Bioensaios de toxicidade genética com Tradescantia. Infoteca-e

Resumo: Dentre a variedade de bioensaios disponíveis para estudos de detecção e monitoração dos efeitos tóxicos genéticos de substâncias e ambientes contaminados, os bioensaios que empregam a planta Tradescantia destacam-se pela sensibilidade e facilidade de realização de estudos de curto prazo in situ. Este trabalho apresenta uma revisão extensiva dos estudos de mutagênese ambiental empregando Tradescantia como bioindicador, e é complementado com resultados experimentais e indicações metodológicas.

DNA damage in dab (Limanda limanda) and haddock (Melanogrammus aeglefinus) from European seas ArchiMer

Dab (Limanda limanda) and haddock (Melanogrammus aeglefinus) were collected from coastal and offshore locations in the Baltic (dab only), North Sea (haddock from one location only) and Iceland. Blood was analysed for DNA strand breaks using the comet assay and liver samples for DNA adduct concentrations. DNA strand breaks were at background levels in dab from the two Iceland locations and from the Dogger Bank. The highest levels were observed in dab from the Firth of Forth, Ekofisk and the German Bight. Hepatic DNA adducts in dab were highest at Ekofisk, in the Baltic and Dogger Bank, below detection limit in dab from Iceland and low in dab from the Firth of Forth and German Bight. Single cell gel electrophoresis (SCGE or comet) assay allows quantification of DNA damage in individual cells and is an ideal tool for use within biological monitoring programmes. Comet assay can be used on a range of cell types including somatic, reproductive (gametes) or circulatory cells in many different species including both marine bivalves and flatfish. The assay can be employed with simple equipment available in most laboratories, is sensitive to environmentally relevant levels of DNA damage (Frenzilli and Lyons, 2009), accurately demonstrates a linear dose response to exposure (Collins et al., 1996), and can be adapted for use on most nucleated cell types. Use of low density polyethylene membranes for assessment of genotoxicity of PAHs in the Seine River ArchiMer

The genotoxicity of river water dissolved contaminants is usually estimated after grab sampling of river water. Water contamination can now be obtained with passive samplers that allow a time-integrated sampling of contaminants. Since it was verified that low density polyethylene membranes (LDPE) accumulate labile hydrophobic compounds, their use was proposed as a passive sampler. This study was designed to test the applicability of passive sampling for combined chemical and genotoxicity measurements. A new gravel-contact assay using rainbow trout, Oncorhynchus mykiss, embryos was developed to assess the toxicity of polycyclic aromatic hydrocarbons (PAHs) and other hydrophobic compounds. Environmentally realistic exposure conditions were mimicked with a direct exposure of eyed rainbow trout embryos incubated onto chemical-spiked gravels until hatching at 10 °C. The aims of this PhD were to determine (1) the effects of the herbicide diuron on the genome of the oyster, Crassostrea gigas, (2) the possible transmission to the offspring of damaged DNA and (3) its consequences on oyster physiology. The genotoxicity of diuron and the vertical transmission of DNA damage were highlighted after parental exposure to environmental concentrations of diuron during the gametogenesis. Primary DNA lesions were detected in both somatic cells and germ cells of exposed genitors. Development of a comet-FISH assay for the detection of DNA damage in hemocytes of Crassostrea gigas ArchiMer

In this work, the DNA-damaging effect of hydrogen peroxide on the structural integrity of nucleolar organizer regions (NORs) was studied for the first time by comet-FISH in the Pacific oyster Crassostrea gigas. Global DNA damage was assessed in hemocytes using an alkaline version of the comet assay. Next, NOR sensitivity was analysed by mapping major rDNA repeat unit by fluorescence in situ hybridisation (FISH) on the same comet slides. Comet assay in phytoplankton as biomarker of genotoxic effects of environmental pollution ArchiMer

The alkaline comet assay was tested on different microalgae: the clinofiagellates, Karenia mikimotoi and Alexandrium minuturn, and the diatom, Chaetoceros gracilis. The microalgae were exposed during their exponential growth to the model direct genotoxicant, hydrogen peroxide (1 h, 5 and 100 pM H2O2). Following H2O2 exposure, the comet assay was validated only for K. mikimotoi for which genotoxicity was observed from the lowest tested concentration of 5 pM with a concentration-dependent effect. The aim of this Ph‟D was to study the biotransformation and the genotoxicity of two PAH of interest, namely the benzo[a]pyrene (BaP) and the fluoranthene (Fluo), in the common sole Solea solea, which is a benthic species sensitive to chemical pollution. The use of in vitro exposure models and the application of two different chemical analytical techniques allowed demonstrating that, as in higher vertebrates, the dihydrodiols and the phenols of BaP and Fluo are the major metabolites produced by sole liver enzymes. Cytological and cytogenic effects induced by thiabendazole on Allium cepa root meristem JBAG

The toxicity and genotoxicity of thiabendazole (TBZ) in its commercial formulation Foldan® was studied in Allium cepa meristematic cells at concentrations ranging between 10 and 250 µg / ml. Our results show that exposure to TBZ induces a significant increase in the frequency of anaphase telophase chromosomal aberrations, micronuclei and binucleated cells at all the concentrations of TBZ assayed. The frequency of anaphase telophase aberrations was observed to reach a plateau at the concentration of 50 µg / ml of TBZ, which could be probably due to TBZ mitotic toxicity manifested at concentrations of 100 and 250 µg / ml.
