Validation of reference genes for accurate normalization by quantitative polymerase chain reaction in sugarcane drought stress studies using two cultivars Ciência Rural

ABSTRACT: The aim of this study was to analyze the expression of putative reference genes in sugarcane under drought stress. The varieties RB72454 and RB72910 were cultivated and the treatments control and drought stress applied to 135-day-old plants grown under field conditions. The stress level of the plants was measured by rate of photosynthesis, transpiration, and stomatal conductance. For each biological replicate, expression analyses were conducted using quantitative polymerase chain reaction for the genes α-tubulin, β-tubulin, β-actin, cyclophilin, eukaryotic elongation factor 1, glyceraldehyde 3-phosphate dehydrogenase (GAPDH), histone H3 and ubiquitin. Animal welfare in experimental orthopedic surgery of dogs: an analysis of the Brazilian view Ciência Rural

ABSTRACT: The aim of this paper was to conduct a bibliometry search about Brazilian scientific publications that describe experimental orthopedic surgeries in dogs. It was analyzed 14 publications, and there was wide variation in the number of experimental animals, showing that there is no consensus on the appropriate number for science experiment with dogs. Most articles had been reviewed by an ethics committee (13/14, 92.85%) and only two articles (2/14; 14.28%) did not report the use of analgesic protocol. Among analgesics, morphine was the most widely used, noted in seven studies (7/14; 42.85%). Yield analysis of Physalis ixocarpa Brot. ex Hornem varieties under greenhouse and field conditions Ciência Rural

ABSTRACT: Physalis ixocarpa (husk tomato) is traditionally cultivated in Mexico, and nowadays the yield is low. In this study, four cultivars of husk tomato were evaluated, under greenhouse and field conditions. It was carried out a split-plot experimental design in a 4x2 factorial arrangement. The large plot was the growth conditions and the small plot was the variety. The microclimate, soil and yield variables were recorded. In the field, the Integrated Photosynthetic Active Radiation (IPAR) was higher 12mol m-2 d-1 than in the greenhouse. ABSTRACT: We aimed to apply genomic information based on SNP (single nucleotide polymorphism) markers for the genetic evaluation of the traits “stay-green” (SG), plant architecture (PA), grain aspect (GA) and grain yield (GY) in common bean through Bayesian models. These models were compared in terms of prediction accuracy and ability for heritability estimation for each one of the mentioned traits. Blackleg in cattle: current understanding and future research needs Ciência Rural

ABSTRACT: Blackleg is an endogenous acute infection that principally affects cattle, whose etiologic agent is the anaerobic bacterium Clostridium chauvoei. In recent years, the major virulence factors of C. chauvoei have been discovered and described. However, the pathogenesis of blackleg in cattle, and in particular, the movement of the pathogen from the point of entry to the affected tissues is not yet fully elucidated. Disease control is based on appropriate management and vaccination. This review summarizes the latest research findings that contribute toward the understanding of the disease in cattle, provide a foundation to preventive strategies, and identify future research needs.

ABSTRACT: This study aimed to evaluate the effects of adding probiotic culture (Bifidobacterium animalis subsp. Lactis Bb-12) and prebiotics (fructooligosaccharide - FOS) to yoghurt formulations stored at 4°C for 28 days, using an experimental design (independent variables: (0-3% of FOS and probiotic starter cultures 0-3%). The pH, acidity, fat, syneresis, protein, ºBrix, sugars, FOS and probiotic bacteria count were analyzed. ABSTRACT: Felis catus gammaherpesvirus 1 (FcaGHV1) may causes an asymptomatic infection that result in an efficient transmission and subsequently dissemination of the virus in feline population. This study used molecular detection by qPCR (quantitative PCR) based on DNA polymerase gene fragment amplification to evaluate the occurrence of FcaGHV1 and its correlation with other feline viral pathogens, such as Carnivore protoparvovirus 1 (CPPV-1), Felid alphaherpesvirus 1 (FeHV-1), and feline retroviruses such as feline immunodeficiency virus (FIV) and feline leukemia virus (FeLV). Diagnostic methods for identification of root-knot nematodes species from Brazil Ciência Rural

ABSTRACT: The accurate identification of root-knot nematode (RKN) species (Meloidogyne spp.) is essential for implementing management strategies. Methods based on the morphology of adults, isozymes phenotypes and DNA analysis can be used for the diagnosis of RKN. Traditionally, RKN species are identified by the analysis of the perineal patterns and esterase phenotypes. For both procedures, mature females are required. Over the last few decades, accurate and rapid molecular techniques have been validated for RKN diagnosis, including eggs, juveniles and adults as DNA sources. Here, we emphasized the methods used for diagnosis of RKN, including emerging molecular techniques, focusing on the major species reported in Brazil.

ABSTRACT: This paper aim to evaluate the ultrafiltration (UF) process for constituents recovery from whey. Sequences of factorial designs were performed by varying temperature (5 to 40°C) and pressure (1 to 3 bar), to maximize the proteins concentration using membrane of 100kDa in dead end system. Based on the best result new experiments were performed with membrane of 50kDa and 10kDa. With the membrane of 50 the protein retention was about 3 times higher than the membrane of 100kDa. Detection of virulence genes and the phylogenetic groups of Escherichia coli isolated from dogs in Brazil Ciência Rural

ABSTRACT: This study identified the virulence genes, pathovars, and phylogenetic groups of Escherichia coli strains obtained from the feces of dogs with and without diarrhea. Virulence genes and phylogenetic group identification were studied using polymerase chain reaction. Thirty-seven E. coli isolates were positive for at least one virulence factor gene. Twenty-one (57.8%) of the positive isolates were isolated from diarrheal feces and sixteen (43.2%) were from the feces of non-diarrheic dogs. Enteropathogenic E. coli (EPEC) were the most frequently (62.2%) detected pathovar in dog feces and were mainly from phylogroup B1 and E. Occurrence and antimicrobial resistance of Campylobacter jejuni and Campylobacter coli isolated from domestic animals from Southern Ecuador Ciência Rural

ABSTRACT: Occurrence and antimicrobial resistance profiles of Campylobacter jejuni and C. coli strains isolated from fecal samples of dogs, pigs, cows and hens in southern Ecuador were studied. Of the 250 samples studied, 84 (33.6%) were Campylobacter positive, with C. jejuni being more frequent (78.6%) than C. coli (21.4%), with the exception of porcine samples, from which C. coli was the only species isolated. Multidrug resistance was reported in 10 Campylobacter strains (11.9%), four C. jejuni and six C. coli, and was always associated with nalidixic acid and ciprofloxacin resistance. ABSTRACT: Owing to the high demand for healthier foods, the food industry has been marketing diet and light yogurts. The present study aimed to sensorially evaluate yogurts prepared using natural sweeteners through a simple centroid mixture design with three repetitions at the central point using the preference map methodology. Different formulations were processed using stevia, xylitol, and sucrose in delimited proportions based on the sweetness of the sweetener. A team of tasters was recruited to survey the sensory attributes of the formulations using the check-all-that-apply (CATA) methodology.
