Use of vinasse and sugarcane bagasse for the production of enzymes by lignocellulolytic fungi BABT

In this present work, three strains of Pleurotus and Trichoderma reesei were cultivated in media with pre-treated bagasse and vinasse. Cellulolytic and lignolytic activities and biomass production were analyzed. The treatment of the bagasse with 2% H2O2 + 1.5% NaOH + autoclave resulted in a greater fiber breakage increasing the cellulose level up to 1.2 times and decreasing 8.5 times the hemicellulose content. This treatment also resulted in a high lignolytic activity for all cultures utilized. T. reesei produced laccase, peroxidase and manganese-peroxidase in all the treatments, having its manganese-peroxidase activity raging from 1.9 to 4.8 times higher than the basidiomycetes.

Production and characterization of galactose oxidase produced by four isolates of Fusarium graminearum BABT

A screening aimed to find new galactose oxidase producer isolates and to evaluate the production among Fusarium graminearum strains was conducted. Thirty-five isolates out of 39 analysed produced the enzyme at several levels. The data indicated a wide distribution of galactose oxidase within F. graminearum and also revealed new producer isolates. The enzyme produced by different isolates showed similar thermal activity and stability and were active on same substrates. However, the optimum pH ranged from 7.0 to 7.5. Thus, all evaluated isolates were suitable for the production of galactose oxidase.

Nucleotide excision repair pathway assessment in DNA exposed to low-intensity red and infrared lasers BJMBR

Low-intensity lasers are used for prevention and management of oral mucositis induced by anticancer therapy, but the effectiveness of treatment depends on the genetic characteristics of affected cells. This study evaluated the survival and induction of filamentation of Escherichia coli cells deficient in the nucleotide excision repair pathway, and the action of T4endonuclease V on plasmid DNA exposed to low-intensity red and near-infrared laser light. Carbon sources effect on pectinase production from Aspergillus japonicus 586 BJM

The effect of different carbon sources on the pectinesterases, endo- and exo-polygalacturonase activities from Aspergillus japonicus 586 was evaluated in liquid media (Manachini solutions) supplemented with different substrate concentrations. The culture medium was inoculated with 5.10(6) spores/ml and mantained under agitation (140 rpm), at 30°C, during 122 h. The enzyme evaluation was carried out 24 h after filtration. The crude extract from A. japonicus 586 indicated that the best enzymatic activities were afforded in the presence of 0.5% pectin (pectinesterease), 0.2% pectin and 0.2% glycerol (endopolygalacturonase), and 0.5% pectin associated to 0.5% glucose (exopolygalacturonase). Laccase production by Lepista sordida BJM

A Lepista sordida laccase has been characterized. Laccase and manganese peroxidase were detected in liquid medium with ammonium phosphate, yeast extract and ammonium molybdidate as nitrogen sources after 3 days of cultivation. Laccase optimal temperature and pH were 45ºC and 3.5, respectively.

Highly thermostable and alkaline α-amylase from a halotolerant-alkaliphilic Bacillus sp. AB68 BJM

An alkaliphilic and highly thermostable α-amylase producing Bacillus sp. was isolated from Van soda lake. Enzyme synthesis occurred at temperatures between 25ºC and 40ºC. Analysis of the enzyme by SDS-PAGE revealed a single band which was estimated to be 66 kDa. The enzyme was active in a broad temperature range, between 20ºC and 90ºC, with an optimum at 50ºC; and maximum activity was at pH 10.5. The enzyme was almost completely stable up to 80ºC with a remaining activity over 90% after 30 min pre-incubation. Thermostability was not increased in the presence of Ca2+. Estimating the turnover number in enzyme kinetic reactions using transient and stationary state data BJPS

Substrate and product concentration data obtained by simulating enzyme-substrate reaction rate equations were used to test two proposed kinetic rate constant estimation techniques in this study. In the first technique, the turnover number, k3, was calculated using early transient time domain data, which are difficult to obtain experimentally. The technique used an iterative approach to calculate k3 with a pair of data and the value of k3 could be retrieved with 35% error. The second technique calculated k3 using stationary domain data and the value of k3 could be retrieved with less than 5% error. Activity of fungal phytases stored in two ways in response to the period of storage at room temperature Ciência Rural

Two distinct experiments were conducted simultaneously with phytases of Aspergillus oryzae and A. niger for determining enzyme activity in response to storage period (up to 180 days at room temperature - RT). In the first experiment, enzymes were stored as they were acquired (pure form) and the activity was measured periodically during 180 days of storage at RT. In the second experiment, the phytases were incorporated to a supplement containing vitamins, minerals, and amino acids; and then this supplement was stored at RT up to 180 days, so that every 30 days of storage was collected one aliquot from each replicate for determining enzymatic activity. The objective of this study was to evaluate the effect of reduced-crude protein (CP) and available phosphorus (aP) diets, added with phytase and amino acids for broilers in the growing (14 to 21 days old) and retirement (35 to 42 days) phases on nitrogen, potassium and phosphorus balance.
