Identificação dos insetos infestantes de alimentos através da micromorfologia de seus fragmentos Zoologia
Vargas,Carlos Henrry Bellot; Almeida,Armando Antunes de.
The identification of insect fragments showed that the infestation was done, mainly, by Coleoptera of the genus Sitophilus Sch›nherr, 1838 (Curculionidae) and Rhizoperthadominica (Fabricius, 1792) (Bostrychidae), that are caracteristic of internai infestation, and in lesser percentage by insects charac-teristic of externai infestation, as Crylolestes spp. (Cucujidae), Tribolium spp. (Tene-brionidae), Oryzaephilussurinamensis(Linnacus, 1758) (Cucujidae) and Lasioderma serricorne (Fabricius, 1792) (Anobiidae). From the total of recovered and identified insect fragments, 59% were fragments of elytra, 23% of mandibles, and in minor percentage by tarsal segments, claws, tibial spurs, legs, tarsus, eyes, antennae, exuviae and larvae urogomphi.

Screening of whole yeast free-cells and optimization of pH and temperature for fructooligosaccharides production Biological Sciences
Deffert, Flávia; Agustini, Bruna Carla; Picheth, Geraldo; Bonfim, Tania Maria Bordin.
Fructooligosaccharides are catalyzed by β–fructofuranosidase enzyme, produced by many microorganisms. However, in order to achieve a more profitable, low time-consuming process with lower cost, researchers have sought alternatives. This study aimed to select and identify yeasts able to produce fructooligosaccharides and evaluate the influence of pH and temperature on their synthesis. Yeast suspensions, solutions of 500 g L-1 sucrose and three values of pH (4.5, 5.5, and 6.5) and temperature (40, 50, and 60ºC) were tested. Yeast species were identified by molecular techniques. Among 141 yeast isolates from grapes, 65 were able to synthesize fructooligosaccharides. The maximum concentration of fructooligosaccharides was 4.8% (w v-1), and Saccharomyces...

Identification of Tibicen cicada species by a Principal Components Analysis of their songs Anais da ABC (AABC)
Ohya,Eiji.
Specific identification of three Tibicen cicadas, T. japonicus, T. flammatus and T. bihamatus, by their chirping sounds was carried out using Principal Components Analysis (PCA). High quality recordings of each species were used as the standards. The peak and mean frequencies and the pulse rate were used as the variables. Out of 12 samples recorded in the fields one fell in the vicinity of T. japonicus and all other were positioned near T. bihamatus. Then the cluster analysis of the PCA scores clearly separated each species and allocated the samples in the same way.

Thrips species (Insecta: Thysanoptera) associated to Fabaceae of agricultural importance in Cerrado and Amazon-Caatinga ecotone from Brazilian Mid-North Biota Neotropica
Lima,Élison Fabrício Bezerra; Monteiro,Renata Chiarini; Zucchi,Roberto Antonio.
Little is known about the Thysanoptera fauna from Brazilian Mid-North. In spite of being a region with at least three biomes and some ecotones, the studies on this insect order in the region are related to associations to a few cultivated plant species. On Fabaceae, plant family with the second largest number of records of thrips species associated in Brazil and huge economic importance for Mid-North, there are thrips records only on cowpea (Vigna unguiculata (L.) Walp.). Due to the lack of knowledge on the Thysanoptera fauna in such a diverse area and the importance of crops of this family in the region, this research aimed to survey the thrips species associated to Fabaceae of agricultural importance in areas of Cerrado and Amazon-Caatinga ecotone in...

An illustrated key to male Actinote from Southeastern Brazil (Lepidoptera, Nymphalidae) Biota Neotropica
Francini,Ronaldo Bastos; Penz,Carla M..
In certain times of the year, Actinote butterflies can be vastly abundant in Southeastern Brazil, thus representing conspicuous denizens of our fauna. Their wing coloration allows for the recognition of five major patterns, which probably resulted from mimicry. Furthermore, examination of series of field-collected and reared specimens clearly demonstrates a high intra-specific variation in wing color pattern. In concert, inter-specific resemblance (mimicry) and intra-specific variation make it difficult to discriminate Actinote species from each other. This key aims at providing a tool for identification of male Actinote. To that effect, we provide diagnostic characters and illustrations of wings and male genitalia for 22 species. We hope that the clear...

Species determination of Brazilian mammals implicated in the epidemiology of rabies based on the control region of mitochondrial DNA BJID
Carnieli Junior,Pedro; Fahl,Willian de Oliveira; Castilho,Juliana Galera; Brandão,Paulo Eduardo; Carrieri,Maria Luiza; Kotait,Ivanete.
Identification of animals that are decomposing or have been run over or burnt and cannot be visually identified is a problem in the surveillance and control of infectious diseases. Many of these animals are wild and represent a valuable source of information for epidemiologic research as they may be carriers of an infectious agent. This article discusses the results obtained using a method for identifying mammals genetically by sequencing their mitochondrial DNA control region. Fourteen species were analyzed and identified. These included the main reservoirs and transmitters of rabies virus, namely, canids, chiroptera and primates. The results prove that this method of genetic identification is both efficient and simple and that it can be used in the...

d'Azevedo,Pedro Alves; Siquiera,Itacy; Gugel,Juliana; Antunes,Ana Lúcia Souza; Secchi,Carina; Pasternak,Jacyr; Martino,Marinês Dalla Vale.
Automated instruments offer many advantages for clinical laboratories. Nevertheless, they can have problems identifying and determining susceptibilities of some pathogens. Vitek® 2 (bioMérieux) is an automated system that was recently introduced to Brazil. We evaluated the performance of this equipment for Brazilian isolates that had been characterized using reference identification and antimicrobial susceptibility testing methods. Ninety-nine strains of Gram-positive cocci from a local reference center collection were analyzed, consisting of 50 coagulasenegative Staphylococcus (CoNS) and 49 Enterococcus and related species. Vitek® 2 correctly identified 79.8% (79/99) of the isolates. Oxacillin resistance was detected in 76% (19/25) of resistant S....

Species distribution and antifungal susceptibility patterns of Candida isolates from a public tertiary teaching hospital in the Eastern Cape Province, South Africa BJMBR
Mnge,P.; Okeleye,B.I.; Vasaikar,S.D.; Apalata,T..
Candida species are the leading cause of invasive fungal infections, and over the past decade there has been an increased isolation of drug resistant Candida species. This study aimed to identify the species distribution of Candida isolates and to determine their unique antifungal susceptibility and resistance patterns. During a cross-sectional study, 209 Candida isolates (recovered from 206 clinical samples) were collected and their species distribution was determined using ChromAgar Candida. The Vitek-2 system (Biomerieux, South Africa) was used to determine minimum inhibitory concentrations (MICs) to azoles (fluconazole, voriconazole), echinocandins (caspofungin, micafungin), polyenes (amphotericin B) and flucytosine. Four species of Candida were...

Rapid identification of Mycobacterium tuberculosis complex by a novel hybridization signal amplification method based on self-assembly of DNA-streptavidin nanoparticles BJM
Wang,Haihe; Zhao,Chunyan; Li,Fan.
Rapid detection of Mycobacterium tuberculosis complex (MTBC) is a critical step in controlling tuberculosis (TB). In this study, we used IS6110 as the specific identification target to develop a novel hybridization signal amplification method (HSAM) for the rapid and direct detection of MTBC from clinical sputum specimens. This system consists of magnetic bead-linked capture probes for target isolation, dextranbased nanoparticles for amplifying the reporter molecule (biotinylated-FITC), and detection probes (2B-DNA) for binding the nanoparticles. Both the capture and detection probes were specific to the IS6110 target sequence. Our results determined that as few as 10 copies of the IS6110 sequence or 10 M. tuberculosis bacteria could be detected,...

16s-23S rDNA: polymorphisms and their use for detection and identification of Xylella fastidiosa strains BJM
Martinati,Juliana Camargo; Pacheco,Flávia Tereza Hansen; Miranda,Vitor Fernandes Oliveira de; Tsai,Siu Mui.
Strains of Xylella fastidiosa from several hosts (coffee, citrus, grape, almond, oleander, peach, plum, etc.) were characterized by analyzing the content of the nucleotide sequences of 16S-23S rDNA (coding for a small subunit ribosomal RNA) spacer region (ITS). Current methods for sequencing the ITS region yields partial sequences that do not contribute with significant information. According to this fact, new primers have been designed in order to obtain a complete sequence and facilitate the sequencing. The complete 16S-23S sequences from 08 strains were amplified through PCR using the primers designed in our lab. The 16S-23S sequences obtained were compared with 52 others sequences entries in GenBank database. The results revealed a higher level of...

Identification and adhesion profile of Lactobacillus spp. strains isolated from poultry BJM
Rocha,Ticiana Silva; Baptista,Ana Angelita Sampaio; Donato,Tais Cremasco; Milbradt,Elisane Lenita; Okamoto,Adriano Sakai; Andreatti Filho,Raphael Lucio.
In the aviculture industry, the use of Lactobacillus spp. as a probiotic has been shown to be frequent and satisfactory, both in improving bird production indexes and in protecting intestine against colonization by pathogenic bacteria. Adhesion is an important characteristic in selecting Lactobacillus probiotic strains since it impedes its immediate elimination to enable its beneficial action in the host. This study aimed to isolate, identify and characterize the in vitro and in vivo adhesion of Lactobacillus strains isolated from birds. The Lactobacillus spp. was identified by PCR and sequencing and the strains and its adhesion evaluated in vitro via BMM cell matrix and in vivo by inoculation in one-day-old birds. Duodenum, jejunum, ileum and cecum were...

Isolation, identification, and biocontrol of antagonistic bacterium against Botrytis cinerea after tomato harvest BJM
Shi,Jun-Feng; Sun,Chang-Qing.
ABSTRACT Tomato is one of the most important vegetables in the world. Decay after harvest is a major issue in the development of tomato industry. Currently, the most effective method for controlling decay after harvest is storage of tomato at low temperature combined with usage of chemical bactericide; however, long-term usage of chemical bactericide not only causes pathogen resistance but also is harmful for human health and environment. Biocontrol method for the management of disease after tomato harvest has great practical significance. In this study, antagonistic bacterium B-6-1 strain was isolated from the surface of tomato and identified as Enterobacter cowanii based on morphological characteristics and physiological and biochemical features combined...

Identification of pathogenic and nonpathogenic Leptospira species of Brazilian isolates by Matrix Assisted Laser Desorption/Ionization and Time Flight mass spectrometry BJM
Karcher,Daniel; Grenfell,Rafaella C.; Moreno,Andrea Micke; Moreno,Luisa Zanolli; Vasconcellos,Silvio Arruda; Heinemann,Marcos B.; Almeida Junior,Joao N. de; Juliano,Luiz; Juliano,Maria A..
ABSTRACT Matrix Assisted Laser Desorption/Ionization and Time of Flight mass spectrometry (MALDI-TOF MS) is a powerful tool for the identification of bacteria through the detection and analysis of their proteins or fragments derived from ribosomes. Slight sequence variations in conserved ribosomal proteins distinguish microorganisms at the subspecies and strain levels. Characterization of Leptospira spp. by 16S RNA sequencing is costly and time-consuming, and recent studies have shown that closely related species (e.g., Leptospira interrogans and Leptospira kirschneri) may not be discriminated using this technology. Herein, we report an in-house Leptospira reference spectra database using Leptospira reference strains that were validated with a collection...
