Survey for neoplasia in Macoma balthica from the Gulf of Gdansk by flow cytometry ArchiMer

Using flow cytometry, 234 Macoma balthica were examined during a survey to determine frequency of neoplasia in the Gulf of Gdansk (Poland). Clams were collected in 4 locations and DNA content in gill tissue cells was determined by flow cytometry using propidium iodide staining. Cell permeabilization was induced by osmotic shock. Prevalence of neoplasia ranged from 9.6 to 26.7 % depending on location. DNA content in aneuploid cells was higher than in normal dividing cells. The fluorescence value for aneuploid cells corresponded to tetraploid/pentaploid cells. Monitoring hydrology is an essential part of studying an ecosystem such as the Marennes-Oléron Bay. Since 1977, bimonthly samples have been taken from five stations in the bay. These samples provide important information on physical characteristics (temperature, oxygen, salinity), nutrients (NH4, NO3, NO2, SiO3, PO4) and particulate matter composition (seston, chlorophyll a and pheophytin).

This study focuses on the situation of wild oyster beds of Crassostrea gigas against OsHV-1 μvar and presents a spatio-temporal description of the OsHV-1 contamination in wild oyster beds in the bay of Marennes-Oléron from a range of sampling based on a hydrodynamic model and analyzes conducted in 2010. The objectives were to describe wild populations of pacific oysters in Marennes Oleron bay; to characterize these populations regarding OsHV-1 prevalence for spat and adults in connection with the isolation level from reared stocks and the sampling periods.

Summer mortality of Pacific oysters is known in several countries. However no specific pathogen has been systematically associated with this phenomenon. A complex combination of environmental and biological parameters has been suggested as the cause and is now starting to be identified. A high genetic basis was found for survival in oysters when a first generation (G1) was tested in three sites during summer. The role of freshwater is more difficult to comprehend. It might act as a detrital organic matter "vector" (during floods) or an anthropic waste "vector" (toxic molecules from agriculture or other industries; nutritive salts from leaching...). Its effect has been demonstrated in the Bay of Veys (Ropert and Kopp, 2000; Misko, 2003), and especially its intense but transient action, presenting high freshenings, as recorded by high-frequency multiparameter probes since 1996 [15]. On this oyster culture site, mortality could then result from osmotic shocks. The "mortality-flow" relationship, demonstrated in 2003 in the Bay of Veys (Misko 2003), has to be specified. Depuis 2014, des épisodes de mortalité anormale affectent les cheptels mytilicoles tant adultes que juvéniles. Les années 2015 et 2016 ont permis de dégager des éléments de connaissance et des pistes relatifs aux facteurs biotiques et abiotiques qui semblent intervenir dans le phénomène de hausse de mortalité des moules depuis 2014. Le cheptel d'huîtres creuses, Crassostrea gigas, du Bassin de Marennes-Oléron a subi ces dernières années des morta lités (mai 88, mai 91, été 1992 et hiver 92-93). Les examens histopathologiques pratiqués durant celles-ci n'ont pas permis de déceler d'agents pathogènes. Les causes avaient été attribuées essentiellement, pour les premiers cas, à un manque de nourriture corrélé avec des conditions de sécheresse (donc un manque d'apport d'eau douce pour le bassin) et à des surcharges trop importantes des biomasses de fiItreurs, huîtres y compris. The "trophic risk" has been demonstrated in the bioenergetic model developed in vivo and based on various diet conditions similar to those of different types of oyster culture basins (eutrophic, heterotrophic and oligotrophic) [22, 23]. The trophic resource – mortality relationship differs from one basin to another, according to the quantity of this resource. Déterminisme des mortalités estivales d'huîtres creuses Crassostrea gigas dans le bassin de Marennes-Oléron : choix d'un site atelier pilote, prééchantillonnage et perspectives ArchiMer

Since 1988, seasonal, chronic mortality (up to 30-40 %) has been occurring at oyster farms in the Marennes-Oléron Bay, namely at the Ronce and Perquis sand flats. The particular geographic position on the southern part of the bay, between the Seudre River and the Maumusson Strait, confers a relative specificity to these oyster farming areas. Production in this area is on the order of 8,000 to 10,000 t of the 30,000 t of oysters produced annually in the Bay. Although limited in surface area (180 ha out of a total of 4,500 ha), this high proportion of production can be attributed to the fact that this zone is mostly dedicated to the finishing process before commercialisation. Due to its economic importance, oyster farming is vital for local businesses.

The objective of this document is to provide an overview of the research and studies carried out by Ifremer in Poitou-Charentes since the emergence of this phenomenon. Thiis report aims to study the oyster Crwsosrrea gigas growth and production over a 12 years time' s span in the bay of Marennes-DIeron. Results of 12 years of oyster growlh did not show any significant oyster biological productivity decrease, nor trend. ln contrast, annual growth showed a large variability, which will be correlated with environmental conditions. Traditionnellement pratiqué dans les marais maritimes autour du bassin de Marennes Oléron, l'affinage des huîtres en claires est destiné à améliorer leur qualité tant d'un point de vue organoleptique et engraissement que du durcissement des animaux. Le but recherché est d'obtenir un gain de poids de chair, une augmentation des réserves glucidiques, une saveur particulière, et un renforcement de la coquille. La claire ostréicole constitue par ailleurs une zone de stockage privilégiée du cheptel avant la commercialisation de la production au cours de la deuxième quinzaine du mois de décembre. These deaths possibly result from the cumulative effects of various stressors such as seasonal temperature variations, pesticide application and the release of contaminants from the resuspension of sediments.
