DNA damage in BALB/c mice infected with Lacazia loboi and its relation to nutritional status J. Venom. Anim. Toxins incl. Trop. Dis.
Barbosa,Adriana Sierra Assencio Almeida; Oliveira,Larissa Ragozo Cardoso de; Tavares,Francilene Capel; Lima,Carlos Roberto Gonçalves de; Diório,Suzana Madeira; Calvi,Sueli Aparecida; Vilani-Moreno,Fátima Regina; Pereira,Paulo Câmara Marques.
AbstractBackground Jorge Lobo’s disease, also known as lacaziosis, is a cutaneous-subcutaneous mycosis with chronic evolution. It is caused by the fungus Lacazia loboi. Herein we report a study that relates the genotoxicity caused by L. loboi in isogenic mice with nutritional status, through a normal or restricted diet. Methods DNA damage was assessed in the peripheral blood by the comet assay (tail intensity). Results The results for leukocytes showed increases in the mean tail intensity in mice under dietary restriction, in infected mice under dietary restriction and in infected mice ingesting a normal diet. Conclusion These results indicate that dietary restriction and L. loboi infection may increase DNA damage levels in mice, as detected by the comet assay.

Effects of manganese on fat snook Centropomus parallelus (Carangaria: Centropomidae) exposed to different temperatures Neotropical Ichthyology
Tuzuki,Bárbara Louise L.; Delunardo,Frederico Augusto C.; Ribeiro,Luciana N.; Melo,Caroliny P. de; Gomes,Levy Carvalho; Chippari-Gomes,Adriana Regina.
ABSTRACT This study evaluates the effects of exposure to manganese (Mn2+) for 96 hours at two different temperatures (24 and 27°C) on juveniles of Centropomus parallelus through the activities of glutathione S-transferase (GST) and catalase (CAT), micronuclei test (MN) and comet assay. The GST activity did not show any significant difference between the groups exposed to Mn2+ and the respective control groups; in contrast, a major increase in the CAT activity was observed at 27°C in the group exposed to Mn2+ compared to the control group. The genotoxic analyses showed that in all animals exposed to Mn2+, the number of red cells with micronuclei increased significantly compared to the respective control groups. There was also a significant increase in the...

DNA fragmentation and membrane damage of bocachico Prochilodus magdalenae (Ostariophysi: Prochilodontidae) sperm following cryopreservation with dimethylsulfoxide and glucose Neotropical Ichthyology
Martínez,José Gregorio; García,Víctor Atencio; Carrasco,Sandra Pardo.
The endangered bocachico Prochilodus magdalenae is a native freshwater fish of Colombia, the most captured species locally and one of the most important species for ex-situ conservation (germplasm banks). The aim of this study was to examine the effect of three concentrations of Dimethylsulfoxide (DMSO) (5%, 10%, 15%) and three of glucose (305, 333, 361 mM) in the extender on spermatic DNA fragmentation (F-DNA) (by Halomax®, Chromatin dispersion) and membrane damage (D-Me) (by eosin-nigrosin staining). After assessment of sperm quality by computer analysis of motility, one part of semen from males was diluted separately with three parts of extender and filled into 0.5 ml straws. Freezing was carried out in liquid nitrogen vapor dry shipper for 30 minutes...

Integrated biomarker response index using a Neotropical fish to assess the water quality in agricultural areas Neotropical Ichthyology
Vieira,Carlos Eduardo Delfino; Almeida,Mayara da Silva; Galindo,Bruno Ambrssio; Pereira,Lindalva; Martinez,Claudia Bueno dos Reis.
Aquatic ecosystems in areas with intense agricultural activity are subject to pesticide contamination, which may compromise the health of the fish. In order to verify the quality of the water and the possible effects of pesticides on fish, a method that combines different biomarker responses into an index named "integrated biomarker response" (IBR) was applied using the biological alterations in the Neotropical fish Astyanax altiparanae. Fish were maintained in situ at five sites along a stream that runs in an agricultural area and in a stream within a forest fragment, considered a reference site. After seven days of exposure the following alterations were observed in fish confined at experimental sites: increased activity of glutathione-S-transferase...

The agricultural impact of pesticides on Physalaemus cuvieri tadpoles (Amphibia: Anura) ascertained by comet assay Rev. Bras. Zool.
Gonçalves,Macks W.; Gambale,Priscilla G.; Godoy,Fernanda R.; Alves,Alessandro Arruda; Rezende,Pedro H. de Almeida; Cruz,Aparecido D. da; Maciel,Natan M.; Nomura,Fausto; Bastos,Rogério P.; Marco-Jr,Paulo de; Silva,Daniela de M..
ABSTRACT Amphibians inhabiting agricultural areas are constantly exposed to large amounts of chemicals, which reach the aquatic environment during the rainy season through runoff, drainage, and leaching. We performed a comet assay on the erythrocytes of tadpoles found in the surroundings of agricultural fields (soybean and corn crops), where there is an intense release of several kinds of pesticides in different quantities. We aimed to detect differences in the genotoxic parameters between populations collected from soybeans and cornfields, and between them and tadpoles sampled from non-agricultural areas (control group). Tadpoles collected from ponds located at soybean fields had significantly more DNA damage, followed by tadpoles collected from...
