Polymorphism in Litopenaeus vannamei genes and cross-species amplification in other shrimp species PAB
Santos,Camilla Alves; Farias,Flavio; Teixeira,Ana Karina; Rocha,João Luís; Guerrelhas,Ana Carolina; Freitas,Patrícia Domingues de.
Abstract: The objective of this work was to assess polymorphisms within Litopenaeus vannamei candidate genes related to growth performance, and a possible association between weight genotypes and phenotypes in two shrimp closed lines. Three EST-SSR (serine-arginine, 60S ribosomal, and troponin) and one EST-SNP (crustacyanin) were selected and evaluated in L. vannamei and seven other marine shrimp species, besides two freshwater species. Genotypes had no effect on total body weight. Although no association was observed in the sampled population, these markers showed amplification for most of the evaluated species and, therefore, may be useful in further genetic studies on shrimp species.

Mapping of quantitative trait loci for thermosensitive genic male sterility in indica rice PAB
Alcochete,Antonio Alberto Neves de; Rangel,Paulo Hideo Nakano; Ferreira,Marcio Elias.
The objective of this work was to select and use microsatellite markers, to map genomic regions associated with the genetic control of thermosensitive genic male sterility (TGMS) in rice. An F2 population, derived from the cross between fertile and TGMS indica lines, was used to construct a microsatellite-based genetic map of rice. The TGMS phenotype showed a continuous variation in the segregant population. A low level of segregation distortion was detected in the F2 (14.65%), whose cause was found to be zygotic selection. There was no evidence suggesting a cause-effect relationship between zygotic selection and the control of TGMS in this cross. A linkage map comprising 1,213.3 cM was constructed based on the segregation data of the F2 population....

Association between microsatellites and resistance to Mal de Río Cuarto in maize by discriminant analysis Phyton
Bonamico,NC; Balzarini,MG; Arroyo,AT; Ibañez,MA; Díaz,DG; Salerno,JC; Di Renzo,MA.
Resistance to Mal de Río Cuarto (MRC) disease in maize (Zea mays L.) is important in Argentina because the crop area involves a wide region where the disease is endemic. Molecular marker-assisted selection could be used as an additional selection tool to enhance precision of the genotype selection for resistance. It demands the identification of informative markers. Microsatellite (SSR) markers linked to gene(s) associated with resistance to MRC have been reported from previous QTL analyses. These analyses have been made on linkage maps derived from a relatively early mapping population. In advanced populations, where highly distinct genotypes are easily classified, discriminant analysis (DA) represents a complementary strategy to marker identification;...

Gaino,Ana Paula Silva Campos; Moraes,Mario Luiz Teixeira de; Moreira,Juliana Prado; Cardin,Laila Toniol; Moraes,Marcela Aparecida; Silva,Alexandre Marques da; Freitas,Miguel Luiz Menezes; Sebbenn,Alexandre Magno.
The aims of this study were to investigate the mating system of a fragmented population of the dioecious tropical tree Myracrodruon urundeuva Allemão, using five microsatellite loci and the mixed mating and correlated mating models. The study was conducted in the Estação Ecológica de Paulo de Farias (436 ha), where the population occupies about 142 ha. The mating system was estimated using 514 open-pollinated offspring, collected from 30 seed-trees. Estimates of the multilocus outcrossing rate confirm that the species is dioecious (t m = 1.0). Low levels of mating among relatives were detected in the population (1 - t s = 0.020). The estimate of paternity correlation (r p(m)) indicated that offsprings were composed of mixtures of half-sibs and full-sibs,...

Characterization of Hilly Chickens in Consideration of Climate Change Factors: Light and Heat Rev. Bras. Ciênc. Avic.
Khan,MKI; Ali,M; Akter,MA; Zaman,S; Miah,G.
ABSTRACT Hilly chickens were characterized from January 2015 to September 2016 considering climate factors (light and heat). The experimental birds were divided into three groups (heat stress; light and control). The heat was generated in the chicken’s shelter by a black shaded light (Lantern) for two to three hours more after sunset. A lighting device (Lantern) was kept in the chicken’s shelter for the same period for increasing daylight duration. The individual chicken’s egg production, egg weight and mature live weight were studied from onset of egg production. It was observed that reddish brown hilly type chicken produced more (100.80 no/year/chicken) and larger sized (43.21g) eggs than spotted chicken (83.4 no/year/chicken and 40.46g). Among the three...

Genetic diversity of four cattle breeds using microsatellite markers R. Bras. Zootec.
Machado,Marco Antonio; Schuster,Ivan; Martinez,Mário Luiz; Campos,Ana Lúcia.
Four cattle breeds (Gyr, Nellore, Guzerat and Holstein) were analyzed by amplification of genomic DNA using microsatellite loci to evaluate the genetic diversity within and among them. DNA samples of 18 animals from each breed were collected to access the genetic content of them. Allele frequencies were calculated and used to generate a Nei's genetic distance matrix what was used to build a dendrogram following UPGMA clustering. As expected, Holstein breed was the most distinct from the other breeds: 1.15 in relation to Gyr, 1.12 in relation to Nellore and 0.94 in relation to Guzerat. The closest genetic distance was 0.25 between Guzerat and Nellore. A total of 64 alleles in all four breeds were detected using nine microsatellite primers. Each breed showed...

Assessment of inter- and intra-cultivar variations in olive using SSR markers Scientia Agricola
Ipek,Ahmet; Barut,Erdogan; Gulen,Hatice; Ipek,Meryem.
Olive (Olea europaea L.) production in the world has been made by using many cultivars, and the genetic uniformity of commercial cultivars is important for standard olive oil and table olive production. The genetic variation among and within commonly cultivated olive cultivars in Turkey was analyzed using SSR markers. A total of 135 leaf samples were collected from 11 commonly cultivated olive cultivars from 11 provinces in four geographical regions of Turkey. Seven SSR primer pairs generated 46 SSR markers, and the number of SSR markers per primer pair ranged from 4 (UDO-14) to 9 (GAPU-89) with an average of 6.57. This high level of SSR polymorphism suggests that olive production in Turkey has been made using genetically diverse olive cultivars and this...

Abdelhamid,Sofiane; Grati-kamoun,Naziha; Marra,Francesco; Caruso,Tiziano.
Olive (Olea europaea L. subsp. europaea var. europaea) is one of the oldest fruit tree in the Mediterranean basin, and is cultivated for oil and canned fruit. Part of this interest is driven by the economic importance of olive oil which is increasing throughout the world due to its beneficial effect to human health. In Tunisia, olive has great socio-economic importance, with more than 60 millions olive trees cultivated for olive oil production including a wide range of cultivars which are widely extended from the north to the south regions of the country for its high economic value. Here, we applied microsatellites (SSRs) molecular markers to assess the genetic variability of the most important Tunisian olive cultivars. In total, the 10 simple sequence...

Species boundary and extensive hybridization and introgression in Petunia Acta Botanica
Turchetto,Caroline; Schnitzler,Carolina K.; Freitas,Loreta B..
ABSTRACT Studying the role of hybridization in the speciation of plants is one of the most thrilling areas of evolutionary biology. Hybridization in natural populations can act in opposition to divergence, contribute to adaptation through introgression or foster the emergence of new lineages via hybrid speciation. Species of the plant genus Petunia grow in open areas in southern South America. Some natural interspecific hybrid events have been described for the genus, such as between the endemic P. exserta and the widespread P. axillaris. Both species occur in sympatry in Serra do Sudeste (Brazil), where they occur in diverse habitats and exhibit floral divergence, which has been related to the attraction of different primary pollinators. The present study...

The bush dog (Speothos venaticus) is a South American canid, included in the IBAMA (Brazilian Institute of Environment and Renewable Natural Resources) official list of animals threatened with extinction, in the vulnerable category. As a preservation and conservation strategy, specimens kept in captivity by Brazilian Institutions are monitored by a management plan. In order to characterize and analyze the genetic variability of bush dog specimens, a cytogenetic analysis was carried out, and microsatellite data were also obtained through the use of 15 primers, originally developed for the domestic dog (Canis familiaris). All tested primers showed transferability and amplified fragment sizes similar to those described for the canine genome. From the total...

ABSTRACT We analysed 114 DNA samples collected in different locations within the stock G breeding grounds, including Ecuador and the northern and southern Pacific of Colombia. We genotyped 15 microsatellite loci, sexed all samples, and performed genotype comparisons. Genotype comparisons were done using a DNA register of previously genotyped individuals from the Colombian breeding ground and the Antarctic Peninsula feeding Area. We confirmed connectivity between feeding grounds in Antarctic Peninsula and breeding grounds in Colombia and Ecuador using microsatellite loci. We found recaptures within each sampling location in the same year, but we also found a few recaptures in the same locations between years both in breeding and feeding grounds....

Utilization of microsatellites for the analysis of genomic alterations in colorectal cancers in Brazil BJMBR
Fuzikawa,A.K.; Haddad,L.A.; da-Cunha-Melo,J.R.; Brasileiro-Filho,G.; Pena,S.D.J..
Two different pathogenetic mechanisms are proposed for colorectal cancers. One, the so-called "classic pathway", is the most common and depends on multiple additive mutational events (germline and/or somatic) in tumor suppressor genes and oncogenes, frequently involving chromosomal deletions in key genomic regions. Methodologically this pathway is recognizable by the phenomenon of loss of heterozygosity. On the other hand, the "mutator pathway" depends on early mutational loss of the mismatch repair system (germline and/or somatic) leading to accelerated accumulation of gene mutations in critical target genes and progression to malignancy. Methodologically this second pathway is recognizable by the phenomenon of microsatellite instability. The distinction...

Bioethanol strains of Saccharomyces cerevisiae characterised by microsatellite and stress resistance BJM
Reis,Vanda Renata; Antonangelo,Ana Teresa Burlamaqui Faraco; Bassi,Ana Paula Guarnieri; Colombi,Débora; Ceccato-Antonini,Sandra Regina.
Abstract Strains of Saccharomyces cerevisiae may display characteristics that are typical of rough-type colonies, made up of cells clustered in pseudohyphal structures and comprised of daughter buds that do not separate from the mother cell post-mitosis. These strains are known to occur frequently in fermentation tanks with significant lower ethanol yield when compared to fermentations carried out by smooth strains of S. cerevisiae that are composed of dispersed cells. In an attempt to delineate genetic and phenotypic differences underlying the two phenotypes, this study analysed 10 microsatellite loci of 22 S. cerevisiae strains as well as stress resistance towards high concentrations of ethanol and glucose, low pH and cell sedimentation rates. Genetic variability within Fusarium solani specie as revealed by PCR-fingerprinting based on pcr markers BJM
Brasileiro,Bereneuza Tavares Ramos Valente; Coimbra,Maria Raquel Moura; Morais Jr,Marcos Antonio de; Oliveira,Neiva Tinti de.
Fusarium solani fungus (teleomorph Haematonectria haematococca) is of relevance for agriculture, producing a disease that causes significant losses for many cultivars. Moreover, F. solani is an opportunistic pathogen to animals and humans. The complexity associated to its correct identification by traditional methods justifies the efforts of using molecular markers for isolates characterization. In this work, three PCR-based methods (one PCR-ribotyping and two PCR-fingerprinting) were used to investigate the molecular variability of eighteen F. solani isolates from four Brazilian States, collected from different substrates. Genetic analysis revealed the intraspecific variability within the F. solani isolates, without any correlation to their geographical...

Genetic Structure and Diversity of Four Chilean Sheep Breeds Chilean J. Agric. Res.
de la Barra,Rodrigo; Uribe,Héctor; Latorre,Etel; San Primitivo,Fermín; Arranz,Juan.
Chilean sheep breeds have a significant founder effect since they descend from very few parents, in some cases population inbreeding has increased and in others there have been significant differentiation processes as compared to their original population. The objective of this study was to estimate the current status of genetic diversity in sheep herds, which comprise the main sheep breeds in Chile, using molecular methods. Animals from four breeding herds were sampled and analyzed. The breeds studied were Corriedale, Suffolk Down, Romney Marsh and Chilota, these are the predominant numerical pure breeds in Chile. From each breed population 40 animals were sampled. Genetic characterization was done using nine microsatellite molecular markers (SSR)...

Analysis of genetic diversity in Argentinian heterotic maize populations using molecular markers Ciencia e Investigación Agraria
Morales,Marcelo; Decker,Viviana; Ornella,Leonardo.
Over the past three decades, traditional Argentinean Orange Flint maize cultivars have been replaced by the higher yielding U.S. Yellow Dent germplasms. However, fint cultivars are potentially resistant to biotic and/or abiotic stress. Thus, knowledge of genetic diversity and relationships among fint inbred lines would help reduce genetic vulnerability and broaden the genetic base of crops in national improvement programs. In this study, we report the analysis of 25 inbred Orange Flint germplasms and one dent using 21 microsatellite markers or Simple Sequence Repeats (SSR). The aim was to assess genetic diversity among these accessions and evaluate the usefulness of SSR markers for defning heterotic groups in temperate germplasm. Genetic diversity values...
