Created in 1992, the REPAMO network (Network of mollusc pathology), is a national zoosanitary surveillance network of shellfish health status along the French coasts. Ils activities are in keeping with two European Directives, 91167/EEC and 95170/EC and are a part of the institutional tasks of Ifremer. The aims of the network are to prevent the introduction and spread of exotic pathogens and to sm'vey the evolution of notifiable pathogens already present in France. The network focused on the survey of listed diseases (bonamiosis and marteiliosis of flat oyster) in two areas under agreement process (Granville bed and zone X). In 2005, only samples of the Granville bed could be analysed due to the difflculty to obtain flat oysters (sparse oyster beds, beds... |

Bonamia ostreae is an intracellular protozoan which is recognized as a cause of mortality in European populations of flat oysters (Ostrea edulis). Based on the recent characterization of actin genes of B. ostreae, specific primers were designed for real-time PCR using SYBR® Green chemistry. Specificity was demonstrated by the unique melting temperature peak observed in positive samples and by the lack of amplification in samples of oysters infected by closely related parasites, including Bonamia exitiosa. A calibration curve using a cloned template was defined to estimate copy number. The assay had a 6 log- dynamic range, mean inter- and intra-assay variation coefficients of <1% and a minimum detection limit of 50 gene copies per reaction. Using... |

The REPAMO network (network ofmollusc pathology), created in 1986, ensures the survey of shellfish health status along French coasts according? to European Directives 91/67/EEC and 95/70/EEC. The aims of the network are 1 - to prevent the introduction and spread of exotic pathogens, 2-to study the means to decrease the impact of pathogens already present and to survey their evolution. In 2003, the REPAMO network focused on the survey oflisted diseases (bonamiosis and marteiliosis) in two areas under agreement process (Banc de Granville and zone X) and on the study of abnormal morlalities. Number of reported abnormal morlalities has increased in 2003. This rise can be partly explained by the recent network restructuring, but also by the summer heatwave and... |

Viral gametocytic hypertrophy was reported for the first time in 2001 in Pacific oyster Crassostrea gigas in France. Since this date, the number of reported cases and the distribution area have increased every year; however, the cases are not associated with macroscopic signs or increased mortality rates. Both male and female gametes were hypertrophied and basophilic inclusions were observed in gamete nuclei, Transmission electron microscopy revealed the presence of viral particles in these intranuclear basophilic inclusions. These particles had characteristics similar to those of the Papillomaviridae and Polyomaviridae families: they were small, non-enveloped, icosahedral, and 44 to 56 nm in diameter, The viral particles were found in male, female and... |

The French network for the surveillance and monitoring of mollusc health (REPAMO) was officially set up in 1992 to control the shellfish status along the French coasts according to the European directives 91/67/EEC and 95/70/EC. The objectives are 1) the surveillance of notifiable diseases present in France (Bonamiosis and Marteiliosis of flat oysters Ostrea edulis), 2) the surveillance of health status of cultured and wild populations of molluscs, 3) the study of abnormal mortality and 4) the control of exchanges from European and third countries. In this context, a new surveillance programme, called Active Targeted Survey, part of the REPAMO, was applied in 2004 in order to improve the monitoring of the health status of the French mollusc populations... |

Bonamia ostreae is an intracellular protistan parasite affecting flat oysters Ostrea edulis. It can be detected in juveniles but mortalities mainly affect oysters which are more than 2 years old. The parasite is usually observed inside haemocytes and sometimes free, notably in gill epithelia suggesting a parasite release through this organ. However, the infective form and ways of entry and release remain undetermined. Flat oysters incubate their larvae in their pallial cavity for 8–10 days before releasing them into the water column. Flat oysters in Bay of Quiberon in South Brittany (France) are known to be infected with B. ostreae since 1979 and is the most important area in France for O. edulis spat collection. Flat oysters incubating larvae were sampled... |

The European flat oyster Ostrea edulis has been considered as the traditional French oyster for a very long time and has now nearly disappeared from our farms, as a consequence of the two successive epizootic of the Marteilia refringens parasite and Bonamia ostreae. The actual quasi monoculture of Crassostrea gigas has made necessary the search for a diversification in oyster culture, including the search for a new start of the indigenous flat oyster culture. The Ifremer has been conducting parallel works since 1985 in order to better understand the biology of the parasite as well as to try and select different lines of flat oyster resistant to Bonamia ostreae. |

Bonamiosis was first reported in association with mass mortality of flat oysters, Ostrea edulis, in June 1979 in L'Ile Tudy, Brittany. The disease rapidly spread to all of the oyster farming areas in France but also in other European countries. The French flat oyster production, which already suffered from another protozoan disease, marteiliosis, decreased from 5500 mt in 1979 to less than 2000 mt after 1980. In 2001, about 350 French farms sell 1650 mt of flat oysters. This production mainly relies on natural spat collection which specially occurs in Quiberon bay. One third of this spat is transferred from South to North Brittany for further growth. Quiberon bay constitutes an interesting site regarding the surveillance of bonamiosis because of the... |

The flat oyster Ostrea edulis L. is widespread along the Italian coasts. In particular, the Manfredonia Gulf (Adriatic Sea) represents an important site where natural beds subsist. Previous monitoring conducted in 1990 by light microscopy and ultrastructural studies revealed the presence of Bonamia-like microcell parasites in some flat oysters: following this observation, a new sampling of O. edulis was carried out at this location in 2007. Of 750 oysters collected, 3 showed the presence of uninucleated microcells (2 to 3 mu m diameter) free or inside the haemocyte cytoplasm by cytology and histopathology. Molecular analysis confirmed that the microcells in 2 oysters were B. exitiosa, whereas in the third oyster the microcells were B. ostreae. Moreover,... |
