The chief importance of hemocytes as immune effectors in molluscs has led to develop researches for in vitro culture. The chemiluminescence technique has been used to estimate the phagocytosis capacity of Crassostrea gigas hemocytes according to different culture media and times. Sea water was used as a basic medium. Additives, such as antibiotics or glucose and buffers, such as Tris or Hepes, were tested as weIl as artificial sea water. Cryopreservation is widely used for long-term conservation of various tissues, embryos or gametes. However, few studies have described cryopreservation of invertebrate primary cell cultures and more particularly of marine invertebrate somatic cells. This technique would however be of great interest to facilitate the study of various metabolic processes which vary seasonally. The aim of the present study was to develop a protocol for cryopreservation of Crassostrea gigas vesicular cells. Different parameters were adjusted to improve recovery of cells after freezing. Previous studies have shown that magnetic resonance imaging is a very appropriate non-invasive technique for quantifying the growth of somatic and gonadic tissues and sex determination in the Pacific oyster, Crassostrea gigas. Despite a thinner gonad, harder to distinguish than that of the Pacific oyster, we showed in this study that it was possible to distinguish and quantify the development of the gonad of the European flat oyster, Ostrea edulis, using a 3D MRI sequence with a suitable finer resolution. The study of maturation and spawning of the oyster is part of a research program to investigate the summer mortalities of the oysters, Crassostrea gigas in Marennes-Oléron Bay. Four maturity stages were simultaneously obtained by diet and thermal conditioning (immature, low maturation, mature and post-spawning stages). Measurements of clearance, filtration, absorption and respiration rates allowed a calculation of the scope for growth and hence an estimation of the oyster's energetic budget at various maturity stages. The protozoan Bonamia ostreae (Ascetospora) is the etiological agent of bonamiosis, or hemocyte disease of the European fiat oyster, Ostrea edulis (Pichot et al., 1980). Since this infection was first reported from the coast of Brittany (Comps et al., 1980), the parasite has spread rapidly along the French Atlantic coasts (Grizel & Tigé, 1982), and in the European countries including the UK, Holland, lreland and Spain (Bannister & Key, 1982; Van Banning, 1982; Polanco et al., 1984). The aim of this study was to evaluate the importance of biotransformation of paralytic shellfish toxins during the detoxification process in contaminated oysters. Mathematical models based upon the detoxification patterns of digestive gland and other tissues were developed. It was demonstrated that biotransformations do not seem to play an important role in digestive gland or other tissue detoxification kinetics with our data set.
