Membrane adsorber for endotoxin removal BJPS
Almeida,Karina Moita de; Almeida,Maiara Maciel; Fingola,Fernando Faria; Ferraz,Helen Conceição.
ABSTRACT The surface of flat-sheet nylon membranes was modified using bisoxirane as the spacer and polyvinyl alcohol as the coating polymer. The amino acid histidine was explored as a ligand for endotoxins, aiming at its application for endotoxin removal from aqueous solutions. Characterization of the membrane adsorber, analysis of the depyrogenation procedures and the evaluation of endotoxin removal efficiency in static mode are discussed. Ligand density of the membranes was around 7 mg/g dry membrane, allowing removal of up to 65% of the endotoxins. The performance of the membrane adsorber prepared using nylon coated with polyvinyl alcohol and containing histidine as the ligand proved superior to other membrane adsorbers reported in the literature. The...

Animal experimentation and scientific knowledge: a thought style? BJPS
Tréz,Thales de Astrogildo e.
Animal experimentation, besides a research method extensively applied in the production of scientific knowledge, is also considered essential to science and with undeniable historical relevance in advances in human health. In this survey, a questionnaire was applied to a group of researchers involved with research based on non-animal models (n =18), and to another group involved with research based on animal models (n =18). The data analysis was grounded in Ludwik Fleck (1896 -1961) epistemological assumptions. The results suggested that there are at least two thought styles operating in consonance on the same research problem (advances in human health conditions) with significantly different conceptions not only concerning the research practices involved,...

Estimating the turnover number in enzyme kinetic reactions using transient and stationary state data BJPS
Uludag-Demirer,Sibel; Duran,Jorge; Tanner,Robert D..
Substrate and product concentration data obtained by simulating enzyme-substrate reaction rate equations were used to test two proposed kinetic rate constant estimation techniques in this study. In the first technique, the turnover number, k3, was calculated using early transient time domain data, which are difficult to obtain experimentally. The technique used an iterative approach to calculate k3 with a pair of data and the value of k3 could be retrieved with 35% error. The second technique calculated k3 using stationary domain data and the value of k3 could be retrieved with less than 5% error. This second technique also offered internal consistency in the calculation of k3 by calculating k3 both from the intercept and the slope of the linear plot...

Arginine enzymatic deprivation and diet restriction for cancer treatment BJPS
Zam,Wissam.
ABSTRACT Recent findings in amino acid metabolism and the differences between normal, healthy cells and neoplastic cells have revealed that targeting single amino acid metabolic enzymes in cancer therapy is a promising strategy for the development of novel therapeutic agents. Arginine is derived from dietary protein intake, body protein breakdown, or endogenous de novo arginine production and several studies have revealed disturbances in its synthesis and metabolism which could enhance or inhibit tumor cell growth. Consequently, there has been an increased interest in the arginine-depleting enzymes and dietary deprivation of arginine and its precursors as a potential antineoplastic therapy. This review outlines the most recent advances in targeting...

Basic considerations in the dermatokinetics of topical formulations BJPS
Nair,Anroop; Jacob,Shery; Al-Dhubiab,Bandar; Attimarad,Mahesh; Harsha,Sree.
Assessing the bioavailability of drug molecules at the site of action provides better insight into the efficiency of a dosage form. However, determining drug concentration in the skin layers following topical application of dermatological formulations is a great challenge. The protocols followed in oral formulations could not be applied for topical dosage forms. The regulatory agencies are considering several possible approaches such as tape stripping, microdialysis etc. On the other hand, the skin bioavailability assessment of xenobiotics is equally important for topical formulations in order to evaluate the toxicity. It is always possible that drug molecules applied on the skin surface may transport thorough the skin and reaches systemic circulation....
