nucleotides in length, excluding the 3' poly(A) tail, and contains two large open reading frames (ORFs) that are separated by an intergenic region of 207 nucleotides. The ORFs are flanked by a 377 nucleotide 5' untranslated region (UTR) and a 226 nucleotide 3' UTR followed by a poly(A) tail. The predicted amino acid sequence of ORF1 revealed sequence motifs characteristic of a helicase, a protease and an RNA-dependent RNA polymerase, similar to the non-structural proteins of several plant and animal RNA viruses. In addition, a short amino acid sequence located in the N-terminal region of ORF1 presented a significant similarity with a baculovirus IAP repeat (BIR) domain of inhibitor of... |

Bivalve molluscs are filter feeders and as a consequence they may bioaccumulate in their tissues viruses that infect humans and higher vertebrates. However, there have also been described mortalities of bivalve molluscs associated with viruses belonging to different families. Mass mortalities of adult Portuguese oysters, Crassostrea angulata, among French livestocks (between 1967 and 1973) were associated with irido-like virus infections. Herpesviruses were reported in the eastern oyster, Pacific oyster and European flat oyster and lately in scallops and clams. Disseminated neoplasia, a proliferative cell disorder of the circulatory system of bivalves, although of uncertain aetiology, has been suggested to be caused by retroviral infections. Other viruses... |

Pacific oysters, Crassostrea gigas, are one of the most productive aquaculture species in the world. However, they are threatened by the spread of Ostreid herpesvirus-1 (OsHV-1) and its microvariants (collectively “µvars”), which cause mass mortalities in all life stages of Pacific oysters globally. Breeding programs have been successful in reducing mortality due to OsHV-1 variants following viral outbreaks; however, an OsHV-1-resistant oyster line does not yet exist in the United States (US), and it is unknown how OsHV-1 µvars will affect US oyster populations compared to the current variant, which is similar to the OsHV-1 reference, found in Tomales Bay, CA. The goals of this study were to investigate the resistance of C. gigas juveniles produced by the... |

In a recent report by risk assessment experts on the identification of food safety priorities using the Delphi technique, foodborne viruses were recognized among the top rated food safety priorities and have become a greater concern to the food industry over the past few years. Food safety experts agreed that control measures for viruses throughout the food chain are required. However, much still needs to be understood with regard to the effectiveness of these controls and how to properly validate their performance, whether it is personal hygiene of food handlers or the effects of processing of at risk foods or the interpretation and action required on positive virus test result. This manuscript provides a description of foodborne viruses and their... |
