Isolation of alkalophilic CGTase-producing bacteria and characterization of cyclodextrin-glycosyltransferase BABT
Higuti,lma Hiroko; Grande,Simone Wichert; Sacco,Roberta; Nascimento,Aguinaldo José do.
One hundred and twenty five soil samples were collected from the regions of roots of corn, cassava, potato, bean, sugar cane, soya, and pumpkin. From these, 75 strains were isolated that produced a yellowish halo surrounding the colonies, due to a phenolphtalein-cyclodextrin (CD) complex, and these were selected as alkalophilic CGTase-producing bacteria. All the 75 strains were identified as Bacillus firmus by microscopy and biochemical tests. The activity of the CGTase's varied from 2² to 2(10) dilutions, when assayed by CD-trichloroethylene (TCE)-complex precipitation. Strain 31 that produced the enzyme at the higher level was selected, and its enzyme was partially purified by starch adsorption (x 17) in a yield of 51%. Maximum enzyme activity occurred at...

Microbial Synthesis and Characterization of Biodegradable Polyester Copolymers from Burkholderia Cepacia and Cupriavidus Necator Strains Using Crude Glycerol as Substrate BABT
Rodrigues,Plínio Ribeiro; Silvério,Tatiane Aparecida Barroso; Druzian,Janice Izabel.
Abstract Polymers are materials of pronounced importance in the modern world, since they are massively present in everyday life, especially in the form of packaging. However, most of these materials are derived from non-renewable sources and their disposal generates large volumes of waste, which is extremely damaging to natural environments. In this context, microbial biopolymers appear as a powerful alternative in the substitution of several applications of synthetic plastics, causing less harm to the environment, as they are biodegradable and produced from renewable sources. In this study, evaluation and characterization of polyhydroxyalkanoates (PHAs) produced by Cupriavidus necator (IPT 026 and IPT 027) and Burkholderia cepacia (IPT 119 and IPT 400),...

Purification and Characterization of a Polygalacturonase Produced by Wickerhamomyces anomalus BABT
Martos,María Alicia; Butiuk,Ana Paula; Rojas,Natalia Lorena; Hours,Roque Alberto.
The aim of this work was to study the purification and physicochemical properties of an endo-polygalacturonase (PG) produced by Wickerhamomyces anomalus isolated from the citrus fruit peels. The enzyme was purified to homogeneity from the culture filtrate of W. anomalus grown on the yeast nitrogen base medium with glucose as carbon and energy source and citrus pectin as inductor. After anion-exchange chromatography and gel filtration chromatography, PG activity was eluted as a single peak, yielding 21% of the original activity. After dialysis and cation-exchange chromatography, only one fraction with PG activity was obtained, recovering 56% of initial enzyme activity and 1.3-fold increase in specific activity. The molecular weight of the enzyme was...

Ribotyping, biotyping and capsular typing of Haemophilus influenzae strains isolated from patients in Campinas, southeast Brazil BJID
Lancellotti,Marcelo; Pace,Fernanda de; Stehling,Eliana Guedes; Villares,Maria Cecília Barisson; Brocchi,Marcelo; Silveira,Wanderley Dias da.
Forty-five Haemophilus influenzae strains isolated from patients were characterized based on biochemical characteristics. Their capsular types were determined by polymerase chain reaction (PCR); they were compared, using two molecular methods [ribotyping with a specific DNA probe amplified from the 16S rDNA region from H. influenzae and through restriction fragment length polymorphism (RLFP) of an amplified 16S DNA region]. The strains were better discriminated by the ribotyping technique that used the 16S probe and by the combination of both techniques. Biotypes I and IV were the most common, followed by biotypes VI, VIII and III. Biotypes II and VII were not found. Most of the capsular samples were nontypable (89%), with capsular types a and b found in 2...

Molecular epidemiology of Shigella spp strains isolated in two different metropolitam areas of southeast Brazil BJM
Angelini,Michelle; Stehling,Eliana Guedes; Moretti,Maria Luiza; Silveira,Wanderley Dias da.
Shigella spp., the human pathogen responsible for shigellosis, is highly infectious even at low levels. The incidence rate of shigellosis varies with geographical distribution, location human development index, and age groups, being higher among children aged under 5 years. In Brazil, a few works indicate that shigellosis cases are underestimated, with S. flexneri and S. sonnei strains being the major agents responsible for the shigellosis cases. The present study used pulsed field gel electrophoresis (PFGE) to investigate the molecular epidemiology of 119 strains of S. sonnei and S. flexneri isolated from shigellosis cases that occurred in the metropolitan areas of Ribeirão Preto and Campinas Cities, São Paulo Sate, southeast Brazil. The results indicated...

Isolation and characterization of novel lectins from Canavalia ensiformis DC and Dioclea grandiflora Mart. ex Benth. seeds Braz. J. Plant Physiol.
Melgarejo,Luz Marina; Vega,Nohora; Pérez,Gerardo.
Two lectins were isolated from Canavalia ensiformis and Dioclea grandiflora seeds. Gel filtration produced a fraction corresponding to Con A or D. grandiflora lectin while erythroagglutination assays revealed a distinct fraction presenting a lectin that agglutinates human red blood cells (RBCs) but not rabbit RBCs. Hydrophobic interaction chromatography showed that the latter fraction yielded a protein that readily agglutinates human erythrocytes; the lectin was also purified by affinity chromatography on Lac-Sepharose showing similar properties to that of the Phenyl-Sepharose-purified lectin. Despite minor differences (carbohydrate content or A1%1cm), the two lectins showed similar molecular properties in that they consisted of two non-covalently linked...
