Defense mechanisms of Solanum tuberosum L. in response to attack by plant-pathogenic bacteria Biol. Res.
POIATTI,VERA A D; DALMAS,FERNANDO R; ASTARITA,LEANDRO V.
The natural resistance of plants to disease is based not only on preformed mechanisms, but also on induced mechanisms. The defense mechanisms present in resistant plants may also be found in susceptible ones. This study attempted to analyze the metabolic alterations in plants of the potato Solanum tuberosum L. cv. Agata that were inoculated with the incompatible plant-pathogenic bacteria X. axonopodis and R. solanacearum, and the compatible bacterium E. carotovora. Levels of total phenolic compounds, including the flavonoid group, and the activities of polyphenol oxidase (PPO) and peroxidase (POX) were evaluated. Bacteria compatibility was evaluated by means of infiltration of tubers. The defense response was evaluated in the leaves of the potato plants....

Evaluation of Anticancer Activity of Extracted Flavonoids from Morus Alba Leaves and its interaction with DNA BABT
Fallah,Soudabeh; Hajihassan,Zahra; Zarkar,Nasrin; Chadegani,Azra Rabbani-; Mohammadnejad,Javad; Hajimirzamohammad,Milad.
ABSTRACT Morous Alba, known as white mulberry contains many oxidative flavonoids, widely used in the treatment of many diseases like hyperglycemia, inflammation, fever and cancer. In the present study we investigated the interaction of extracted flavonoids from Iranian Morus Alba leaves with DNA as a main target for anticancer drugs. Various spectroscopic techniques (UV/Vis, CD and Fluorescence Spectroscopy) were used to detect the interaction. In vivo studies also were done to confirm the effectiveness of the extracted flavonoids. The spectroscopic results showed that the extracted flavonoids bind to DNA especially to the sugar-phosphate backbone and making DNA conformational changes upon this binding. Experiments on the cancerous mice with solid tumors...

Teixeira,Maria Gessi; Carvalho,Marília; Leite,Marco Aurélio; Barbosa,Sandro; Santos filho,Plinio Rodrigues dos; Santos,Breno Régis.
Abstract The aim of this work was to evaluate the effect of Salicylic acid, 2,4-D and 2-iP on the production of total phenolic and flavonoids as well in the levels of fukugetin and 7-epiclusianone in callus of Garcinia brasiliensis zygotic embryos. For this, Bacupari callus were exposed to different concentrations (0.0; 0.1; 1; 10; 100 µM) of Salicylic acid (SA) in the presence or in the absence of 72 μM 2-iP and 28.73 μM 2,4-D. The highest concentration of total phenolic occurred in the treatment with 100 μM SA in callus subcultured in the absence of 2,4-D and 2-iP, and with 10 μM SA in callus subcultured in the presence of these regulators. Concerning flavonoids, 2,4-D and 2-iP supplementation without the presence of the SA was sufficient for the highest...

High-yield production of scutellaria radix flavonoids (baicalein, baicalin and wogonin) by liquid-culture of Scutellaria baicalensis root-derived cells BABT
Ohtsuki,Takashi; Himeji,Mika; Fukazawa,Harumi; Tanaka,Miho; Yamamoto,Hisako; Mimura,Akio.
Production of baicalein, baicalin and wogonin by liquid culture of Scutellaria baicalensis cells derived from the plant root was studied. The maximum production obtained were 119 mg/L of baicalein at two week, 1372 mg/L of baicalin at eight week, and 14 mg/L of wogonin at two week. In addition, the production of baicalin was drastically increased to 1000 mg/L level at 3-week culture, and the extremely high production rate (339 mg/L•week) was obtained. In the comparison of total antioxidative activities among baicalein, baicalin and wogonin, evaluated by thiocyanate method, it was suggested that the location of hydroxyl groups both at 5- and 6-position contributed to enhancement of radical scavenging activity, and/or methoxylation at 8-position diminished...

Neuroprotection by flavonoids BJMBR
Dajas,F.; Rivera-Megret,F.; Blasina,F.; Arredondo,F.; Abin-Carriquiry,J.A.; Costa,G.; Echeverry,C.; Lafon,L.; Heizen,H.; Ferreira,M.; Morquio,A..
The high morbidity, high socioeconomic costs and lack of specific treatments are key factors that define the relevance of brain pathology for human health and the importance of research on neuronal protective agents. Epidemiological studies have shown beneficial effects of flavonoids on arteriosclerosis-related pathology in general and neurodegeneration in particular. Flavonoids can protect the brain by their ability to modulate intracellular signals promoting cellular survival. Quercetin and structurally related flavonoids (myricetin, fisetin, luteolin) showed a marked cytoprotective capacity in in vitro experimental conditions in models of predominantly apoptotic death such as that induced by medium concentrations (200 µM) of H2O2 added to PC12 cells in...

The action of red wine and purple grape juice on vascular reactivity is independent of plasma lipids in hypercholesterolemic patients BJMBR
Coimbra,S.R.; Lage,S.H.; Brandizzi,L.; Yoshida,V.; da Luz,P.L..
Although red wine (RW) reduces cardiovascular risk, the mechanisms underlying the effect have not been identified. Correction of endothelial dysfunction by RW flavonoids could be one mechanism. We measured brachial artery reactivity by high-resolution ultrasonography, plasma lipids, glucose, adhesion molecules (ICAM-1 and VCAM), and platelet function in 16 hypercholesterolemic individuals (8 men and 8 women; mean age 51.6 ± 8.1 years) without other risk factors. Twenty-four normal subjects were used as controls for vascular reactivity. Subjects randomly received RW, 250 ml/day, or purple grape juice (GJ), 500 ml/day, for 14 days with an equal wash-out period. At baseline, all 16 subjects were hypercholesterolemic (mean LDL = 181.0 ± 28.7 mg/dl) but HDL,...

Wine, alcohol and atherosclerosis: clinical evidences and mechanisms BJMBR
Luz,P.L. da; Coimbra,S.R..
Atherosclerosis is a chronic inflammatory disease which may cause obstructions of the coronary, cerebral and peripheral arteries. It is typically multifactorial, most often dependent on risk factors such as hypercholesterolemia, diabetes, smoking, hypertension, sedentarism, and obesity. It is the single main cause of death in most developed countries due to myocardial infarction, angina, sudden death, and heart failure. Several epidemiological studies suggest that moderate alcohol intake, especially red wine, decrease cardiac mortality due to atherosclerosis. The alcohol effect is described by a J curve, suggesting that moderate drinkers may benefit while abstainers and heavy drinkers are at higher risk. Experimental studies indicate that most beneficial...

P-glycoprotein-mediated resistance to chemotherapy in cancer cells: using recombinant cytosolic domains to establish structure-function relationships BJMBR
Di Pietro,A.; Dayan,G.; Conseil,G.; Steinfels,E.; Krell,T.; Trompier,D.; Baubichon-Cortay,H.; Jault,J.-M..
Resistance to chemotherapy in cancer cells is mainly mediated by overexpression of P-glycoprotein (Pgp), a plasma membrane ATP-binding cassette (ABC) transporter which extrudes cytotoxic drugs at the expense of ATP hydrolysis. Pgp consists of two homologous halves each containing a transmembrane domain and a cytosolic nucleotide-binding domain (NBD) which contains two consensus Walker motifs, A and B, involved in ATP binding and hydrolysis. The protein also contains an S signature characteristic of ABC transporters. The molecular mechanism of Pgp-mediated drug transport is not known. Since the transporter has an extraordinarily broad substrate specificity, its cellular function has been described as a "hydrophobic vacuum cleaner". The limited knowledge...

Binding effect of proline-rich-proteins (PRPs) on in vitro antimicrobial activity of the flavonoids BJM
Ansari,Jawaad Ahmed; Naz,Shahina; Tarar,Omer Mukhtar; Siddiqi,Rahmanullah; Haider,Muhammad Samee; Jamil,Khalid.
The interaction of the cyanidin, pelargonidin, catechin, myrecetin and kaempferol with casein and gelatin, as proline rich proteins (PRPs), was studied. The binding constants calculated for both flavonoid-casein and flavonoid-gelatin were fairly large (105–107 M−1) indicating strong interaction. Due to higher proline content in gelatin, the binding constants of flavonoid-gelatin (2.5 × 105–6.2 × 107M−1) were found to be higher than flavonoid-casein (1.2 × 105–5.0 × 107 M−1). All the flavonoids showed significant antibacterial activity against the tested strains. Significant loss in activity was observed due to the complexation with PRPs confirming that binding effectively reduced the concentration of the free flavonoids to be available for antibacterial...

Quercetin: a flavonoid with the potential to treat asthma BJPS
Fortunato,Laila Rigolin; Alves,Claudiney de Freitas; Teixeira,Maxelle Martins; Rogerio,Alexandre Paula.
Allergic asthma is a complex inflammatory disorder characterized by airway hyperresponsiveness, eosinophilic inflammation and hypersecretion of mucus. Current therapies include β2-agonists, cysteinyl leukotriene receptor 1 antagonists and corticosteroids. Although these drugs demonstrate beneficial effects, their adverse side effects limit their long-term use. Thus, the development of new compounds with similar therapeutic activities and reduced side effects is both desirable and necessary. Natural compounds are used in some current therapies, as plant-derived metabolites can relieve disease symptoms in the same manner as allopathic medicines. Quercetin is a flavonoid that is naturally found in many fruits and vegetables and has been shown to exert...
