CD19+ B cell subsets in the peripheral blood and skin lesions of psoriasis patients and their correlations with disease severity BJMBR
Lu,J.; Ding,Y.; Yi,X.; Zheng,J..
T lymphocytes are important in the pathogenesis of psoriasis, and increasing evidence indicates that B cells also play an important role. The mechanisms of action, however, remain unclear. We evaluated the ratios of CD19+ B cells in peripheral blood mononuclear cells (PBMCs) from 157 patients with psoriasis (65 patients with psoriasis vulgaris, 32 patients with erythrodermic psoriasis, 30 patients with arthropathic psoriasis, and 30 patients with pustular psoriasis) and 35 healthy controls (HCs). Ratios of CD19+ B cells in skin lesions were compared with non-lesions in 7 erythrodermic psoriasis patients. The Psoriasis Area Severity Index (PASI) was used to measure disease severity. CD19+ B cell ratios in PBMCs from psoriasis vulgaris (at both the active...

Rhein induces apoptosis of human gastric cancer SGC-7901 cells via an intrinsic mitochondrial pathway BJMBR
Li,Yiwen; Xu,Yuqing; Lei,Bo; Wang,Wenxiu; Ge,Xin; Li,Jingrui.
Rhein is a primary anthraquinone found in the roots of a traditional Chinese herb, rhubarb, and has been shown to have some anticancer effects. The aim of the present study was to investigate the effect of rhein on the apoptosis of the human gastric cancer line SGC-7901 and to identify the mechanism involved. SGC-7901 cells were cultured and treated with rhein (0, 50, 100, 150, and 200 µM) for 24, 48, or 72 h. Relative cell viability assessed by the MTT assay after treatment was 100, 99, 85, 79, 63% for 24 h; 100, 98, 80, 51, 37% for 48 h, and 100, 97, 60, 36, 15% for 72 h, respectively. Cell apoptosis was detected with TUNEL staining and quantified with flow cytometry using annexin FITC-PI staining at 48 h after 100, 200 and 300 µm rhein. The percentage...

Age-related changes of the multidrug resistance P-glycoprotein function in normal human peripheral blood T lymphocytes BJMBR
Machado,C.G.; Calado,R.T.; Garcia,A.B.; Falcão,R.P..
The multidrug resistance P-glycoprotein is a transmembrane efflux pump expressed by lymphocytes and is involved in their cytolytic activity. In the present study, we investigated the age-related changes of P-glycoprotein function in normal peripheral blood lymphocytes. Blood samples from 90 normal volunteers (age range, 0 to 86 years) were analyzed. P-glycoprotein function was assessed by the flow cytometric rhodamine 123 assay. P-glycoprotein function was highest in cord blood and progressively declined with age in peripheral blood T CD4+ and CD8+ cells. In contrast, P-glycoprotein function did not vary with age in CD19+ B or CD16+CD56+ natural killer cells. These data suggest that the decline in P-glycoprotein function in T CD4+ and CD8+ lymphocytes as a...

Distribution of HNA and LNA bacterial groups in the Southwest Atlantic Ocean BJM
Andrade,Luciana; Gonzalez,Alessandra M.; Rezende,Carlos Eduardo; Suzuki,Marina; Valentin,Jean Louis; Paranhos,Rodolfo.
Bacterioplankton was studied in a large area of Southwest Atlantic Ocean between 13 and 25ºS and 28 and 42ºW. Samples were collected in 108 stations at 20 m depth. Bacteria were enumerated by flow cytometry after nucleic acid staining with syto13 and two subgroups were differentiated: low nucleic acid content (LNA) and high nucleic acid content (HNA) bacteria. Total bacterial numbers varied from 0.37 to 5.53 10(5) cells mL-1. HNA cells represented 15 to 70% of the total number while LNA cells represented 30 to 85%. Heterotrophic bacterial production was determined by incorporation of tritiated leucine and ranged from 2.7 to 171.07 ng C L-1 h-1. No significant correlation was found between abundance and production. Nevertheless with support of multivariate...

Goat umbilical cord cells are permissive to small ruminant lentivirus infection in vitro BJM
Martins,Gabrielle R.; Marinho,Rebeca C.; Bezerra Junior,Rosivaldo Q.; Alves,Antoniel de O.; Câmara,Lilia M.C.; Albuquerque-Pinto,Luiz C.; Teixeira,Maria F. da S..
Abstract Small ruminant lentiviruses isolated from peripheral blood leukocytes and target organs can be propagated in vitro in fibroblasts derived from goat synovial membrane cells. These cells are obtained from tissues collected from embryos or fetuses and are necessary for the establishment of the fibroblast primary culture. A new alternative type of host cells, derived from goat umbilical cord, was isolated and characterized phenotypically with its main purpose being to obtain cell monolayers that could be used for the diagnosis and isolation of small ruminant lentiviruses in cell culture. To accomplish this goal, cells were isolated from umbilical cords; characterized phenotypically by flow cytometry analysis; differentiate into osteogenic,...

Heterotrophic bacteria abundances in Rodrigo de Freitas Lagoon (Rio de Janeiro, Brazil) BJM
Gonzalez,Alessandra M.; Paranhos,Rodolfo; Lutterbach,Márcia S..
The Rodrigo de Freitas Lagoon (RJ, Brazil) is an important coastal ecosystem that has been submitted to an accelerated degradation process. The aim of this work was to determine the abundance and the spatial distribution of total heterotrophic (by flow cytometry) and cultivated bacteria ("pour plate" method on R2A agar). Another objective was to evaluate the lagoon's influence on water quality of Ipanema and Leblon beaches. Physical and chemical data were acquired too. Sub-superficial water samples were taken monthly, from December 1999 to October 2000. On lagoon, the cultivated bacteria abundance varied from 6.9x10(5) to 5.0x10(7) CFU.100 mL-1. On Ipanema and Leblon beaches, this parameter yielded 1.4x10(5) and 2.8x10(6) CFU.100 mL-1, respectively. Total...

Thomas,Angel Treasa; Rao,Josyula Venkata; Subrahmanyam,Volety Mallikarjuna; Chandrashekhar,Hariharapura Raghu; Maliyakkal,Naseer; Kisan,Tukaram Kedar; Joseph,Alex; Udupa,Nayanabhirama.
Extracts from natural products, especially microorganisms, have served as a valuable source of diverse molecules in many drug discovery efforts and led to the discovery of several important drugs. Identification of microbial strains having promising biological activities and purifying the bio-molecules responsible for the activities, have led to the discovery of many bioactive molecules. Extracellular, as well as intracellular, extracts of the metabolites of thirty-six bacterial and twenty-four fungal isolates, grown under unusual conditions such as high temperature, high salt and low sugar concentrations, were in vitro tested for their cytotoxic potential on various cancer cell lines. The extracts were screened on HeLa and MCF-7 cell lines to study the...

Effects of different concentrations of sodium azide and chloramphenicol on the preservation of raw milk samples Ciência Rural
Vancin,Franciele Rampazzo; Webber,Bruna; Bondan,Carlos; Daroit,Luciane; Santos,Luciana Ruschel dos; Rodrigues,Laura Beatriz.
ABSTRACT: The preservation of milk samples for microbiological analyses by the Brazilian Network of Milk Quality Control Laboratories requires the addition of preservatives to maintain the microbiota from the time of sample collection to the moment of analysis. The number of microorganisms can change as a result of the active ingredients and concentration of the preservative, as well as due to interactions between the preservatives, incubation time, and packaging temperature. The objective of this research was to evaluate the conservation potential of different concentrations of sodium azide and chloramphenicol on the analytical shelf life of milk samples. Two farms were selected, one with a low bacterial count and one with a high bacterial count. The milk...
