In the present work, cultured atrial cells isolated from oyster (Crassostrea gigas) were used as an experimental model to investigate the toxicity of tributyltin (TBT) after short-time exposure at concentrations representative of those that can be measured in seawater, marine sediments and/or bivalves bioaccumulating this pollutant. Purified DNA was then digested by a mix of 3 restriction enzymes (AluI, RsaI, HaeIII). Impact of intensive larval rearing practices in oyster hatcheries : selective processes and loss of genetic variability (Poster) ArchiMer

Effects of the herbicide Basamais (bentazon) and the fungicide Opus (epoxiconazole) on oyster spat (Crassostrea gigas) were assessed using in-situ microcosms in a field experiment lasting 13 days. Six-week-old hatchery spat (mean size 1.1 mm), previously collected on PVC plates, was immersed in glass bottles filled with 200 mu m filtered seawater. Vibrio aestuarianus zinc metalloprotease causes lethality in the Pacific oyster Crassostrea gigas and impairs the host cellular immune defenses ArchiMer

Extracellular products (ECPs) of the pathogenic Vibrio aestuarianus 01/32 were previously reported to display lethality in Crassostrea gigas oysters and to cause morphological changes and immunosuppression in oyster hemocytes. To identify the source of this toxicity, biochemical and genetic approaches were developed. ECP protease activity and lethality were shown to be significantly reduced following incubation with metal chelators, suggesting the involvement of a zinc metalloprotease. If mechanistic models have been proposed, molecular studies aiming to substantiate these models have remained scarce. We reveal here the transcriptional signature associated with immune priming in the oyster Crassostrea gigas. Hybridization and morphological differences between the two closely related oyster taxa Crassostrea angulata and C. gigas ArchiMer

Detection of ostreid herpesvirus-1 (OsHV-1) by PCR using a rapid and simple method of DNA extraction from oyster larvae ArchiMer

A DNA extraction procedure was developed for the detection of ostreid herpesvirus-1 (OsHV-1) using the polymerase chain reaction (PCR) in oyster larvae. The DNA extraction procedure developed was tested on 8 larval samples. Abnormal nuclei with characteristic features associated with OsHV-1 infections were only observed in samples in which the viral DNA was detected by PCR. A previously described competitive PCR method was applied to detect inhibition during PCR reactions. The results show that the method can be used on small amounts of oyster larvae (3 mg) for the detection of OsHV-1 DNA by PCR.
