International study on <i>Artemia</i> : LX. Allozyme data suggest that a new <i>Artemia</i> population in southern Chile (50° 29' S; 73° 45' W) is <i>A. persimilis</i> 27
Gajardo, G.; Mercado, C.; Beardmore, J.A.; Sorgeloos, P..
An <i>Artemia</i> population from a site recently described in Chile and located in a semi-arid and humid hydrographical zone (Torres del Paine National Park), was electrophoretically characterized and cross-bred (followed through <i>F</i>3) to reference samples of <i>A. franciscana</i> (San Francisco Bay, California, USA) and <i>A. persimilis</i> (Buenos Aires, Argentina), which are the species most likely to be found in Chile. The population exhibited high levels of genetic variability as judged by the percentage of polymorphic loci (<i>P</i>=53.3), mean number of alleles per locus (<i>n</i>=2.2) and mean heterozygosity (<i>H</i>=0.176). Based on...

Genetic diversity and relationship in American and African oil palm as revealed by RFLP and AFLP molecular markers 86
Barcelos,Edson; Amblard,Philippe; Berthaud,Julien; Seguin,Marc.
The objective of this work was to evaluate the genetic diversity, its organization and the genetic relationships within oil palm (Elaeis oleifera (Kunth) Cortés, from America, and E. guineensis (Jacq.), from Africa) germplasm using Restriction Fragment Length Polymorphism (RFLP) and Amplified Fragment Length Polymorphism (AFLP). In complement to a previous RFLP study on 241 E. oleifera accessions, 38 E. guineensis accessions were analyzed using the same 37 cDNA probes. These accessions covered a large part of the geographical distribution areas of these species in America and Africa. In addition, AFLP analysis was performed on a sub-set of 40 accessions of E. oleifera and 22 of E. guineensis using three pairs of enzyme/primer combinations. Data were...

Exon-primed intron-crossing (EPIC) markers as a tool for ant phylogeography 92
Ströher,Patrícia R.; Li,Chenhong; Pie,Marcio R..
Exon-primed intron-crossing (EPIC) markers as a tool for ant phylogeography. Due to their local abundance, diversity of adaptations and worldwide distribution, ants are a classic example of adaptive radiation. Despite this evolutionary and ecological importance, phylogeographical studies on ants have relied largely on mitochondrial markers. In this study we design and test exon-primed intron-crossing (EPIC) markers, which can be widely used to uncover ant intraspecific variation. Candidate markers were obtained through screening the available ant genomes for unlinked conserved exonic regions interspersed with introns. A subset of 15 markers was tested in vitro and showed successful amplification in several phylogenetically distant ant species. These...

Genetic structure of populations of Pissodes castaneus (De Geer) (Coleoptera, Curculionidae) using amplified fragment length polymorphism 92
Zaleski,Scheila R. M.; Lazzari,Sonia M. N.; Lazzarotto,Crisleide M.; Panzavolta,Tiziana; Iede,Edson T.; Marques,Francisco de A..
Genetic structure of populations of Pissodes castaneus (De Geer) (Coleoptera, Curculionidae) using amplified fragment length polymorphism. The objective of this study was to determine the genetic structure of populations of Pissodes castaneus from different areas and on different species of Pinus using the PCR-AFLP technique. Twenty samples were analyzed, representing 19 populations from Brazil and one from Florence, Italy, which is the region of origin of P. castaneus. The four combinations of primers generated a total of 367 fragments of DNA, and 100% of polymorphic loci, indicating high degree of molecular polymorphism. The dendrogram did not reveal trends for grouping the populations in relation to origin. The low genetic similarity (0.11 between the...

Danko Brncic and the flower-flies 97
CARSON,HAMPTON L..
This is a personal essay commemorating Brncic as a keen naturalist/evolutionist in the best Darwinian sense. His innate curiosity, so important in a scientist, led him to the discovery of cenain unique drosophilid flies in Chile. The life cycle of one species of flower-fly (Drosophila flavopilosa) is of special interest. Females oviposit in fresh flowers of a solanaceous plant that is widespread in Chilean valleys. Twenty years of combined ecological and genetic work, carried out on this species, pioneers the use of a highly specialized Drosophila species in the study of population genetics and evolution. Many eco-genetic features are revealed that are important supplements to observations made on the widely-studied generalist species of the genus

Is the evolutionary theory still useful?: A review with examples 97
NESPOLO,ROBERTO F.
Evolutionary biology is experiencing an exceptional process of revisión and outreach because of the 200-anniversary if the birth of Charles Darwin. As a consequence, the study of organic evolution and also its teaching are being discussed at several levels, by evolutionary biologists, biologists and scholars outside evolutionary biology and by the general public. In this scenario, a didactic explanation of how biologists address evolutionary research in real populations seems to be useful. Using actual research examples, here I tried to outline how the classic theory (termed here as the "basic scheme") is useful to answer relevant questions in biology and how a less dogmatic paradigm (or a more versatile one) would be needed when dealing with the most...

Genetic variability in Pseudoplatystoma reticulatum from a breeding program in Brazil 124
Albuquerque,Daniele Menezes; Oliveira,Carlos Antonio Lopes de; Rodriguez,Maria Del Pilar Rodriguez; Ribeiro,Ricardo Pereira.
ABSTRACT The aim of this study was to characterise genetic variability in five groups of Pseudoplatystoma reticulatum: Mato Grosso I (MT-I), Mato Grosso II (MT-II), Mato Grosso III (MT-III), Mato Grosso do Sul I (MS-I) and Mato Grosso do Sul II (MS-II), taken from a centre for genetic improvement. In order to determine the allelic frequency, observed heterozygosity (HO), inbreeding coefficients (FIS), Hardy-Weinberg equilibrium (HW-p) and genetic differentiation, eight polymorphic microsatellite loci (Pcor01, Pcor05, Pcor08, Pcor10, Ppu01, Ppu04, Ppu09 and Ppu10) were selected, in which a total of 62 alleles were observed in the individuals (n = 211). The MS-I population had the highest (Pcor08 allele 83: 0.5000) and lowest (Pcor10 allele 50: 0.1667) mean...

Structure of genetic diversity of Bemisia tabaci (Genn.) (Hemiptera: Aleyrodidae) populations in Brazilian crops and locations 63
Fontes,Fernanda von Hertwig Mascarenhas; Colombo,Carlos Augusto; Lourenção,André Luiz.
Bemisia tabaci (Genn.) represents a complex of cryptic species that causes losses of many valuable crops. Even though there are differences in their ability to colonize hosts, transmit phytovirus and develop resistance to insecticides, the genetic differentiation of the insect populations is important for the adoption of control measures. Therefore, the genetic diversity of B. tabaci populations in economically important crops in Brazilian locations was characterized through a microsatellite analysis. Eight microsatellite markers were used for the analysis of eight populations, three in Solanum tuberosum (States of São Paulo, Minas Gerais and Bahia), two in Glycine max (States of São Paulo and Mato Grosso), one in Phaseolus vulgaris and Brassica oleracea...

Intrapopulation fixation index dynamics in finite populations with variable outcrossing rates 63
Coelho,Alexandre Siqueira Guedes; Vencovsky,Roland.
The intrapopulation fixation index ( f ) is inversely related to the outcrossing rate (t). Results obtained from data on molecular markers of natural populations have shown that these values are highly variable, even when measured in the same group of individuals. It is thus suggested that factors besides those described in Wright's genetic equilibrium must be operating. Using simulated data sets this study shows that the finite size condition of a population is sufficient to spread the estimated f values along a range at equilibrium, as opposed to keeping them at the theoretical equilibrium point. The variation in outcrossing rates can amplify this range considerably. Correlation between estimated f values obtained from different loci in this condition...

Genetic diversity in Calibrachoa pygmaea (Solanaceae): A hawkmoth-pollinated nightshade from the Pampas 39
Mäder,Geraldo; Backes,Alice; Reck-Kortmann,Maikel; Freitas,Loreta B..
ABSTRACT Calibrachoa pygmaea is a unique species of Calibrachoa, especially concerning its flower morphology and the environment where it occurs. The species is self-incompatible and is narrowly distributed in wet and flooded fields of the Pampas region. We characterize the genetic diversity of the species based on traditional plastid markers and newly developed nuclear microsatellites to identify drivers that guide its evolution. Our results identified markers that are informative and useful for studying the population structure of C. pygmaea, as well as that of other species of Calibrachoa. Both marker sets were congruent in developing conclusions regarding the evolutionary scenario of C. pygmaea, and revealed that the genetic variability and population...

Development and characterization of microsatellite markers for Tibouchina hatschbachii (Melastomataceae), an endemic and habitat-restricted shrub from Brazil 3
Maia, Fabiano Rodrigo; Sujii, Patricia Sanae; Goldenberg, Renato; Pereira, Viviane Silva; Zucchi, Maria Imaculada.
Tibouchina hatschbachii Wurdack (Melastomataceae) is an autogamous shrub restricted to granite (GO) and sandstone (SO) rock outcrops from subtropical Brazil. We designed primers for the amplification of microsatellite regions for T. hatschbachii, and characterized these primers to estimate genetic diversity parameters and contemporary genetic structure patterns. Eight loci were successfully amplified and were characterized using 70 individuals from three natural populations. Polymorphic information content ranged from 0.200 to 0.772 per locus. All loci were polymorphic, with allele numbers ranging from two to eight. The low degree of polymorphism may be explained by the fact that T. hatschbachii has disjunct populations and a recent genetic bottleneck, and...

SARZI,DEISE S.; JUSTOLIN,BEATRIZ; SILVA,CLARIANE DA; LEMOS,RAFAEL P.M.; STEFENON,VALDIR MARCOS.
Abstract: Eugenia uniflora L. (Myrtaceae) is a tree species widely distributed in South America suffering the effects of the exploitation of natural populations. In this study, we employed low coverage sequencing of the E. uniflora genome for mining of SSR markers. The de novo assembly generated 2,601 contigs with an average length of 1139 bp and spans 3.15 Mb. A total of 76 dimer, 33 trimer and two compound SSR loci were identified. Twelve selected SSR loci were employed to genotype 30 individuals from two natural populations. A total of 73 alleles were detected (mean A= 6.1) were observed, the mean effective number of alleles was Ae = 3.91, mean Ho was 0.23 and mean HE was 0.70). The mean Wright’s within population fixation index was FIS = 0.66 and...

Amplification of microsatellites in two haemulid species (Haemulon aurolineatum and Haemulon steindachneri) 50
Pabón Quintero,Paula E.; Tavera,José Julián; Millán-Márquez,Ana María; Acero P,Arturo.
ABSTRACT Genetic-population studies in marine fish have allowed to study patterns of dispersal and connectivity between habitats. One important tool in population genetics is the use of microsatellite molecular markers. Cross-amplification of microsatellite is a method that consists in using primers designed for one species in a different one but phylogenetically related. Because of the importance of genetic studies of populations in artisanal fisheries species, primers were evaluated and designed for the species Haemulon aurolineatum and Haemulon steindachneri. Samples were collected from the artisanal fisheries in Baru-Colombia. Amplification was standardized for 12 microsatellites which ten were polymorphic for H. aurolineatum and nine for H....

Genetics of two marine shrimp hatcheries of the Pacific white shrimp Litopenaeus vannamei (Boone, 1931) in Pernambuco, Brazil 65
Lima,Ana Patrícia Souza de; Silva,Suzianny Maria Bezerra Cabral da; Oliveira,Karine Kelly Cavalcanti; Maggioni,Rodrigo; Coimbra,Maria Raquel Moura.
The shrimp industry has grown significantly over the past 10 years in Brazil, especially the farmed production of the exotic Pacific white shrimp, Litopenaeus vannamei. In 2004, this industry was marked by a productivity crisis, which stirred interest towards genetic improvement of shrimp stocks. Shrimp breeders importation was banned in Brazil by a govern Normative Instruction in 1997, as a sanitary precaution. Since then, broodstock replacement in hatcheries has been based on domestic stocks, raising concerns on the decline of genetic diversity and if the existing diversity would allow effective genetic improvement programs. In the present research, genetic parameters such as number of alleles, effective allele number, expected and observed...

Storage protein variability in natural populations of maté (Ilex paraguariensis) in Brazil 65
Gregianini,Tatiana Schäffer; Winge,Helga.
ABSTRACT: Samples of 168 maté trees (Ilex paraguariensis) from four natural populations of different states of Brazil (Rio Grande do Sul, Santa Catarina, Paraná and Mato Grosso do Sul) were analyzed for their variability in storage proteins seed, which were used to estimate the degree of differentiation among and within species, because are relatively stable during evolution. Data on band presence/absence from 80 different polypeptides were used to perform similarity coefficient of Jaccard. A high level of genetic variability was reported within sampled populations (SJ intra=0.374). Lower levels of diversity were observed between populations (SJ inter=0.308). Total genetic diversity of maté was Hsp=0.264 estimated by Shannon measure. Partition of that...
