Population genomics and gene introgression in goat herds naturally adapted to Brazil Rev. Ciênc. Agron.
Moura,Jeane de Oliveira; Campelo,José Elivalto Guimarães; Bajay,Miklos Maximiliano; Sarmento,José Lindenberg Rocha; Araújo,Adriana Mello de.
ABSTRACT The aim of this study was to apply the Illumina 50 K goat SNP Chip to analyse population genomic structure in two herds of Marota goats in the State of Piauí, one private and the other an official conservation herd, and to investigate evidence of genetic erosion in these herds caused by the Anglo-Nubian goat. To that end, 86 Marota and 10 Anglo-Nubian animals were genotyped. Genetic diversity was analysed by comparing minor allele frequency (MAF) in the herds. Population structure and genetic differentiation were evaluated using a Bayesian approach, principal component analysis (PCA) and the fixation index (FST). High genetic differentiation (FST = 0.16) was seen in the Marota population in relation to the Anglo-Nubian. The private herd shared a...

Non-hierarchical clustering of Manihot esculenta Crantz germplasm based on quantitative traits Rev. Ciênc. Agron.
Oliveira,Eder Jorge de; Aud,Fabiana Ferraz; Morales,Cinara Fernanda Garcia; Oliveira,Saulo Alves Santos de; Santos,Vanderlei da Silva.
ABSTRACT The knowledge of the phenotypic variation of cassava (Manihot esculenta Crantz) germplasm allows the estimative of the genetic variability to support the selection of contrasting genitors. Therefore, the aim of this work was to define homogeneous groups of cassava germplasm based on yield traits, disease resistance and root quality using K-means as a non-hierarchical method. Breeding values estimated by Best Linear Unbiased Predictor (BLUP) were used for the cluster analysis. The number of groups was defined according to the stabilization of the smallest within-group sum of squares. Seventeen clusters were defined to represent the diversity of the germplasm, whose number of accessions ranged from 7 (Group 15) to 69 (Group 9). In general,...

SSR markers to detect gene flow from upland to mocó cotton Rev. Ciênc. Agron.
Dantas,Ana Carolina Assis; Barroso,Paulo Augusto Vianna; Hoffmann,Lúcia Vieira; Alves,Milena Ferreira; Andrade,Francisco Pereira de.
Mocó cotton was the most important crop in Northeastern Brazil. Local varieties had been developed before governmental breeding programs, which should be identified and preserved as a genetic resource. The aim of this work was to select SSR markers which can be used to monitor gene flow from herbaceous to mocó cotton plants, using non transgenic upland cotton as a pollen source. Unique mocó cotton feral populations were identified in the state of Rio Grande do Norte, and mother plants and their seeds were collected. Allele frequencies were studied among fifteen plants from these populations using 64 SSR primer pairs. Ten SSR markers, with alleles exclusive to mocó or upland genotypes, were selected to monitor gene flow by paternity analysis (BNL3627; 2960;...

ABSTRACT: Genetic redundancy in cassava (Manihot esculenta Crantz) presents a challenge to efficient management of genetic resources. This study aimed to identify and define the genetic structure of duplicates in cassava germplasm from various Embrapa research units, using single-nucleotide polymorphism (SNP) markers. We evaluated 2,371 accessions with 20,712 SNPs. The identification of duplicates was performed based on multilocus genotypes (MLG), adopting a maximum genetic distance threshold of 0.05. The population structure was defined based on discriminant analysis of principal components (DAPC). A total of 1,757 unique and 614 duplicate accessions were identified. The redundancy of the collections ranged from 17 % (Belém, PA – Brazil) to 39 %...

Screening of Lycopersicon sp. accessions for resistance to Pepper yellow mosaic virus Scientia Agricola
Juhász,Ana Cristina Pinto; Silva,Derly José Henriques da; Zerbini Júnior,Francisco Murilo; Soares,Bruno Oliveira; Aguilera,George Alberto Hial.
The tomato is a crop of great economical importance, however it is susceptible to a large number of pests and diseases, including viral disease for which the best control strategy is genetic resistance. The disease, caused by Pepper yellow mosaic virus (PepYMV) has become a recent problem. Consequently, the idea of this work was to screen 376 accessions of Lycopersicon sp. to find possible sources of resistance to PepYMV. Out of 355 accessions of L. esculentum inoculated with PepYMV, 52 did not express symptoms. However, the virus reached high concentration in the tissues as measured by indirect ELISA, and therefore they were not considered as safe sources of resistance. Among 21 accessions of wild Lycopersicon species, one of L. hirsutum was shown to be...

Heterotic responses of tropical elite maize accessions from Latin America with Brazilian testers Scientia Agricola
Santos,Manoel Xavier dos; Pollak,Linda Maria; Carvalho,Hélio Wilson Lemos de; Pacheco,Cleso Antônio Patto; Gama,Elto Eugenio Gomes e; Guimarães,Paulo Evaristo de Oliveira; Andrade,Ramiro Vilela de.
Little emphasis has been placed on identifying new sources of tropical maize germplasm that can be used in breeding programs. Additional information on the performance and heterotic classification of tropical germplasm is needed. This study was conducted to identify elite maize accessions from Latin America that could contribute to increase the level of heterosis with the best heterotic patterns of Brazil. Seventy-two elite accessions from the Latin American Maize Project (LAMP) were crossed with the testers BR 105 (flint kernel Suwan background) and BR 106 (dent kernel Tuxpeño background). The 72 crosses plus 9 checks were evaluated using a simple 9 × 9 lattice in four locations that represent one tropical region. The combined analysis showed highly...

Cytogenetic identification of wheat-Psathyrostachys huashanica amphiploid × triticale progenies for English grain aphid resistance Scientia Agricola
Xie,Quan; Kang,Houyang; Sparkes,Debbie Louise; Tao,Shan; Hu,Zhiqin; Xu,Lili; Fan,Xing; Sha,Lina; Zhang,Haiqin; Wang,Yi; Zeng,Jian; Zhou,Yonghong.
English grain aphid (EGA, Sitobion avenae Fabricius) is an important pest in wheat (Triticum aestivum L.). To develop EGA-resistant varieties, introducing the desirable genes from related species is regarded as an efficient avenue. In this study, the F1, F2 and F3 plants derived from the cross of EGA-susceptible wheat-Psathyrostachys huashanica Keng ex Kuo amphiploid (PHW-SA, AABBDDNsNs) and EGA-resistant triticale (Zhongsi 828, AABBRR) were analyzed for EGA resistance. Consequently, PHW-SA was moderately susceptible while Zhongsi 828 and their F1 hybrids were immune, suggesting that the resistance is dominant. All the F2 plants showed high resistance or immunity over two years, indicating that EGA resistance genes are more likely carried by the rye...

Genetic diversity of table grape based on morphoagronomic traits Scientia Agricola
Leão,Patrícia Coelho de Souza; Cruz,Cosme Damião; Motoike,Sérgio Yoshimitsu.
The conservation and characterization of grape (Vitis spp) genetic resources in germplasm banks have been the basis of its use in breeding programs that result in development of new cultivars. There are at least 10,000 grape cultivars kept in germplasm collection. The genetic diversity in 136 table grape accessions from the state of Bahia, Brazil, was evaluated. Continuous and discrete morphoagronomic traits were assessed. The clustering analysis by the Tocher otimization method resulted in 30 clusters (considering continuous morphoagronomic traits), and 9 clusters (taking into consideration multicategorical traits). There was no agreement between clusters obtained by both, continuous or discrete phenotypic descriptors, independent of the cluster method...

Development of an SSR-based identification key for Tunisian local almonds Scientia Agricola
Gouta,Hassouna; Ksia,Elhem; Buhner-Zaharieva,Tatiana; Mliki,Ahmed; Gogorcena,Yolanda.
Ten simple sequence repeat (SSR) loci were used to study polymorphism in 54 almond genotypes. All genotypes used in this study originated from almond-growing areas in Tunisia with different climatic conditions ranging from the sub-humid to the arid and are preserved in the national collection at Sidi Bouzid. Using ten SSR, 130 alleles and 250 genotypes were revealed. In order to develop an identification key for each accession, the data were analysed separately for each microsatellite marker. The most polymorphic microsatellite (CPDCT042) was used as a first marker. Two microsatellite loci (CPDCT042 and CPDCT025) were sufficient to discriminate among all accessions studied. Neighbour-joining clustering and principal coordinate analysis were performed to...

Pre-breeding: a link between genetic resources and maize breeding Scientia Agricola
Nass,Luciano Lourenço; Paterniani,Ernesto.
Activities related to genetic resources are characterized by high cost and long term return. Thus the conservation of genetic variability for the future and the efficient utilization of available accessions are two important goals to be attained. However, the low utilization of germplasm banks is not restricted to Brazil but to other developing countries as well. Therefore, pre-breeding is a promising alternative to link genetic resources and breeding programs. Several aspects for maize are discussed such as concept, importance, methods and results. Problems to be investigated and suggestions are also presented.

New opportunities for developing tomato varieties with enhanced carotenoid content Scientia Agricola
Leiva-Brondo,Miguel; Valcarcel,Mercedes; Martí,Raúl; Roselló,Salvador; Cebolla-Cornejo,Jaime.
ABSTRACT The development of varieties with a high content of antioxidant compounds, such as carotenoids, has become a major focus in the marketing of tomato. Several mutants have been used in the development of high pigment varieties, but the significant influence of the environment on carotenoid content and the presence of negative side effects in vegetative growth and yield have limited the success of these variants. Consequently, the identification of alternative sources of variation in the quest for high carotenoid content is ongoing. In this study, 12 accessions of Solanum lycopersicum (including the former var cerasiforme) and S. pimpinelifolium have been evaluated in three different environments: open field and glasshouse cultivation at two sites....

Use of AFLPS to distinguish landraces of pejibaye (Bactris gasipaes) in brazilian Amazonia Scientia Agricola
Clement,Charles R.; Sousa,Nelcimar Reis; Rodrigues,Doriane Picanço; Astolfi-Filho,Spártaco; Moreno,Yolanda Núñez; Pascual,Vicente Torres; Rodríguez,Francisco Javier Gallego.
Although the first inhabitants of western Amazonia domesticated pejibaye (Bactris gasipaes Kunth, Palmae) or peach palm for its fruits, today it is widely planted for its heart-of-palm. Like other domesticates, pejibaye presents a complex hierarchy of landraces developed before the conquest of the Americas. The existence of three landraces (Pará, Solimões, Putumayo) was proposed along the Amazonas and Solimões Rivers, Brazil, based on morphological characteristics. There are some questions remaining about the intermediate landrace being an artifact of the morphometric analysis. AFLPs were used to evaluate the relationships among samples of these putative landraces. DNA was extracted from 99 plants representing 13 populations maintained in the Pejibaye...

Traditional Eastern Spanish varieties of tomato Scientia Agricola
Cortés-Olmos,Carles; Valcárcel,José Vicente; Roselló,Josep; Díez,Maria José; Cebolla-Cornejo,Jaime.
Despite the importance of traditional varieties of tomato (Solanum lycopersicum L.) as sources of variation in breeding programs and varieties targeted to high-price quality markets that value their exceptional organoleptic quality, little is known regarding the structure of these materials at the morphological level. In this study, a collection of 166 populations (137 of them during two years) of traditional varieties of tomato from the east coast of Spain has been characterized using 41 descriptors. The characterization revealed a considerable variation. The segregation observed in several populations (28 %) suggests that apart from the configuration as population varieties, the high variation present in these landraces may be partially due to possible...
