Esterase electrophoresis of Clostridium perfringens bovine strains J. Venom. Anim. Toxins incl. Trop. Dis.
Baldassi,L.; Barbosa,M.L.; Bach,E.E.; Iaria,S.T..
Eighty-nine of 144 isolates of Clostridium perfringens obtained from 187 samples of 71 bovine in several Brazilian states were submitted to esterase electrophoresis for typing. Mobilites electrophoresis, as parameter, were settled down by isolates from ATCC pattern of types A, B, C, and D. Of the 89 isolates, 43 (48.3%) were characterized as electrophoretic type A, 20 (22.5%) as D, 18 (20.2%) as C, and 3 (3.4%) as B. Five (5.6%) isolates did not identity with any type. Similarly, esterase electrophoresis enabled the typing of 94.4 % of the isolates, demonstrating to be an appropriate method for animal sample analyses.

Virulence exaltation of Clostridium perfringens strains from bovines J. Venom. Anim. Toxins incl. Trop. Dis.
Baldassi,L.; Barbosa,M.L.; Bach,E.E.; Iaria,S.T..
Ten out of eighty-nine strains biochemically identified as Clostridium perfringens, isolated from bovine organs, were selected by their different results showed in toxigenicity test on mice. Those and the standard strains, ATCC types A, B, C, and D, had their virulence exalted through serial intramuscular inoculation into guinea pigs. Results showed that, for toxigenic strains (6), one or two passages were enough to cause exaltation, while for the atoxigenic (4), five or six inoculations were needed. Esterase electrophoresis of standard and isolated strains, with and without exaltation, was performed. Electrophoresis analysis permits the following conclusions: strains that do not show any clinical symptoms in mice, when exalted demonstrate decreased...

Gene Expression of 11β-HSD and Glucocorticoid Receptor in the Bovine (Bos taurus) Follicle During Follicular Maturation and Atresia : The Role of Follicular Stimulating Hormone OAK
TETSUKA, Masafumi; NISHIMOTO, Hiromi; MIYAMOTO, Akio; OKUDA, Kiyoshi; HAMANO, Seizo;
Glucocorticoids modulate ovarian function in cattle. However, their regulatory mechanisms have not been fully elucidated. In the present study, we examined gene expression of two glucocorticoid-metabolizing enzymes, a bidirectional 11β-HSD type 1 (11HSD1) and a dehydrogenase 11β-HSD type 2 (11HSD2), and glucocorticoid receptor (GR) in bovine follicles during follicular maturation and atresia. Granulosa cells (GCs) and theca interna layers (TIs) were harvested from follicles classified as small growing, dominant, preovulatory, early atretic and late atretic follicles. The expression levels of 11HSD1, 11HSD2 and GR mRNA were quantified by real-time PCR. In the healthy follicles, expression of 11HSD1 mRNA increased as follicles matured, both in GCs and TIs. A...

N-acetylglucosaminyltransferase I Activity of Bovine Oviduct Epithelial Cells: Stimulation by Luteinizing Hormone, Vascular Endothelial Growth Factor and Tumor Necrosis Factor α OAK
NOZAKI, Hirofumi; WIJAYAGUNAWARDANE, Missaka P.B.; KODITUWAKKU, Suranga P.; YOSHIDA, Takashi; NAKAMURA, Tadashi; ARAI, Ikichi; URASHIMA, Tadasu; MIYAMOTO, Akio;
N-acetylglucosaminyltransferase I (GnT I; EC 2.4.1.101), which catalyzes the first step in the conversion of oligomannose to complex or hybrid N-glycans of glycoproteins, was found in media cultured with bovine oviduct epithelial cells (BOEC) obtained from non-pregnant cows during the follicular phase. Combined treatment with specific hormones increased GnT I release from BOEC. Luteinizing hormone (LH; 10 ng/ml) alone slightly, but together with 17β-estradiol (E2; 1 ng/ml), synergistically increased GnT I activity. Vascular endothelial growth factor (VEGF) and tumor necrosis factor (TNF) α, which have been shown to have their highest activities in the bovine oviduct during the periovulatory period, also increased in GnT I activity. This study provides the...

The activity of α3-D-mannoside-β-1,2-N-acetylglucosaminyltransferase I (GnT I; EC 2.4.1.101), which catalyzes the first step in the conversion of oligomannose to complex or hybrid N-glycans of glycoproteins, was detected in bovine follicular fluid (bFF). The GnT I activity in bFF had a pH optimum of 5.8 and an absolute requirement for either Co2+, Mn2+, or Mg2+, the activity being stimulated by these cations in the above order. The apparent Km value for α1-3α1-6 mannopentaose of GnT I in bFF was 2.17 mM. The substrate specificity for the GnT I activity decreased in the following order: α1-3α1-6 mannopentaose> α1-3α1-6 mannotriose>α1-3 mannobiose. The GnT I activity in bFF from large atretic follicles was significantly higher than in that from large...

The purpose of this study was to investigate the effect of X-ray tube-cassette distance and image scanning time on image quality to establish the benefit of Computed Radiography (CR) in bovine clinical practice. The tube-cassette distance had no effect on the visual evaluation score (graininess and sharpness). The image scanning time correlated with graininess but not sharpness. From these results,it was concluded that accidental variations in the tube-cassette...

El-Sayed Ahmed Awad EL-SHAFAEY1) 2); AOKI, Takahiro; ISHII, Mitsuo; YAMADA, Kazutaka;
Twenty-one limbs of bovine cadavers (42 digits) were exposed to interdigital cassetteless imaging plate using computed radiography. The radiographic findings included exostosis, a rough planta surface, osteolysis of the apex of the distal phalanx and widening of the laminar zone between the distal phalanx and the hoof wall. All these findings were confirmed by computed tomography. The hindlimbs (19 digits) showed more changes than the forelimbs (10 digits), particularly in the lateral distal phalanx. The cassetteless computed radiography technique is expected to be an easily applicable method for the distal phalanx rather than a conventional cassette-plate and/or the film-screen cassetteless methods.

ACOSTA, Tomas J.; TETSUKA, Masa; MATSUI, Motozumi; SHIMIZU, Takashi; BERISHA, Bajram; SCHAMS, Dieter; MIYAMOTO, Akio;
The aim of the present in vivo study was to monitor real-time fluctuations of cortisol (Cr) in the wall of preovulatory follicles using a microdialysis system (MDS) implanted in the theca layer as well as changes in ovarian venous plasma (OVP) and jugular venous plasma (JVP). Seven cows were superovulated using FSH and prostaglandin F2α injections. Dialysis capillary membranes were surgically implanted into the theca layer of mature follicles and connected to a microdialysis system. Fractions of the perfusates were collected from Day -1 (Day 0=LH surge) to Day 3. No difference in the concentrations of Cr between JVP and OVP was detected throughout the experiment. Circulating concentrations of Cr ranged from 20 to 35 ng/ml 8 h after surgery in ovulatory and...

A Field study to Estimate the Prevalence of Bovine African Trypanosomosis in Butaleja District, Uganda OAK
JING, Zhang; MAGONA, Joseph W; SAKURAI, Tatsuya; THEKISOE, Oriel M. M; OTIM, Charles P; SUGIMOTO, Chihiro; INOUE, Noboru;
Prevalence of bovine trypanosomosis was determined from a total of 203 blood samples collected from Butaleja district, eastern Uganda. All samples were examined by microhematocrit centrifuge test (MHC), PCR and ELISA. ELISA was performed in accordance with the OIE standard procedures using Trypanosoma brucei gambiense procyclic form crude antigens. PCR were utilized to identify the species and the subspecies of trypanosome. The overall prevalence of bovine African trypanosomosis was 8.9% by MHC, and 45.3% by the ELISA. Since substantial number (12 out of 18) of MHC positive samples were negative in the PCR tests, we could not conclude the most epidemic trypanosome species in the studied area. Nevertheless, the PCR results suggests that the most prevalent...

We have previously shown that polymorphonuclear neutrophils (PMNs) are present in bovine oviduct fluid under physiological conditions, and that the oviduct provides a microenvironment that protects sperm from phagocytosis by PMNs. Alpha 1-acid glycoprotein (AGP) is a major acute-phase protein produced mainly in the liver that has immunomodulatory functions. AGP mRNA is expressed in extrahepatic organs, such as the lung, kidney, spleen, lymph node, uterus, and ovary. Therefore, in this study, we investigated, 1) the local production of AGP in the bovine oviduct, 2) the effect of AGP on the phagocytic activity of PMNs for sperm and superoxide production and 3) the impact of AGP desialylation on the PMN phagocytosis of sperm. The AGP gene was expressed in...

Computed tomography (CT) was used for diagnosis of brain abscess in a 6-month-old, Japanese black calf presented with neurological dysfunction, compulsive circling and vision disturbance. CT images showed asymmetric lateral ventricles, and presence of intra-cranial multiple low absorption lesions surrounded by capsule suggestive of abscess in the right cerebral hemisphere. Postmortem examination revealed marked swelling of right cerebral hemisphere and olfactory bulb. Multilocular large abscess containing creamy pus was found to occupy most area of periventricular and lateral ventricle. Fusobacterium necrophrum was isolated from the abscess contents as the causative agent. These results demonstrate that CT is useful tool for tentative diagnosis of bovine...

Classification of Bovine Follicles Based on the Concentrations of Steroids, Glucose and Lactate in Follicular Fluid and the Status of Accompanying Follicles OAK
NISHIMOTO, Hiromi; HAMANO, Seizo; HILL, Glen Alan; MIYAMOTO, Akio; TETSUKA, Masafumi;
A simple and clear means to identify the physiological status of follicles is essential for study of follicular biology. In the present study, we verified a novel classification procedure based on analysis of the follicular population and glucose concentration in follicular fluid (FF) as an alternative method to classify bovine follicles. Paired ovaries were collected from heifers, and the number of follicles and stage of the CL were recorded. Follicles were initially divided into the following 3 groups according to diameter and the ratio of E2 and P4 (E/P): E2 active (E-A: E/P≥1), E2 inactive (E-I: E/P<1, ≥8.5 mm) and small follicles (E/P<1, <8.5 mm). E-A follicles were easily identified as E2-rich dominant follicles and were further classified...

This study was to investigate the localization and distribution of eosinophils (EOS) in the bovine oviduct throughout the estrous cycle. Histological studies revealed more abundant EOS in the infundibula of the oviducts ipsilateral to the preovulatory dominant follicle and the ovulated ovary. The number of EOS was higher in the infundibula of the oviducts ipsilateral to the ovulated ovary than those of the oviducts contralateral to the ovulated ovary. The infundibula of the oviducts ipsilateral to the preovulatory dominant follicle had higher number of EOS than those of the oviducts ipsilateral to the mid-cycle corpus luteum. The number of EOS in the isthmus, but not in the ampulla, was higher in the outer layers (tunica muscularis and tunica serosa) than...

Bovine Oocytes and Early Embryos Express mRNA Encoding Glycerol Kinase but Addition of Glycerol to the Culture Media Interferes with Oocyte Maturation OAK
OKAWARA, Sumika; HAMANO, Seizo; TETSUKA, Masafumi;
Glycerol plays multi-functional roles in cellular physiology. Other than forming the backbone molecule for glycerophospholipid and triglyceride (TG), glycerol acts as an energy substrate for glycolysis. Spermatozoa are known to utilize glycerol for energy production, but there are no reports of this in oocytes. In this study, the value of glycerol as an energy substrate for bovine oocyte maturation (Exp. 1) and the gene expression of glycerol kinase (GK), an enzyme crucial for cellular glycerol utilization, in bovine oocytes and early embryos (Exp. 2) were examined. In Exp. 1, in vitro maturation (IVM) was conducted using synthetic oviduct fluid supplemented with/without glucose (1.5 mM) and/or glycerol (1.0 mM), and maturation rate, degree of cumulus...
