Background The high capacity of chloroplast genome response to integrate and express transgenes at high levels makes this technology a good option to produce proteins of interest. This report presents the stable expression of Pectin lyase (PelA gene) and the first stable expression of manganese peroxidase (MnP-2 gene) from the chloroplast genome. Results pES4 and pES5 vectors were derived from pPV111A plasmid and contain the PelA and MnP-2 synthetic genes, respectively. Sugarcane bagasse as feedstock for cellulase production by Trichoderma harzianum in optimized culture medium Electron. J. Biotechnol.

This work aimed at the production of cellulases from pretreated sugarcane bagasse by the filamentous fungus Trichoderma harzianum IOC 3844 and their application in the hydrolysis of this same substrate, for a future use in second-generation ethanol production. The production of cellulases was optimized, which resulted in high enzymatic activities after 42 hrs of process in an instrumented bioreactor (CMCase 27,017 U x L-1; FPase 1,225 U x L-1; and β-glucosidase 609 U x L-1). Callus induction and plant regeneration of Ulex europaeus Electron. J. Biotechnol.

A callus induction and plant regeneration protocol was developed from leaf and thorn explants for the plant Ulex europaeus. Explants were incubated on 2% sucrose half-strength Murashige and Skoog Medium (MS) with various combinations of plant growth regulators and antioxidants. The best frequency of callus and shoot formation was obtained with 2,4-dichlorophenoxyacetic acid (2,4-D) 1 mg/l x kinetin (Kin) 0.2 mg/l (DK Medium; callus induction) and zeatin (Z) 1 mg/l (DK medium; shoot induction). Effect of glutaraldehyde biocide on laboratory-scale rotating biological contactors and biocide efficacy Electron. J. Biotechnol.

The effect of glutaraldehyde, a commercial biocide widely used in paper and pulp industry, on the performance of laboratory-scale rotating biological contactors (RBCs) as well as biocide efficacy was studied. Biofilms were established on the RBCs and then exposed to 0 - 180 ppm glutaraldehyde at a dilution rate of 1.60 h-1. The results showed that the biofilms became acclimated to glutaraldehyde and eventually could degrade it. Acclimation to the biocide took longer at the higher biocide concentrations. The degree of biocide degradation and chemical oxygen demand (COD) removal depended on acclimation period, the presence of other organic matters and the amount of mineral salts available. Simple sequence repeats (SSR) or micro-satellites are becoming standard DNA markers for plant genome analysis and are being used as markers in marker assisted breeding. De novo generation of micro-satellite markers through laboratory-based screening of SSR-enriched genomic libraries is highly time consuming and expensive. An alternative is to screen the public databases of related model species where abundant sequence data is already available. All the genomic sequences of Medicago from the public domain database were searched and analysed of di, tri, and tetra nucleotide repeats. Repetitive element palindromic PCR (rep-PCR) as a genetic tool to study interspecific diversity in Euphorbiaceae family Electron. J. Biotechnol.

Background The classification of diversity in germplasm collections is important for plant breeding. The repetitive element palindromic-polymerase chain reaction (rep-PCR) technique was used to investigate inter-specific diversity within 17 species from the Euphorbiaceae family using REP and BOX primers. Results The agglomerative cluster analysis was used to evaluate the scoring data. BOX and REP gave amplification with polymorphism of 98.84% and 100% respectively. Background: A study of the correlation between the particle size of lignocellulosic substrates and ultrasound pretreatment on the efficiency of further enzymatic hydrolysis and fermentation to ethanol. Results: The maximum concentrations of glucose and, to a lesser extent, di- and trisaccharides were obtained in a series of experiments with 48-h enzymatic hydrolysis of pine raw materials ground at 380-100 rpm for 30 min. Molecular modeling of manganese peroxidase from the lignin-degrading fungus Ceriporiopsis subvermispora and structural comparison with other peroxidases Electron. J. Biotechnol.

Ceriporiopsis subvermispora is a white-rot basidiomycete that produces several isoenzymes of manganese peroxidase (MnP· ). A cDNA of one of them (MnP13-1) has been isolated and sequenced. The deduced aminoacid sequence shows about 60% similarity with the MnPs from Phanerochaete chrysosporium. Based on the crystal structures of MnP and lignin peroxidase (LiP) from P. chrysosporium, and of a peroxidase from Arthromyces ramosus (ARP), we have modeled by homology the three dimensional structure of MnP13-1 using standard modeling procedures.
