Thirteen deuteromycete ligninolytic fungal strains were grown in media containing polycyclic aromatic hydrocarbons (PAHs), for 6 and 10 days. The PAHs were added directly with the inocula or on the third day of cultivation. A selection of the best strains was carried out based on the levels of degradation of the PAHs and also on the ligninolytic activities produced by the fungi. The selected strains were cultivated for 3, 6, 9, 12 and 15 days in the PAHs-containing media. Degradation of PAHs, as measured by reversed-phase HPLC on a C18 column, varied with each strain as did the ligninolytic enzymes present in the culture supernatants. Highest degradation of naphthalene (69%) was produced by the strain 984, having Mn-peroxidase activity, followed by strain... |

Polycyclic aromatic hydrocarbons (PAH) are carcinogenic compounds which contaminate water and soil, and the enzymes can be used for bioremediation of these environments. This study aimed to evaluate some environmental conditions that affect the production and activity of the catechol 1,2-dioxygenase (C12O) by Mycobacterium fortuitum in the cell free and immobilized extract in sodium alginate. The bacterium was grown in mineral medium and LB broth containing 250 mg L-1 of anthracene (PAH). The optimum conditions of pH (4.0-9.0), temperature (5-70 ºC), reaction time (10-90 min) and the effect of ions in the enzyme activity were determined. The Mycobacterium cultivated in LB shown higher growth and the C12O activity was two-fold higher to that in the mineral... |

Several naphthalene and anthracene degrading bacteria were isolated from rhizosphere of Populus deltoides, which were growing in non-contaminated soil. Among these, four isolates, i.e. Kurthia sp., Micrococcus varians, Deinococcus radiodurans and Bacillus circulans utilized chrysene, benzene, toluene and xylene, in addition to anthracene and naphthalene. Kurthia sp and B. circulans showed positive chemotactic response for naphthalene and anthracene. The mean growth rate constant (K) of isolates were found to increase with successive increase in substrate concentration (0.5 to 1.0 mg/50ml). B. circulans SBA12 and Kurthia SBA4 degraded 87.5% and 86.6% of anthracene while, Kurthia sp. SBA4, B. circulans SBA12, and M. varians SBA8 degraded 85.3 %, 95.8 % and... |

Abstract Phenol and phenolic compounds are environmental pollutants present in industrial wastewaters such as coal tar, oil refineries and petrochemical plants. Phenol removal from industrial effluents is extremely important for the protection of environment. Usually, phenol degradation is carried out by physicochemical methods that are costly and produce hazardous metabolites. Recently, phenol biodegradation has been considered. Yeasts are the most important phenol biodegraders. In this study, the phenol-degrading yeast from environmental samples (soil and wastewater) was isolated from the coking plant of Zarand, Kerman. Then total heterotrophic yeasts were counted. The soil samples had higher rates of yeast degrader, in comparison to wastewater samples.... |

A Gram-negative bacterium, designated as strain KB2, was isolated from activated sludge and was found to utilize different aromatic substrates as sole carbon and energy source. On the basis of morphological and physiochemical characteristics and 16S rRNA gene sequence analysis, the isolated strain KB2 was identified as Stenotrophomonas maltophilia. Strain KB2 is from among different Stenotrophomonas maltophilia strains the first one described as exhibiting the activities of three types of dioxygenases depending on the structure of the inducer. The cells grown on benzoate and catechol showed mainly catechol 1,2- dioxygenase activity. The activity of 2,3-dioxygenase was detected after phenol induction. Protocatechuate 3,4-dioxygenase was found in crude cell... |

ABSTRACT Polychlorinated biphenyls (PCBs), the chlorinated derivatives of biphenyl, are one of the most prevalent, highly toxic and persistent groups of contaminants in the environment. The objective of this study was to investigate the biodegradation of PCBs in northeastern (Heilongjiang Province), northern (Shanxi Province) and eastern China (Shanghai municipality). From these areas, nine soil samples were screened for PCB-degrading bacteria using a functional complementarity method. The genomic 16S rDNA locus was amplified and the products were sequenced to identify the bacterial genera. Seven Pseudomonas strains were selected to compare the capacity of bacteria from different regions to degrade biphenyl by HPLC. Compared to the biphenyl content in... |

A minimal liquid medium containing benzene (B), toluene (T) and xylene (X) and mixtures thereof, was used to evaluate degradation activity of Pseudomonas putida CCMI 852 containing a TOL plasmid. Experiments were developed with B, T and X (100 mg L-1), with mixtures of BT, BX, and TX (50 + 50 mg L-1 each) and BTX (33.3 + 33.3 + 33.3 mg L-1 each), added to 500 mL of medium. After 18 to 24 hours, the inoculum was added and solvent disappearance was determined after 24 to 25 hours by GC. Results showed that P. putida CCMI 852 was able to metabolize T and X, but B was not metabolized. In a BTX mixture, B was not metabolized and T and X degradation rate decreased 50%. |

ABSTRACT The evaluation of the bioremediation potential of microbial with dyes in solid and liquid culture media has been described, but prior studies have not mentioned which culture method is most appropriate for selection of microorganisms. Therefore, the aim of this work was to evaluate the bioremediation potential of filamentous fungi in liquid and solid culture media with methylene blue. The fungi isolates tested were Pleurotus ostreatoroseus (POR-SP1, POR-SP2), P. ostreatus (DF39, EF58 and EF60), Pycnoporus sanguineus (PS) and Fusidium sp. (FUS). The methylene blue concentrations tested were 0, 5, 10, and 50 mg L-1 in the solid medium and 0, 5, 25, 50, and 100 mg L-1 in the liquid medium. In the solid medium, the mycelial diameters of DF39, EF58,... |
