Two amylase genes, A and B, from the oyster Crassostrea gigas were characterized. Using PCR-RFLP, 6 and 4 alleles, respectively, were described for the amylase genes A and B. The roles of A and B amylase genes were investigated experimentally. They are expressed during larval and adult stages, and A transcripts are more abundant than B. The A transcript increases significantly with temperature, in high trophic conditions. However, A and B transcript levels do not change when food quantity increases although amylase activity augments. The level of B is correlated with dietary starch quantities, whereas the amount of A appears to remain constant ; simultaneously amylase activity decreases and the KM increases. Summer mortality associated with juveniles of the oyster Crassostrea gigas is probably the result of a complex interaction between the host, pathogens and environmental factors. Genetic variability in the host appears to be a major determinant in its sensitivity to summer mortality. Previously, divergent selection criteria based on summer survival have been applied to produce oyster families with resistant and susceptible progeny. In this paper, we describe the use of suppression subtractive hybridization to generate 150 C gigas clones that were differentially regulated between resistant and susceptible F2 progeny. Image analysis : a new method for estimating triploidy in commercial bivalves ArchiMer

For determination of ploidy levels in bivalves, three techniques are now routinely used: karyological determination. microfluorimetry and flow cytometry. This paper proposes an alternative technique which is based on determination of the optical density (OD) of stained nuclei using image analysis. A karyological and image analysis comparative experiment conducted on juveniles of Crassostrea gigas (Thunberg) and Ruditapes philippinarum (Adam & Reeve) is described and highlights the advantages of the image analysis technique. Infectivity and RNA Persistence of a Norovirus Surrogate, the Tulane Virus, in Oysters ArchiMer

Oysters, being filter feeders, can accumulate some human pathogens such as norovirus, a highly infectious calicivirus, most common cause of acute gastroenteritis worldwide. Accumulated virus decays over a period of days to weeks, possibly rendering contaminated oysters safe again. Sensitive molecular methods have been set up for shellfish analysis but without answering the question of infectious virus detection. Causes of decline of oyster production (Crassostrea virginica) in the Maryland portion of the Chesapeake Bay : a literature study ArchiMer

The historical landings of Eastern oyster Crassostrea virginica are described for the Maryland part of the Chesapeake Bay. The different trends are analyzed concurrently with the main events and management strategies which occurred. Three main periods are identified : (1) the great fishery when annual oyster landings from 1840 to 1890 reached 600,000 metric tons. This period was characterized by gross overfishing and the destruction of oyster habitat by the oyster gears; (2) the reduced but stable landings from 1900 to 1980, with failure of the reseeding plan connected to heavy sedimentation and anoxie summer conditions. Investigation of a hepatitis A outbreak due to oyster consumption in France in 2007 ArchiMer

Following the notification of nine hepatitis A cases partly clustered near an oyster growing area in the Côtes d’Armor district (Brittany, France), epidemiological, environmental and microbiological investigations were performed in order to identify the source and vehicle of contamination and devise control measures. The investigations identified 111 cases, all of whom lived or had stayed in the district. Of these cases, 87% had eaten raw bivalve molluscs and 81% had specifically eaten oysters. The raw molluscs originated from a single shellfish farm located at the north of Paimpol bay. We Studied the prevalence and intensity of the parasitic copepod Myicola ostreae in 2 closely related oysters Crassostrea angulata and C. gigas and their F1 hybrids. The effects on host and host reaction were also analysed to better understand host-parasite relationships between copepods and bivalve molluscs. Full reciprocal crosses were carried out between C. angulata and C. gigas and the progenies were reared in the wild in Ria Formosa Lagoon (Portugal), allowing natural infestation by M. ostreae. Improvement of remote setting of the Pacific oyster (Crassostrea gigas) on French platic pipes ArchiMer

The oysters culture in France is based on a natural spatfall, collected on cultches in setting areas. The collectors are then transfered on intertidal parcs for development during 1.5 to 3 years. The subsequent ongrowing stage is performed after separating the oysters and putting them in bags or by off-bottom cultivation. In order to improve the stock turnover and to improve the rearing conditions for the juveniles, experiments were performed to define optimal conditions for remote setting. The want of C. gigas spat-catching in the Marennes-oleron basin in 1981 can be due to various different reasons which are analysed here. Physico -chemical and trophic factors that can induce variations in the quality of gametogenesis, are successively in comparison with the year 1979 when reproduction was good. The biochemical evolution and a study of gonads slow a lack in maturation in 1981 even during the summer period. Late maturation of gonads bring about physiological consequences which induced mortality of cultivated oysters at the end of winter. A feeble recruit and a decrease of the cultivated stock have economic consequences which here sketched up.

Herpes-like virus infecting Japanese oyster (Crassostrea gigas) spat ArchiMer

During the summer of 1993, abnormal sporadic mass mortalities (80-90%) and morbidities occurred among four batches of young Japanese oyster (Crassostrea gigas) spat from the French Atlantic coast. These mortalities were only observed in July. Samples of affected animaIs were examined by electron microscopy. This paper reports the presence and appearance of herpes viruses in these samples.

A selective breeding program for Crassostrea virginica was established in 1997 as part of an initiative in Virginia to address declining oyster harvests caused by the two oyster pathogens Haplosporidium nelsoni (MSX) and Perkinsus marinus (Dermo). Housed in the Aquaculture Genetics and Breeding Technology Center (ABC), the objective of the program was to develop and disseminate disease-resistant lines that would enable an oyster culture industry. Le naissain d’huître creuse Crassostrea gigas est touché chaque année depuis 2008 par des mortalités causées par un microvariant de l’ostreid herpesvirus 1 (OsHV-1 μVar). Plusieurs études montrent que ces évènements de mortalité se produisent préférentiellement lorsque la température de l’eau est comprise entre 16°C et 24°C. Par conséquent, une exposition rapide des huîtres infectées par OsHV-1 à des températures supérieures à 24°C pourrait limiter les mortalités causées par le virus.
