The neurotoxic effects of vitamin A and retinoids Anais da ABC (AABC)
OLIVEIRA,MARCOS ROBERTO DE.
Vitamin A (retinol) and its congeners - the retinoids - participate in a panoply of biological events, as for instance cell differentiation, proliferation, survival, and death, necessary to maintain tissue homeostasis. Furthermore, such molecules may be applied as therapeutic agents in the case of some diseases, including dermatological disturbances, immunodeficiency, and cancer (mainly leukemia). In spite of this, there is a growing body of evidences showing that vitamin A doses exceeding the nutritional requirements may lead to negative consequences, including bioenergetics state dysfunction, redox impairment, altered cellular signaling, and cell death or proliferation, depending on the cell type. Neurotoxicity has long been demonstrated as a possible...

Biochemical and histological characterization of tomato mutants Anais da ABC (AABC)
Monteiro,Carolina C.; Rolão,Milca B.; Franco,Mônica R.; Peters,Leila P.; Cia,Mariana C.; Capaldi,Flávia R.; Carvalho,Rogério F.; Gratão,Priscila L.; Rossi,Mônica L.; Martinelli,Adriana P.; Peres,Lázaro E.P.; Azevedo,Ricardo A..
Biochemical responses inherent to antioxidant systems as well morphological and anatomical properties of photomorphogenic, hormonal and developmental tomato mutants were investigated. Compared to the non-mutant Micro-Tom (MT), we observed that the malondialdehyde (MDA) content was enhanced in the diageotropica (dgt) and lutescent (l) mutants, whilst the highest levels of hydrogen peroxide (H2O2) were observed in high pigment 1 (hp1) and aurea (au) mutants. The analyses of antioxidant enzymes revealed that all mutants exhibited reduced catalase (CAT) activity when compared to MT. Guaiacol peroxidase (GPOX) was enhanced in both sitiens (sit) and notabilis (not) mutants, whereas in not mutant there was an increase in ascorbate peroxidase (APX). Based on PAGE...

Low dissolved oxygen levels increase stress in piava (Megaleporinus obtusidens): iono-regulatory, metabolic and oxidative responses Anais da ABC (AABC)
COPATTI,CARLOS E.; BOLNER,KEIDI C.S.; LONDERO,ÉRIKA P.; ROSSO,FELIPE L. DE; PAVANATO,MARIA A.; BALDISSEROTTO,BERNARDO.
Abstract: The aquatic environment presents daily and/or seasonal variations in dissolved oxygen (DO) levels. Piava faces different DO levels in the water due to its distributional characteristics. The goal of this study was to describe the effects of low DO levels on plasma ion, biochemical and oxidative variables in piava juveniles. Fish were exposed to different DO levels, including 1.0, 2.0, 3.0, 4.0 and 5.0 mg L-1 of DO for 96 h, after which blood and tissue samples (liver, kidney, gill and muscle) were collected. The decrease in DO levels decreased plasma Na+, Cl-, K+ and NH3 levels as well as protein and glycogen levels in the liver, kidney and muscle; increased Na+/K+-ATPase activity in the gills and kidney as well as glucose and ammonia levels in...

In vitro effect of globotriaosylceramide on electron transport chain complexes and redox parameters Anais da ABC (AABC)
ALVARIZ,RAFAELA M.; MOREIRA,ISABEL T.D.S.; CURY,GABRIELA K.; VARGAS,CARMEN R.; BARSCHAK,ALETHÉA G..
Abstract: Fabry disease (FD) is an X-linked inherited disease and occurs due to mutations in GLA gene that encodes the α-galactosidase enzyme. Consequently, there is an accumulation of enzyme substrates, namely globotriaosylceramide (GB3). FD is a multisystemic disease, caused by storage of GB3 in vascular endothelia, with significant renal, cardiac and vascular involvement. The aim of this work was to evaluate the in vitro effect of GB3 on electron transport chain complexes (ETC) and redox parameters. Biochemical biomarkers were determined in homogenates of cerebral cortex, kidneys and liver of Wistar rats in the presence or absence of GB3 at concentrations of 3, 6, 9 and 12 mg/L. We found that GB3 caused an increase of ETC complexes II and IV activities,...

LUNARDI,RAFAELA F.; WOHLENBERG,MARIANE; MEDEIROS,NIARA; AGOSTINI,FABIANE; FUNCHAL,CLÁUDIA; DANI,CAROLINE.
Oxidative stress has been considered as one of the factors responsible for hepatic diseases, which sometimes require new ways of treatment. The present study aimed to evaluate the in vitro antioxidant capacity of the tea of Echinodorus grandiforus (“leather hat” plant) in rat liver. Different preparations of tea were evaluated for phenolic composition, antioxidant activity by DPPH assay and ability to inhibit lipid peroxidation induced by copper sulfate. The antioxidant activity was assessed in liver tissue treated with sodium azide in the presence or absence of tea by assays for lipid peroxidation (TBARS), protein oxidation (carbonyl) and the antioxidant enzymes catalase (CAT) and superoxide dismutase (SOD). The results show that different preparations of...

Antioxidant effect of simvastatin throught oxidative imbalance caused by lisdexamfetamine dimesylate Anais da ABC (AABC)
Eger,Guilherme A.; Ferreira,Vinícius V.; Batista,Camila R.; Bonde,Henrique; Lima,Daniela D. de; Wyse,Angela T.S.; Cruz,Júlia N. da; Rodrigues,André F.; Magro,Débora D. Dal; Cruz,José G.P. da.
The present study aims to directly investigate the behavioral and antioxidant effects of simvastatin in a model of bipolar mania induced by lisdexamfetamine dimesylate. Wistar rats were treated for 30 days with simvastatin. On the 24th day after the start of treatment, each rat was administered lisdexamfetamine dimesylate for 7 days. The results suggest that simvastatin combined with lisdexamfetamine dimesylate induced a significant increased locomotion and lisdexamfetamine dimesylate administration causes an oxidative imbalance determined by an increment in lipid peroxidation, protein oxidation and alterations in the activities of antioxidant enzymes in brain areas; moreover, in the presence of simvastatin, most of these effects were prevented. These...

Regular exercise, overweight/obesity and sedentary lifestyle cause adaptive changes in thiol–disulfide homeostasis Anais da ABC (AABC)
GOL,MEHMET; ÖZKAYA,BEYTULLAH; YILDIRIM,CANER; BAL,RAMAZAN.
Abstract: Dynamic thiol–disulfide homeostasis is considered to have critical roles in maintenance of physiological functioning. We aimed to reveal whether there is any specific aberration in thiol–disulfide homeostasis in three distinct categories of individuals, including those who 1) exercise regularly (fitness group), 2) have a sedentary lifestyle (sedentary group) and 3) are overweight or obese (overweight/obese group). 72 male individuals were included in the study, 21 of whom were in fitness group, 28 of whom were overweight or obese and 23 of whom had a sedentary lifestyle. Plasma native thiol (-SH) and total thiol [(-SH) + (-S-S-)] levels were quantitatively determined. Total thiol levels in sedentary group were significantly lower than those in...

Endothelial, renal and hepatic variables in wistar rats treated with Vancomycin Anais da ABC (AABC)
BRUNIERA,FELIPE R.; FERREIRA,FELIPE M.; SAVIOLI,LUIZ R.M.; BACCI,MARCELO R.; FEDER,DAVID; PEREIRA,EDIMAR C.; PEDREIRA,MAVILDE L.G.; PETERLINI,MARIA A.S.; PERAZZO,FÁBIO F.; AZZALIS,LIGIA A.; ROSA,PAULO C.P.; JUNQUEIRA,VIRGINIA B.C.; SATO,MONICA A.; FONSECA,FERNANDO L.A..
Vancomycin (VCM) is indicated in combat against Gram-positive infections, but it is not considered a first-choice drug because of its adverse effects. It is believed that oxidative stress is the primary mechanism of endothelial injury and the consequent VCM toxicity, which varies from phlebitis to nephrotoxicity. Moreover, dose recommendations, dilution, rates and types of infusion are still controversial. The aim of this study was to determine the effect of different VCM dilutions in endothelial, liver and kidney injuries by biochemical parameters and histopathological analysis. Wistar rats were randomly divided into six groups and subjected to femoral vein cannulation for drug administration. Control groups received 0.9 ml of saline and the others...

Abstract Our aim is to investigate the potentially preventive effects of Aliskiren in a carrageenan-induced lung pleurisy model and to compare the standard anti-inflammatory agents, indomethacin and dexamethasone. The pleurisy model was induced through the injection of carrageenan (0.2 ml-%2) into the pleural cavity. After the experiment, serum and lung tissues were collected and biochemical, molecular and pathological examinations were performed. In our study, pleural inflammation decreased superoxide dismutase activity and the glutathione level and increased the malondialdehyde level in the lung of rats, while Aliskiren increased the superoxide dismutase activity and glutathione level and decreased the malondialdehyde level. In addition,...

The pentose phosphate pathway in Trypanosoma cruzi: a potential target for the chemotherapy of Chagas disease Anais da ABC (AABC)
Igoillo-Esteve,Mariana; Maugeri,Dante; Stern,Ana L.; Beluardi,Paula; Cazzulo,Juan J..
Trypanosoma cruzi is highly sensitive to oxidative stress caused by reactive oxygen species. Trypanothione, the parasite's major protection against oxidative stress, is kept reduced by trypanothione reductase, using NADPH; the major source of the reduced coenzyme seems to be the pentose phosphate pathway. Its seven enzymes are present in the four major stages in the parasite's biological cycle; we have cloned and expressed them in Escherichia coli as active proteins. Glucose 6-phosphate dehydrogenase, which controls glucose flux through the pathway by its response to the NADP/NADPH ratio, is encoded by a number of genes per haploid genome, and is induced up to 46-fold by hydrogen peroxide in metacyclic trypomastigotes. The genes encoding...

Abstract Silicone breast implant is associated with complications inherent to the surgical procedure. Prosthesis coating with polyurethane, however, commonly reduces the incidence of such complications. In this paper, the authors evaluated the inflammatory histomorphometric profile and oxidative damage associated to the implant of polyester urethane sheets. Forty-eight Wistar rats were divided into Sham or polyester urethane groups (n = 8/group) and underwent a polyester urethane implant in the dorsal skinfold. Tissue samples were collected on days seven, 30, and 90 after surgery and subjected to histomorphometric analysis and biochemical tests. Results were analyzed by one-way ANOVA (p ≤ 0.05). Peri-implant tissue samples exhibited characteristic...

Lipid peroxidation in female dogs bearing mammary gland carcinomas Arq. Bras. Med. Vet. Zootec.
Souza,I.B.; Cardoso,C.V.; Poppe,S.C.; Bondan,E.F..
