Eudesmane Sesquiterpenes from Verbesina lanata with Inhibitory Activity against Grapevine Downy Mildew Organic Eprints
Ramseyer, Justine; Thuerig, Barbara; De Mieri, Maria; Schärer, Hans-Jakob; Oberhänsli, Thomas; Gupta, Mahabir P.; Tamm, Lucius; Hamburger, Matthias; Potterat, Olivier.
An in-house library of more than 3000 extracts of plant and fungal origin was screened against some major plant pathogens. As one of the hits, an ethyl acetate extract from inflorescences of Verbesina lanata showed significant inhibitory activity in vitro against grapevine downy mildew (Plasmopara viticola), with a MIC100 value of 35 μg/mL. An emulsifiable concentrate formulation with 50 mg/g of the extract was developed for in vivo evaluation. A suspension of the formulation containing 1 mg/mL of extract lowered leaf surface infection of grapevine seedling by 82% compared to the nontreated control. With the aid of HPLC-based activity profiling, the antifungal activity was correlated with a series of lipophilic compounds. Preparative isolation by a...

Versuche 2005 mit neuen Kartoffelsorten für den Biolandbau Organic Eprints
Speiser, Bernhard; Amsler, Thomas; Tamm, Lucius.

Proposals for Regulation of Botanicals Organic Eprints
Tamm, Lucius; Speiser, Bernhard; Mercier, Thierry.
Plants and plant extracts, here called 'botanicals', have been used for plant protection for a long time. Quantitatively, the most important botanical is pyrethrum, followed by azadirachtin, rotenone and essential oils. The current regulatory systm for pesticides is often seen as a major hurdle for the market introduction of new botanicals. The EU-funded Specific Support Action project 'REBECA' has held a series of workshops with stakeholder representatives. The following propopsals for improvement were elaborated: (1) development of a specific guidance document for botanicals; (2) adapted requirements concerning the characterisation of the active substance(s); (3) relaxations concerning identification and analytical methods for 'impurities'; (4) adapted...

Efficacy of a Magnolia officinalis bark extract against grapevine downy mildew and apple scab under controlled and field conditions Organic Eprints
Thuerig, Barbara; Ramseyer, Justine; Hamburger, Matthias; Ludwig, Mathias; Oberhänsli, Thomas; Potterat, Olivier; Schärer, Hans-Jakob; Tamm, Lucius.
In organic agriculture, the control of several diseases is largely depending on copper fungicides. Yet, copper can accumulate in the soil if the annual input exceeds annual uptake by plants, which can have a negative impact on soil fertility. Its use should thus be avoided or reduced. The aim of the present study was to evaluate the efficacy of a bark extract of Magnolia officinalis Rehder and Wilson to control three pathogens including Plasmopara viticola (Berk. & M.A. Curtis) Berl. & De Toni (causing grapevine downy mildew), Venturia inaequalis (Cooke) G. Winter (causing apple scab), and Phytophthora infestans (Mont.) De Bary (causing potato and tomato late blight) under controlled and field conditions, and to identify compounds responsible for...

Site-specific field resistance of grapevine to Plasmopara viticola correlates to altered gene expression and was not modulated by the application of organic amendments Organic Eprints
Thürig, Barbara; Slaughter, Ana; Marouf, Elaheh; Held, Matthias; Mauch-Mani, Brigitte; Tamm, Lucius.
The influence of site on resistance of grapevine (cv. Chasselas) to Plasmopara viticola was evaluated. Grapevine leaves from three vineyards in the region of Lake Neuchâtel (Switzerland) were tested for their susceptibility to P. viticola in the lab in five successive years (2004–2008), and the expression levels of four selected defence-related genes (Glucanase, Lipoxygenase 9, 9-cis epoxycarotenoid dioxygenase, Stilbene synthase) were studied in 1 year. In all 5 years of examination, differences between sites were substantial. In four out of 5 years, plants from site Hauvernier were much less susceptible to P. viticola than plants from site Auvernier. In another year, differences were less pronounced but still significant for one leaf age. Susceptibility...

Re-establishment of suppressiveness to soil- and air-borne diseases by re-inoculation of soil microbial communities Organic Eprints
Thürig, Barbara; Fließbach, Andreas; Berger, Nicole; Fuchs, Jacques G.; Kraus, Noemy; Mahlberg, Nicole; Nietlispach, Bruno; Tamm, Lucius.
The aim of this study was to investigate the potentials and limitations in restoring soil suppressiveness in disturbed soils. Soils from three sites in UK and Switzerland (STC, REC, THE) differing in their level of suppressiveness to soil-borne and air-borne diseases were γ-irradiated and this soil matrix was re-inoculated with 1% (w/w) of either parent native soil or native soil from the other sites (‘soil inoculum’). Suppressiveness to air-borne and soil-borne diseases was quantified by means of the host–pathogen systems Lepidium sativum (cress)–Pythium ultimum, an oomycete causing root rot and seedling damping-off, and Arabidopsis thaliana–Hyaloperonospora parasitica, an oomycete causing downy mildew. Soil microbial biomass, activity and community...
