Biodegradation of acetonitrile by cells of Candida guilliermondii UFMG-Y65 immobilized in alginate, kappa-carrageenan and citric pectin BJM

Different encapsulation matrices were tested for immobilized cells of Candida guilliermondii UFMG-Y65 used for acetonitrile degradation. Acetonitrile degradation by free cells and cells immobilized in Ba-alginate, kappa-carrageenan and citric pectin was studied. The rate of acetonitrile degradation was monitored for 120 h by measuring yeast growth and ammonia concentration. Different alginate concentrations did not affect cell viability, but the period of incubation in BaCl2 solution reduced the number of viable cells. GPo1 alkB gene expression for improvement of the degradation of diesel oil by a bacterial consortium BJM

To facilitate the biodegradation of diesel oil, an oil biodegradation bacterial consortium was constructed. The alkane hydroxylase (alkB) gene of Pseudomonas putida GPo1 was constructed in a pCom8 expression vector, and the pCom8-GPo1 alkB plasmid was transformed into Escherichia coli DH5α. The AlkB protein was expressed by diesel oil induction and detected through SDS-polyacrylamide gel electrophoresis. The culture of the recombinant (pCom8-GPo1 alkB/E. coli DH5α) with the oil biodegradation bacterial consortium increased the degradation ratio of diesel oil at 24 h from 31% to 50%, and the facilitation rates were increased as the proportion of pCom8-GPo1 alkB/E. coli DH5α to the consortium increased. Biodegradation of polycyclic aromatic hydrocarbons by soil fungi BJM

Thirteen deuteromycete ligninolytic fungal strains were grown in media containing polycyclic aromatic hydrocarbons (PAHs), for 6 and 10 days. The PAHs were added directly with the inocula or on the third day of cultivation. A selection of the best strains was carried out based on the levels of degradation of the PAHs and also on the ligninolytic activities produced by the fungi. The selected strains were cultivated for 3, 6, 9, 12 and 15 days in the PAHs-containing media. Degradation of PAHs, as measured by reversed-phase HPLC on a C18 column, varied with each strain as did the ligninolytic enzymes present in the culture supernatants. Draft genome sequence of a caprolactam degrader bacterium: Pseudomonas taiwanensis strain SJ9 BJM

Abstract Pseudomonas taiwanensis strain SJ9 is a caprolactam degrader, isolated from industrial wastewater in South Korea and considered to have the potential for caprolactam bioremediation. The genome of this strain is approximately 6.2 Mb (G + C content, 61.75%) with 6,010 protein-coding sequences (CDS), of which 46% are assigned to recognized functional genes. This draft genome of strain SJ9 will provide insights into the genetic basis of its caprolactam-degradation ability.

Properties of catechol 1,2-dioxygenase in the cell free extract and immobilized extract of Mycobacterium fortuitum BJM

Polycyclic aromatic hydrocarbons (PAH) are carcinogenic compounds which contaminate water and soil, and the enzymes can be used for bioremediation of these environments. This study aimed to evaluate some environmental conditions that affect the production and activity of the catechol 1,2-dioxygenase (C12O) by Mycobacterium fortuitum in the cell free and immobilized extract in sodium alginate. The bacterium was grown in mineral medium and LB broth containing 250 mg L-1 of anthracene (PAH). ABSTRACT Role of microbes in bioremediation of oil spills has become inevitable owing to their eco friendly nature. This study focused on the isolation and characterization of bacterial strains with superior oil degrading potential from crude-oil contaminated soil. Three such bacterial strains were selected and subsequently identified by 16S rRNA gene sequence analysis as Corynebacterium aurimucosum, Acinetobacter baumannii and Microbacterium hydrocarbonoxydans respectively. Biodegradation of naphthalene and anthracene by chemo-tactically active rhizobacteria of populus deltoides BJM

Several naphthalene and anthracene degrading bacteria were isolated from rhizosphere of Populus deltoides, which were growing in non-contaminated soil. Among these, four isolates, i.e. Kurthia sp., Micrococcus varians, Deinococcus radiodurans and Bacillus circulans utilized chrysene, benzene, toluene and xylene, in addition to anthracene and naphthalene. Kurthia sp and B. circulans showed positive chemotactic response for naphthalene and anthracene. The mean growth rate constant (K) of isolates were found to increase with successive increase in substrate concentration (0.5 to 1.0 mg/50ml). Degradation of h-acid by free and immobilized cells of Alcaligenes latus BJM

Alcaligenes latus, isolated from industrial effluent, was able to grow in mineral salts medium with 50 ppm (0.15 mM) of H-acid as a sole source of carbon. Immobilization of Alcaligenes latus in Ca-alginate and polyurethane foam resulted in cells embedded in the matrices. When free cells and immobilized cells were used for biodegradation studies at concentration ranging from 100 ppm (0.3 mM) to 500 ppm (1.15 mM) degradation rate was enhanced with immobilized cells. Cells immobilized in polyurethane foam showed 100% degradation up to 350 ppm (1.05 mM) and 57% degradation at 500 ppm (1.5 mM). ABSTRACT Discharge of coke-oven wastewater to the environment may cause severe contamination to it and also threaten the flora and fauna, including human beings. Hence before dumping it is necessary to treat this dangerous effluent in order to minimize the damage to the environment. Conventional technologies have inherent drawbacks however, biological treatment is an advantageous alternative method. In the present study, bacteria were isolated from the soil collected from the sites contaminated by coke-oven effluent rich in phenol and cyanide. Phenol removal from industrial effluents is extremely important for the protection of environment. Usually, phenol degradation is carried out by physicochemical methods that are costly and produce hazardous metabolites. Recently, phenol biodegradation has been considered. Keratinolytic microorganisms have become the subject of scientific interest due to their ability to biosynthesize specific keratinases and their prospective application in keratinic waste management. Among several bacterial classes, actinobacteria remain one of the most important sources of keratin-degrading strains, however members of the Micrococcaceae family are rarely scrutinized in regard to their applicatory keratinolytic potential. A Gram-negative bacterium, designated as strain KB2, was isolated from activated sludge and was found to utilize different aromatic substrates as sole carbon and energy source. On the basis of morphological and physiochemical characteristics and 16S rRNA gene sequence analysis, the isolated strain KB2 was identified as Stenotrophomonas maltophilia. Further characterization of o-nitrobenzaldehyde degrading bacterium Pseudomonas sp: ONBA-17 and deduction on its metabolic pathway BJM

A previously reported o-nitrobenzaldehyde (ONBA) degrading bacterium Pseudomonas sp. ONBA-17 was further identified and characterized. Based on results of DNA base composition and DNA-DNA hybridization, the strain was identified as P. putida. Its degradation effect enhanced with increase of inoculum amount and no lag phase was observed. Higher removal rate was achieved under shaking conditions. All tested ONBA with different initial concentrations could be completely degraded within 5 d. Analysis of methane biodegradation by Methylosinus trichosporium OB3b BJM

The microbial oxidation of methane in the atmosphere is performed by methanotrophic bacteria that use methane as a unique source of carbon and energy. The objective of this work consisted of the investigation of the best conditions of methane biodegradation by methanotrophic bacteria Methylosinus trichosporium OB3b that oxidize it to carbon dioxide, and the use of these microorganisms in monitoring methods for methane. The results showed that M. trichosporium OB3b was capable to degrade methane in a more effective way with an initial microorganism concentration of 0.0700 g.L-1, temperature of 30ºC, pH 6.5 and using 1.79 mmol of methane. The objective of this study was to investigate the biodegradation of PCBs in northeastern (Heilongjiang Province), northern (Shanxi Province) and eastern China (Shanghai municipality). Benzene, toluene and xylene biodegradation by Pseudomonas putida CCMI 852 BJM

A minimal liquid medium containing benzene (B), toluene (T) and xylene (X) and mixtures thereof, was used to evaluate degradation activity of Pseudomonas putida CCMI 852 containing a TOL plasmid. Experiments were developed with B, T and X (100 mg L-1), with mixtures of BT, BX, and TX (50 + 50 mg L-1 each) and BTX (33.3 + 33.3 + 33.3 mg L-1 each), added to 500 mL of medium. After 18 to 24 hours, the inoculum was added and solvent disappearance was determined after 24 to 25 hours by GC. Results showed that P. putida CCMI 852 was able to metabolize T and X, but B was not metabolized. In a BTX mixture, B was not metabolized and T and X degradation rate decreased 50%.
