Soletchnik, Patrick; Razet, Daniel; Geairon, Philippe; Faury, Nicole; Goulletquer, Philippe.
The study of maturation and spawning of the oyster is part of a research program to investigate the summer mortalities of the oysters, Crassostrea gigas in Marennes-Oléron Bay. Four maturity stages were simultaneously obtained by diet and thermal conditioning (immature, low maturation, mature and post-spawning stages). Measurements of clearance, filtration, absorption and respiration rates allowed a calculation of the scope for growth and hence an estimation of the oyster's energetic budget at various maturity stages. Male and female oysters had similar physiological responses. The filtration rate ranged from 2.4 to 2.6 1.h(-1) at the early stages of maturation and decreased to 1.8 1.h.' during the maturity stage. Growth rate resulting from gonad...

Analysis of Clinical Ostreid Herpesvirus 1 (Malacoherpesviridae) Specimens by Sequencing Amplified Fragments from Three Virus Genome Areas ArchiMer
Renault, Tristan; Moreau, Pierrick; Faury, Nicole; Pepin, Jean-francois; Segarra, Amelie; Webb, Stephen.
Although there are a number of ostreid herpesvirus 1 (OsHV-1) variants, it is expected that the true diversity of this virus will be known only after the analysis of significantly more data. To this end, we analyzed 72 OsHV-1 "specimens" collected mainly in France over an 18-year period, from 1993 to 2010. Additional samples were also collected in Ireland, the United States, China, Japan, and New Zealand. Three virus genome regions (open reading frame 4 [ORF4], ORF35, -36, -37, and -38, and ORF42 and -43) were selected for PCR analysis and sequencing. Although ORF4 appeared to be the most polymorphic genome area, distinguishing several genogroups, ORF35, -36, -37, and -38 and ORF42 and -43 also showed variations useful in grouping subpopulations of this...

Transcriptomic study of 39 ostreid herpesvirus 1 genes during an experimental infection ArchiMer
Segarra, Amelie; Faury, Nicole; Pepin, Jean-francois; Renault, Tristan.
Massive mortality outbreaks have been reported in France since 2008 among Pacific oysters, Crassostrea gigas, with the detection of a particular OsHV-1 variant called μVar. Virus infection can be induced in healthy spat in experimental conditions allowing to better understand the disease process, including viral gene expression. Although gene expression of other herpesviruses has been widely studied, we provide the first study following viral gene expression of OsHV-1 over time. In this context, an in vivo transcriptomic study targeting 39 OsHV-1 genes was carried out during an experimental infection of Pacific oyster spat. For the first time, several OsHV-1 mRNAs were detected by real-time PCR at 0 h, 2 h, 4 h, 18 h, 26 h and 42 h post injection. Several...

Soletchnik, Patrick; Razet, Daniel; Geairon, Philippe; Faury, Nicole; Goulletquer, Philippe.
The study of sexual maturation and spawning in the Pacific oyster (Crassostrea gigas) is part of a vast research programme that endeavours to understand the causes of mortality that occur sporadically during the spring and summer seasons in the Marennes-Oléron Bay. Thermal and diet conditioning were used to obtain oysters at each stage of maturity simultaneously. Using the measured rates of clearance, consumption, absorption and respiration provided estimates of growth potential and gave the energetic budget of oysters at different stages of sexual maturity. Physiological responses were similar for males and females. Filtration decreased from 2.4 to 2.6 l.h (-1) to 1.8 l.h (-1) with increasing maturity. Weight gain was associated with gonad development and...

Antiviral immunity in the Pacific oyster, Crassostrea gigas: last development and perspective ArchiMer
Renault, Tristan; Faury, Nicole; Barbosa, Valerie; Moreau, Kevin; Haffner, Philippe; Pepin, Jean-francois.
Herpes-like viruses have been reported last decades in various marine mollusc species in association with mortality outbreaks throughout the world (Renault & Novoa 2004). One of these viruses isolated during French outbreaks has been characterised as an unassigned member of the Herpesviridae family and named ostreid herpes virus 1 (OsHV-1) or Pacific oyster herpes virus (Davison et al. 2005). Studying anti-viral non-specific defence mechanisms (innate immunity) is of great interest, because they constitute the only one in molluscs. Therefore, innate immunity has be investigated in the Pacific cupped oyster, Crassostrea gigas. The main aim of the work was focused on the identification and the characterisation of genes induced by OsHV-1 in the Pacific...

Analysis of hemocyte parameters in Pacific oysters, Crassostrea gigas, reared in the field — Comparison of hatchery diploids and diploids from natural beds ArchiMer
Gagnaire, Beatrice; Soletchnik, Patrick; Faury, Nicole; Kerdudou, Nolwenn; Le Moine, Olivier; Renault, Tristan.
Internal defence mechanisms of bivalves include the hemocytes which assume several functions. Relationships exist between environmental factors, physiological state of oysters, defence system and pathogens. Hatchery oysters are more and more produced and reared in the field. The characteristics and particularly the defence system of these animals are poorly documented. In this context, two field trials were carried out from May to September 2002 and from April to July 2003 in order to monitor hemocyte parameters of diploid oysters from natural beds and hatchery-produced diploid oysters using flow cytometry. Several hemocyte parameters (cell mortality, granulocyte percentage, phagocytosis, esterase and peroxidase activities) were monitored. For both oyster...

This study aims to assess the broodstock origin and cultural effects on oyster summer mortality rates. Three oyster batches, two originating from the Marennes-Oléron Bay, and one from the Normandy rearing area were deployed in March 2000 on the Ronce Perquis oyster bank, on experimental site since 1996. On bottom cultured oysters were monitored until January 2001. By the end of the experiment, the cumulative mortality ranged from 51 to 58 %. Following on initial mortality rate (8.8 % to 9.3 %), oyster batches showed a critical period early July. In 15 days, mortality rate reached 13.7 %. Over the rearing period, the oyster shell growth ranged between 8.9 g and 12.7 g. The somatic gain represented 0.44-0.48 g of dry meat weight. Spawning event occured in...

First comparison of French and Australian OsHV-1 mu vars by bath exposure ArchiMer
Burge, Colleen A.; Reecez, Kimberly S.; Dhar, Arun K.; Kirkland, Peter; Morga, Benjamin; Degremont, Lionel; Faury, Nicole; Wippel, Bryanda J. T.; Macintyre, Alanna; Friedman, Carolyn S..
Economically devastating mortality events of farmed and wild shellfish due to infectious disease have been reported globally. Currently, one of the most significant disease threats to Pacific oyster Crassostrea gigas culture is the ostreid herpesvirus 1 (OsHV-1), in particular the emerging OsHV-1 microvariant genotypes. OsHV-1 microvariants (OsHV-1 mu vars) are spreading globally, and concern is high among growers in areas unaffected by OsHV-1. No study to date has compared the relative virulence among variants. We provide the first challenge study comparing survival of naive juvenile Pacific oysters exposed to OsHV-1 mu vars from Australia (AUS mu var) and France (FRA mu var). Oysters challenged with OsHV-1 mu vars had low survival (2.5%a exposed to AUS...

Extracellular products (ECPs) of the French V. tubiashii strain 07/118 T2 were previously reported to be toxic for the Pacific oyster Crassostrea gigas. In this study we now assessed host cellular immune responses and bacterial potential effectors by which these ECPs can be associated with host damages. The adhesion capacity (28% inhibition) and phagocytosis ability (56% inhibition) of oyster hemocytes were the main functions affected following in vitro contact between hemocytes and V. tubiashii ECPs. This may be linked to the demonstration of the capability of ECPs to cleave various cellular substrates as oyster collagen. Moreover, a strong metalloproteolytic activity was recorded with general (azocasein) and specific (ADAM) substrates and characterized...

Arnaud, Christophe; Faury, Nicole; Bouquet, Jean-paul.
The Marennes-Oléron is a well known cupped oyster (Crassostrea gigas) and mussel (Mytilus edulis) reproduction area. Every year, a monitoring of the reproduction of these two species is carried out on behalf of the Regional Shellfish Culture Section of Marennes-Oléron. This monitoring aims to determine the periods of larval emission in order to let the professionals know about the best collection times This study presents the main results obtained in 1999 and compares them with those obtained in 1997 and 1998. The year 1999 showed good results as far as larval emission and development are concerned in both oysters and mussels. The comparison of the three consecutive years shows the disparity of larval evolutions.

Robert, Stephane; Goulletquer, Philippe; Soletchnik, Patrick; Geairon, Philippe; Le Moine, Olivier; Razet, Daniel; Faury, Nicole; Taillade, Sylvie.
Within its oyster quality project, the Marennes-Oleron Professional Organization (SRC) aims to improve the zootechnic rearing conditions in ponds for the Pacific cupped oyster C. gigas "fines de claires" and "speciales de claires", presently specified by the French norm AFNOR NF V45-056 (AFNOR, 1985). The proposed modifications concern a seasonal management of the oyster biomass per m2 , using 1 and 3kg/m2, during 2 weeks from April 1rst to October 31, and 3 weeks from November 1 to March 31, respectively. The Shellfish Aquaculture Laboratory of Poitou-Charentes (LCPC) carried out experiments to address the professional and decision-makers request for an assessment of those modifications on the final oyster quality compared to the current rearing...

Ratiskol, Jacqueline; Faury, Nicole.

Cellular and molecular responses of haemocytes from Ostrea edulis during in vitro infection by the parasite Bonamia ostreae ArchiMer
Morga, Benjamin; Renault, Tristan; Faury, Nicole; Chollet, Bruno; Arzul, Isabelle.
Bonamia ostreae is a protozoan, affiliated to the order Haplosporidia and to the phylum Cercozoa. This parasite is intracellular and infects haemocytes, cells notably involved in oyster defence mechanisms. Bonamiosis due to the parasite B. ostreae is a disease affecting the flat oyster, Ostrea edulis. The strategies used by protozoan parasites to circumvent host defence mechanisms remain largely unknown in marine bivalve molluscs. In the present work, in vitro experiments were carried out in order to study the interactions between haemocytes from O. edulis and purified parasite. B. ostreae. We monitored cellular and molecular responses of oyster haemocytes by light microscopy, flow cytometry and real-time PCR 1, 2, 4 and 8 h p.i. Light microscopy was used...

Vibrio gigantis sp nov., isolated from the haemolymph of cultured oysters (Crassostrea gigas) ArchiMer
Le Roux, Frederique; Goubet, A; Thompson, F; Faury, Nicole; Gay, Melanie; Swings, J; Saulnier, Denis.
Polyphasic analysis of four new Vibrio isolates originating from the haemolymph of diseased cultured oysters is described. The new isolates were closely related to Vibrio splendidus, having 98 % 16S rRNA gene sequence similarity. Phylogenetic analysis based on DNA gyrase subunit B (gyrB), RNA polymerase Quo factor (rpoD), replication origin-binding protein (rctB) and transmembrane regulatory protein (toxR) genes, fluorescent amplified fragment length polymorphism and DNA-DNA hybridization experiments clearly showed that the new isolates form a tight genomic group that is different from the currently known Vibrio species. It is proposed that these new isolates should be accommodated in a novel species, Vibrio gigantis sp. nov. Phenotypic features that...

Effect of seasonal variation in trophic conditions and the gametogenic cycle on d13C and d15N levels of diploid and triploid Pacific oysters Crassostrea gigas ArchiMer
Malet, Nathalie; Sauriau, Pierre-guy; Faury, Nicole; Soletchnik, Patrick; Guillou, Gael.
Carbon and nitrogen stable isotopes were investigated in separate organs of diploid and sterile triploid Pacific oysters Crassostrea gigas for 13 mo, together with changes in chemical and isotope composition of suspended matter sampled from an intertidal mudflat within Marennes-Oléron Bay, France. Particulate organic matter (POM) was a mixture of pelagic and benthic material with a predominance of neritic phytoplankton in spring, and resuspended microphytobenthos in summer and autumn. A remarkable shift of +30/00 in d13C was reflected in both diploids and triploids from spring to summer, and further temporal differences were observed amongst their tissues. Seasonal changes in POM d15N were also reflected in oyster tissues, with digestive gland and muscle...
