Molecular variants in populations of Bryconamericus aff. iheringii (Characiformes, Characidae) in the upper Paraná river basin - doi: 10.4025/actascibiolsci.v35i2.11451 Biological Sciences
Mazeti, Camila Montoro; Universidade Estadual de Maringá; Maniglia, Thiago Cintra; Universidade Estadual de Maringá; Prioli, Sônia Maria Alves Pinto; Universidade Estadual de Maringá; Prioli, Alberto José; Universidade Estadual de Maringá.
There are evidences that Bryconamericus aff. iheringii represents a species complex. DNA molecular markers have been effective in studies on phylogeny, taxonomy, and identification of cryptic species. In this study, partial sequences of genes of ATPase 6 and 8 were used to assess genetic diversity within and among populations of B. aff. iheringii of sub-basins of Tibagi, Pirapó and Ivaí rivers, belonging to the Upper Paraná river basin. The analysis of the sequences of genes pointed out high genetic diversity in B. aff. iheringii from the sub-basins studied with genetic distance values comparable to those found among different species. There was a division of the individuals into five groups. The comparison with other species of Bryconamericus that have...

Mitochondrial DNA damage associated with lipid peroxidation of the mitochondrial membrane induced by Fe2+-citrate Anais da ABC (AABC)
Almeida,Andréa M.; Bertoncini,Clélia R.A.; Borecký,Jiri; Souza-Pinto,Nadja C.; Vercesi,Aníbal E..
Iron imbalance/accumulation has been implicated in oxidative injury associated with many degenerative diseases such as hereditary hemochromatosis, beta-thalassemia, and Friedreich's ataxia. Mitochondria are particularly sensitive to iron-induced oxidative stress - high loads of iron cause extensive lipid peroxidation and membrane permeabilization in isolated mitochondria. Here we detected and characterized mitochondrial DNA damage in isolated rat liver mitochondria exposed to a Fe2+-citrate complex, a small molecular weight complex. Intense DNA fragmentation was induced after the incubation of mitochondria with the iron complex. The detection of 3' phosphoglycolate ends at the mtDNA strand breaks by a 32P-postlabeling assay, suggested the involvement of...

Wide-range genetic connectivity of Coney, Cephalopholis fulva (Epinephelidae), through oceanic islands and continental Brazilian coast Anais da ABC (AABC)
Souza,Allyson S. de; Dias Júnior,Eurico A.; Galetti Jr,Pedro M.; Machado,Erik G.; Pichorim,Mauro; Molina,Wagner F..
The Epinephelidae form a group of species of high biological and economical interests. It´s phylogeographic patterns are not well known especially the distributed populations in the western region of the Atlantic Ocean. Among the representatives is a small species called Cephalopholis fulva, Coney, which presents a wide geographical distribution, polychromia, hermaphroditism and is quickly becoming a large target for the exploration of commercial fishing. The genetic and historical demography were obtained through the partial sequence analysis of Control Region from six locations on the coastline of Brazil from the northeast coast to the southwest coast, including the oceanic islands of Rocas Atoll and Fernando de Noronha Archipelago. The analyzed samples...

Development and characterization of two new cell lines from common carp, Cyprinus carpio (Linn) Biol. Res.
Lakra,Wazir S; Goswami,M; Rajaswaminathan,T; Rathore,Gaurav.
Two new cell lines (CCF and CCH) were established from fin and heart tissues of common carp, Cyprinus carpio. The cells were optimally maintained in Leibovitz-15 medium supplemented with 10% fetal bovine serum (FBS) and 10 ng/ml of basic fibroblastic growth factor (bFGF). The effects of temperature, concentration of FBS and bFGF on the growth of CCF and CCH cells were examined. The temperature ranged from 24 to 32 °C for good growth of the cells. The growth rate of cells was higher in medium containing 10% FBS and the addition of bFGF to the medium significantly increased the growth rate. The CCF cells were found to be epithelial, while the CCH cells were fibroblastic in nature. The cytogenetic analysis of the cell lines revealed a diploid number of 100...

Insights into the genetic diversity of the leopard seal (Hydrurga leptonyx), inferred from mitochondrial DNA analysis, at Danco Coast, Antarctic Peninsula Boletín de Investigaciones
Hernández-Ardila,Laura Valentina; Barragán-Barrera,Dalia C.; Negrete,Javier; Poljak,Sebastián; Riet-Sapriza,Federico G.; Caballero,Susana.
ABSTRACT The leopard seal (Hydrurga leptonyx) is a genetically low-studied species. In order to conduct the first genetic diversity assessment of this species in Danco Coast (Antarctic Peninsula), 423 bp of the mitochondrial DNA Control Region (mtDNA-CR) was sequenced from 13 blood samples collected in Primavera Base (62º15’S, 58º39’W) during the 2011-2012 austral summer. Our results showed high haplotype diversity (h = 0.99), with various divergent haplotypes. Our findings suggest that leopard seals in the Danco Coast could represent different phylogroups; however, including more genetic markers are needed to confirm this hypothesis.

Genetic variability of brycon orbignyanus (valenciennes, 1850) (characiformes: characidae) in cultivated and natural populations of the upper paraná river, and implications for the conservation of the species BABT
Panarari-Antunes,Renata de Souza; Prioli,Alberto José; Prioli,Sônia Maria Alves Pinto; Galdino,Alexsandro Sobreira; Julio Junior,Horácio Ferreira; Prioli,Laudenir Maria.
This study aimed to evaluate the genetic variability of B. orbignyanus in cultivated and natural populations of the Upper Paraná River, using molecular RAPD markers and mtDNA control region. Specimens were collected in the Paraná River and in the Piracema fish farm in Maringá, State of Paraná, Brazil. RAPD primers produced 82 loci with consistent expression. The population from the Paraná River showed 28 polymorphic loci, whereas the population from the fish farm presented only 12. Data revealed genetic differentiation between the two populations, although not very pronounced. These results were corroborated by the principal coordinate analysis and by neighbor-joining clustering. The alignment of the D-loop sequences of B. orbignyanus indicated the...

Mitochondrial control region haplotypes of the South American sea lion Otaria flavescens (Shaw, 1800) BJMBR
Artico,L.O.; Bianchini,A.; Grubel,K.S.; Monteiro,D.S.; Estima,S.C.; de Oliveira,L.R.; Bonatto,S.L.; Marins,L.F..
The South American sea lion, Otaria flavescens, is widely distributed along the Pacific and Atlantic coasts of South America. However, along the Brazilian coast, there are only two nonbreeding sites for the species (Refúgio de Vida Silvestre da Ilha dos Lobos and Refúgio de Vida Silvestre do Molhe Leste da Barra do Rio Grande), both in Southern Brazil. In this region, the species is continuously under the effect of anthropic activities, mainly those related to environmental contamination with organic and inorganic chemicals and fishery interactions. This paper reports, for the first time, the genetic diversity of O. flavescens found along the Southern Brazilian coast. A 287-bp fragment of the mitochondrial DNA control region (D-loop) was analyzed. Seven...

Characterization of six rat strains (Rattus norvegicus) by mitochondrial DNA restriction fragment length polymorphism BJMBR
Hilsdorf,A.W.; Krieger,J.E..
Restriction fragment length polymorphism (RFLP) was used to examine the extent of mtDNA polymorphism among six strains of rats (Rattus norvegicus) - Wistar, Wistar Munich, Brown Norway, Wistar Kyoto, SHR and SHR-SP. A survey of 26 restriction enzymes has revealed a low level of genetic divergence among strains. The sites of cleavage by EcoRI, NcoI and XmnI were shown to be polymorphic. The use of these three enzymes allows the 6 strains to be classified into 4 haplotypes and identifies specific markers for each one. The percentage of sequence divergence among all pairs of haplotypes ranged from 0.035 to 0.33%, which is the result of a severe population constriction undergone by the strains. These haplotypes are easily demonstrable and therefore RFLP...

Maternal inheritance on reproductive traits in Brangus-Ibagé cattle Ciência Rural
Henkes,Luis Ernani; Benavides,Magda Vieira; Oliveira,João Francisco Coelho; Moraes,José Carlos Ferrugem; Weimer,Tania Azevedo.
Cytoplasmic inheritance influence on reproductive traits was investigated in the Brangus-Ibagé cattle (3/8 Nelore x 5/8 Aberdeen Angus). Additive genetic effects were responsible for 12% ± 11% of phenotypic variation observed in first calving interval, but their contribution dropped to zero when all calving intervals (CI) were considered. The heritability estimate for age at first calving (AFC, in days) was 0.19 ± 0.09. Mitochondrial lineage (MIT) had negligible effects on phenotypic variances of calving interval (0.0 ± 0.02), calf birth weight (0.0 ± 0.01), and cow weight at calving (0.0 ± 0.01). However, for the age at first calving, MIT accounted for 0.15 ± 0.07 of total variation. Cow weight at calving had a significant linear effect on CI and AFC....
