Cross-disciplinary approaches for measuring parasitic helminth viability and phenotype Anais da ABC (AABC)
Peak,Emily; Hoffmann,Karl F.
Parasitic worms (helminths) within the Phyla Nematoda and Platyhelminthes are responsible for some of the most debilitating and chronic infectious diseases of human and animal populations across the globe. As no subunit vaccine for any parasitic helminth is close to being developed, the frontline strategy for intervention is administration of therapeutic, anthelmintic drugs. Worryingly, and unsurprising due to co-evolutionary mechanisms, many of these worms are developing resistance to the limited compound classes currently being used. This unfortunate reality has led to a renaissance in next generation anthelmintic discovery within both academic and industrial sectors. However, a major bottleneck in this process is the lack of quantitative methods for...

Relationship between number of ovipositions of Cotesia flavipes (Cam.) and number of descendants emerged from its host Diatraea saccharalis (Fabr.) Anais da SEB
Yamauchi,Meire N.; Gobbi,Nivar; Chaud-Netto,José; Campos-Farinha,Ana E..
Females of Cotesia flavipes (Cam.) were individually introduced into Petri dishes containing one larva of its host Diatraea saccharalis (Fabr.). Each C. flavipes female was allowed to oviposit once or twice in the same larva. Time of emergence of larvae and adults, sex-ratio, number of non-viable larvae and pupae, and longevity of adults of C. flavipes were recorded. Data revealed that two successive ovipositions in the host did not affect sex-ratio, but increased number of parasitoids and number of non-viable larvae and pupae.

Intraspecific and interspecific pre-adult competition on the neotropical region colonizer Zaprionus indianus (Diptera: Drosophilidae) under laboratory conditions Bragantia
Galego,Luís Gustavo da Conceição; Carareto,Claudia Marcia Aparecida.
This study analyzes the pre-adult interactions of Zaprionus indianus, a recently-introduced species in Brazil, with two others Drosophilidae under laboratory conditions. The effects of larval residues on the viability and on the developmental time of Z. indianus, Drosophila simulans and D. sturtevanti were used to evaluate pre-adult competitive interactions, conditioning the culture medium with larval residues. Pre-adult interactions between Z. indianus, D. sturtevanti and D. simulans may affect their relative abundance over time, since the viability of Z. indianus was negatively affected by residues of D. sturtevanti, and its residues reduced the viability of D. simulans and the developmental time of both D. simulans and D. sturtevanti.

Physiological changes of Candida tropicalis population degrading phenol in fed batch reactor BABT
Komarkova,Eliska; Paca,Jan; Klapkova,Eva; Stiborova,Marie; Soccol,Carlos R; Sobotka,Miroslav.
Candida tropicalis can use phenol as the sole carbon and energy source. Experiments regarding phenol degradations from the water phase were carried out. The fermentor was operated as a fed-batch system with oxistat control. Under conditions of nutrient limitation and an excess of oxygen the respiration activity of cells was suppressed and some color metabolites (black-brown) started to be formed. An accumulation of these products inhibited the cell growth under aerobic conditions. Another impact was a decrease of the phenol hydroxylase activity as the key enzyme of the phenol degradation pathway at the end of the cell respiration activity. This decrease is linked with the above mentioned product inhibition. The cell death studied by fluorescent probe...

Yeast CUP1 protects HeLa cells against copper-induced stress BJMBR
Xie,X.X.; Ma,Y.F.; Wang,Q.S.; Chen,Z.L.; Liao,R.R.; Pan,Y.C..
As an essential trace element, copper can be toxic in mammalian cells when present in excess. Metallothioneins (MTs) are small, cysteine-rich proteins that avidly bind copper and thus play an important role in detoxification. Yeast CUP1 is a member of the MT gene family. The aim of this study was to determine whether yeast CUP1 could bind copper effectively and protect cells against copper stress. In this study, CUP1 expression was determined by quantitative real-time PCR, and copper content was detected by inductively coupled plasma mass spectrometry. Production of intracellular reactive oxygen species (ROS) was evaluated using the 2',7'-dichlorofluorescein-diacetate (DCFH-DA) assay. Cellular viability was detected using the...

Beijerinckia derxii stimulates the viability of non-N2-fixing bacteria in nitrogen-free media BJM
Barbosa,Heloiza R.; Thuler,Daniela Strauss; Shirakawa,Márcia Aiko; Miyasaka,Natália R. S..
The interactions between the nitrogen-fixing microorganism Beijerinckia derxii with two non-diazotrophic bacteria, either Escherichia coli or a facultative sulphur-oxidizing chemolitotroph, were studied in mixed cultures. Direct and indirect contact between B. derxii and E. coli were tested. B. derxii increased CFU numbers and/or maintained the viability of the non-diazotrophic bacteria, but neither growth nor nitrogenase activity of the nitrogen-fixing bacterium were affected by either partner.

Electrical stimulation of saccharomyces cerevisiae cultures BJM
Araújo,Ofelia Q.F.; Coelho,Maria Alice Z.; Margarit,Isabel C.P.; Vaz-Junior,Carlos A.; Rocha-Leão,Maria Helena M..
Modulation of cell endogenous membrane potential by an external electrical field influences the structure and function of membrane compartments, proteins and lipid bi-layer. In this work, the effects of applied potential on Saccharomyces cerevisiae growth were characterized through simple yet conclusive experiments. Cell growth time profile and cell division were investigated as macroscopic response to the electrical stimulation. Control experiments were conducted under identical conditions except for the absence of applied potential. Through comparative analysis, electrical stimulation was verified to alter cell cycle as smaller sized population was observed, suggesting that a synchrony in cell division was promoted. Power spectral analysis was employed...
