Identification of genes expressed by Cryptococcus gattii during iron deprivation BJM
Paula,Daphine Ariadne Jesus de; Silva,Lívia Kmetzsch Rosa e; Staats,Charley Christian; Vainstein,Marilene H.; Joanoni,Ana Lúcia Pinto; Nakazato,Luciano; Dutra,Valéria.
Cryptococcus neoformans and C. gattii are pathogenic yeasts that cause life-threatening diseases in humans and animals. Iron is an essential nutrient for virtually every organism as it functions as a cofactor in numerous essential enzymatic reactions. In the literature, the competition for iron between microbes and mammalian hosts during infection is well documented. In this study, we used representational difference analysis (RDA) in order to gain a better understanding of how C. gattii responds to iron starvation. A total of 15 and 29 genes were identified as having altered expression levels due to iron depletion after 3 h and 12 h, respectively. Of these, eight genes were identified in both libraries. The transcripts were related to many biological...

The uptake of different iron salts by the yeast Saccharomyces cerevisiae BJM
Gaensly,Fernanda; Picheth,Geraldo; Brand,Debora; Bonfim,Tania M.B..
Yeasts can be enriched with microelements, including iron; however, special physicochemical conditions are required to formulate a culture media that promotes both yeast growth and iron uptake. Different iron sources do not affect biomass formation; however, considering efficacy, cost, stability, and compatibility with Saccharomyces cerevisiae metabolism, ferrous sulphate is recommended.

Mobilization of endogenous glycogen and trehalose of industrial yeasts BJM
Paulillo,Silene Cristina de Lima; Yokoya,Fumio; Basso,Luiz Carlos.
The fermentation of yeast reserve carbohydrates, glycogen and trehalose is a procedure to increase protein level of yeast cells and improve ethanol production. This work studied on the degradation kinetics of glycogen and trehalose carried out with two industrial strains of Saccharomyces cerevisiae (PE-2 and SA-1) and the effect of different temperatures (38º, 40º, 42º and 44ºC) on degradation rate. Endogenous fermentation was carried out in a yeast suspension at 20% (w/v) based on wet weight, suspended in fermented media with 3.0 to 4.5 % (v/v) of ethanol. The degradation of the carbohydrate reserves at 40ºC followed first-order kinetics, showing that its rate is mainly dependent on the carbohydrate concentration in the cell. The degradation rate (k)...

Karimi,Maryam; Hassanshahian,Mehdi.
Abstract Phenol and phenolic compounds are environmental pollutants present in industrial wastewaters such as coal tar, oil refineries and petrochemical plants. Phenol removal from industrial effluents is extremely important for the protection of environment. Usually, phenol degradation is carried out by physicochemical methods that are costly and produce hazardous metabolites. Recently, phenol biodegradation has been considered. Yeasts are the most important phenol biodegraders. In this study, the phenol-degrading yeast from environmental samples (soil and wastewater) was isolated from the coking plant of Zarand, Kerman. Then total heterotrophic yeasts were counted. The soil samples had higher rates of yeast degrader, in comparison to wastewater samples....

Oxidative stress and antioxidant response in a thermotolerant yeast BJM
Mejía-Barajas,Jorge A.; Montoya-Pérez,Rocío; Salgado-Garciglia,Rafael; Aguilera-Aguirre,Leopoldo; Cortés-Rojo,Christian; Mejía-Zepeda,Ricardo; Arellano-Plaza,Melchor; Saavedra-Molina,Alfredo.
Abstract Stress tolerance is a key attribute that must be considered when using yeast cells for industrial applications. High temperature is one factor that can cause stress in yeast. High environmental temperature in particular may exert a natural selection pressure to evolve yeasts into thermotolerant strains. In the present study, three yeasts (Saccharomyces cerevisiae, MC4, and Kluyveromyces marxianus, OFF1 and SLP1) isolated from hot environments were exposed to increased temperatures and were then compared with a laboratory yeast strain. Their resistance to high temperature, oxidative stress, and antioxidant response were evaluated, along with the fatty acid composition of their cell membranes. The SLP1 strain showed a higher specific growth rate,...

Yeast biomass production: a new approach in glucose-limited feeding strategy BJM
Vieira,Érika Durão; Andrietta,Maria da Graça Stupiello; Andrietta,Silvio Roberto.
The aim of this work was to implement experimentally a simple glucose-limited feeding strategy for yeast biomass production in a bubble column reactor based on a spreadsheet simulator suitable for industrial application. In biomass production process using Saccharomyces cerevisiae strains, one of the constraints is the strong tendency of these species to metabolize sugars anaerobically due to catabolite repression, leading to low values of biomass yield on substrate. The usual strategy to control this metabolic tendency is the use of a fed-batch process in which where the sugar source is fed incrementally and total sugar concentration in broth is maintained below a determined value. The simulator presented in this work was developed to control molasses...
