Suitability of the tetrazolium test methodology for recently harvested and stored coffee seeds Ciência e Agrotecnologia
Clemente,Aline da Consolação Sampaio; Carvalho,Maria Laene Moreira de; Guimarães,Renato Mendes.
Germination of coffee seeds occurs in a slow and non-uniform manner and the tetrazolium test (TZ) may be a quick alternative for assessment of viability, as long as the preparation and imbibition of the seeds is established. This study was performed in two stages: in the first, recently harvested seeds from three coffee cultivars were submitted to four methodologies for removal of the parchment: manual extraction or use of sodium hypochlorite (NaClO at 5%) and two periods of imbibition of the seeds in water (36 and 48 hours) for removal of the embryos. In the second phase, manual removal and the use of NaClO were compared for elimination of the parchment of stored seeds for monthly performance of the tetrazolium test for six months. Sodium hypochlorite is...

Absorption and metabolism of volatile fatty acids by rumen and omasum Ciência e Agrotecnologia
Daniel,João Luiz Pratti; Resende Júnior,João Chrysostomo de.
Volatile fatty acids (VFA) absorption and metabolic capacity of rumen and omasum were compared, in vitro. Fragments of rumen wall and omasum laminae were taken from eight adult crossbred bovines. An isolated fragment of the mucosa was fitted in a tissue diffusion chamber. Valeric acid and CrEDTA were added to ruminal fluid and placed on the mucosal side and buffer solution was placed on the serosal side. Fractional absorption rates were measured by exponential VFA:Cr ratio decay over time. Metabolism rate was determined as the difference between VFA absorbed and VFA which appeared on the serosal side over time. Mitotic index was higher in omasum (0.52%) than in rumen epithelium (0.28%). VFA fractional absorption rate was higher in omasum (4.6%/h.cm²) than...

Carvalho,Tereza Cristina de; Krzyzanowski,Francisco Carlos; Ohlson,Osvaldo de Castro; Panobianco,Maristela.
The tetrazolium test is widely used for seed vigor assessment of various plant species. For wheat, however, there is no methodology specifically recommended. This study aimed at determining an efficient procedure to evaluate vigor of wheat seeds by this test; besides allowing the establishment of seed quality classes. Thus, four different wheat seed lots were assessed by determining the moisture content and through tests of germination, accelerated aging, and field seedling emergence. The following methodologies were assessed: 1) longitudinal bisection of seed, with subsequent placement of the two halves on filter paper moistened with a 1.0% tetrazolium solution, at 30º C, for 2 h; 2) longitudinal bisection, with subsequent immersion of one seed half into...

Morphology, mineralogy and micromorphology of soils associated to summit depressions of the Northeastern Brazilian Coastal Plains Ciência e Agrotecnologia
Silva,Elen Alvarenga; Gomes,João Bosco Vasconcellos; Araújo Filho,José Coelho de; Vidal-Torrado,Pablo; Cooper,Miguel; Curi,Nilton.
The scarcity of comprehensive characterizations of soils associated to gentle summit depressions of the Northeastern Brazilian Coastal Plains justifies this work, which had as objective to provide basic information for the more diverse agricultural and non-agricultural uses. For that, representative soils (Spodosols or similar soils) from these environments were selected in Alagoas, Sergipe and Bahia states. This approach included characterization of morphological, mineralogical and micromorphological properties of the soil profiles, employing standard procedures. The morphological characterization corroborated the effect of the podzolization process during the formation of these soils. The mineralogy of the clay fraction of these soils was basically...

Development of sampling for quantification of glyphosate in natural waters Ciência e Agrotecnologia
Tzaskos,Danilla Fernanda; Marcovicz,Crislaine; Dias,Nivea Maria Piccolomini; Rosso,Neiva Deliberali.
Glyphosate is a systemic, post-emergent, non-selective herbicide widely used in agriculture. The objective of this study was to develop a method for sample preparation, to partially purified natural water samples contaminated with residues of glyphosate, and to quantify them by derivation and spectroscopically. To assess the accuracy of the method, samples of water from an artesian well and from a stream were fortified with known amounts of glyphosate. The concentration of glyphosate was determined using a standard curve concentration range from 4 to 14 mg L-1. From the concentration of glyphosate recovered, in both samples of fortified water it was found that DOWEX MWA-1 resin and NaCl 0.100 mol L-1 eluent were effective in the purification of the...

Effect of maltodextrin on the freezing point and thermal conductivity of uvaia pulp (Eugenia piriformis Cambess) Ciência e Agrotecnologia
Villa-Vélez,Harvey Alexander; Telis-Romero,Javier; Higuita,Diana Maria Cano; Telis,Vânia Regina Nicolletti.
The freezing point depression (FPD) of uvaia pulp with and without additives - 10, 16, 22 and 28% of maltodextrin (MD), was measured using a simple apparatus consisting of two major sections: a freezing vessel and a data acquisition system. The thermal conductivity of the pulps was also investigated as a function of the frozen water fraction and temperature using a coaxial dual-cylinder apparatus. Above the initial freezing point, thermal conductivity fitted the polynomial equations well. Below the freezing point, thermal conductivity was strongly affected by both the frozen water fraction and the temperature. Simple equations in terms of the frozen water fraction and temperature could be fitted to the experimental data for freezing point depression and...

Development and description of light functional dulce de leche with coffee Ciência e Agrotecnologia
Guimarães,Ívina Catarina de Oliveira; Leão,Maria Helena Miguez da Rocha; Pimenta,Carlos José; Ferreira,Larissa de Oliveira; Ferreira,Eric Batista.
This study deals with development of a "dulce de leche" (DL) with coffee, with low caloric content and functional properties. Through rotational central composite design, eleven DLs were formulated with different polydextrose and sucralose levels. The results were analyzed by means of response surface and ANOVA. The addition of polydextrose led to the increase in moisture and water activity and reduction in the ash and soluble solids contents. In the formulations with greater levels of sucralose and polydextrose, there was an increase in the ash, acid and fiber content. Greater yield was achieved with greater concentrations of polydextrose and less substitution of sucrose by sucralose, allowing development of a new functional product with low caloric...

Levels of digestible isoleucine on performance, carcass traits and organs weight of gilts (15 - 30 KG) Ciência e Agrotecnologia
Castilha,Leandro Dalcin; Pozza,Paulo Cesar; Nunes,Ricardo Vianna; Lazzeri,Doglas Batista; Somensi,Marcelo Luiz; Pozza,Magali Soares dos Santos.
The ideal protein concept for pigs has allowed reducing levels of crude protein in the diet, since synthetic amino acids are included because the branched chain amino acids may be limiting. In order to determine the digestible isoleucine requirement for gilts from 15 to 30 kg, a performance assay was accomplished, using 40 crossbreed gilts of high genetic potential, averaging 15.00 ± 0.52 kg of body weight, alloted in a randomized blocks design, consisting of five treatments (0.45, 0.52, 0.59, 0.66, 0.73% of digestible isoleucine), four replicates and two animals per experimental unit. Performance traits were determined and at the end of the experiment one animal per experimental unit was slaughtered to determine carcass composition and organs weight....

Ematné,Hugo Junqueira; Souza,João Cândido de; Biudes,Gustavo Barnabé; Nunes,José Airton Rodrigues; Guedes,Fernando Lisboa.
Popcorn is widely consumed in the south of Minas Gerais; however, up to now no specific breeding program for the region has been carried out. Recurrent selection (RS) is a viable alternative for breeding of this crop; nevertheless, it is necessary to frequently evaluate the progress achieved. Consequently, the purpose of this work was to assess the genetic progress obtained after two cycles of RS for the traits of popping expansion (PE) and grain yield (GY). Cycles one and two of the UFLA - E and UFLA - R populations, the original population (UFLA) and two controls were assessed. The experiment was conducted at the UFLA experimental farm in a randomized block design with eleven replications; ten agronomic traits, including PE and GY, were assessed. Gains...

Response to water deficit of Ipomoea cairica (L.) Sweet Ciência e Agrotecnologia
Tognon,Grasiela Bruzamarello; Petry,Cláudia; Cuquel,Francine Lorena.
Green spaces require frequent watering, increasing the pressure on water resources. The use of native plants in landscaping reduces the risks of biological contamination, contributing to preservation of Brazilian biodiversity and provides the value of sustainability in urban green areas. Of these native plants, a restricted number are native lianas which are adapted to Brazilian climate and available on the market. In this regard, the selection and recommending of native plants which are efficient in use of water for planting in urban areas constitute an effective strategy for conservation of water resources. Thus, the purpose of this study was to evaluate the growth and development of I. cairica plants submitted to water deficit. For a period of 100 days...

Evaluation of antifungal activity of seaweed extracts Ciência e Agrotecnologia
Peres,Julio Cesar Fernandes; Carvalho,Luciana Retz de; Gonçalez,Edlayne; Berian,Luis Otávio Saggion; Felicio,Joana D´arc.
Seaweeds are subject to numerous biological interactions and sometimes to extreme abiotic conditions, so they have developed among other defense mechanisms, the ability to produce biologically active substances. Thus, these organisms produce mainly terpenes and phenols. Among others, the antifungal activity, due to its importance in human and animal health and the production of agricultural products, has been the subject of several studies. In the present work, this activity was investigated in ten seaweeds extracts, by direct bioautography assays, compared to Colletotrichum lagenarium and disk diffusion assay, compared to Aspergillus flavus. The organisms studied were: Stypopodium zonale, Laurencia dendroidea, Ascophyllum nodosum, Sargassum muticum,...
